The aim of the present work was to compare the efficacies and levels of selectivity of different culture-dependent and -independent methods for analyzing bifidobacteria in human stool samples. The three different culture media used here significantly differed from each other, particularly with regard to the recovery of Bifidobacterium adolescentis. Bifidobacterium medium failed to recover B. adolescentis; Beerens medium recovered some B. adolescentis organisms (17% of total bifidobacteria), whereas tomato-Eugon medium recovered mainly B. adolescentis organisms (58% of total bifidobacteria). A culture-independent method that combines GC fractionation of bacterial community DNA and 16S rRNA sequencing indicated that B. adolescentis organisms accounted for 85% of all bifidobacteria. Methodological biases, such as those described in this paper, should be taken into account in interpreting earlier studies and designing future experiments.  相似文献

Abstract. Major changes in forest floor vegetation were identified on the basis of three nationwide surveys conducted as part of national forest inventories in 1951–1953, 1985–1986 and 1995. These surveys provided objectively selected, statistically representative samples of all forested land in Finland. The 1951–1953 data consist of over 10000 sample plots, while the later surveys were conducted on ca. 3000 permanent plots. Changes in relative abundance of dominant species (i.e. in the proportions of species of the total cover of forest floor vegetation) were analysed across biogeographical provinces. Spatial correlation, systematic sampling, partial re‐measurement and multiple testing were taken into account in assessment of the statistical significance of the observed changes. The most notable changes in forest floor vegetation were a decrease in the relative abundance of Hylocomium splendens and an increase in Dicranum polysetum. In N Finland, where forests are grazed by semi‐domestic reindeer, we observed a decline in the abundance of Cladina lichens and an increase in Dicranum mosses. Peltigera aphthosa declined throughout the country. Polytrichum juniperinum, Pohlia nutans, and Brachythecium species, which occupy disturbed sites or grow on litter, increased in abundance. The relative abundance of Sphagnum species decreased, particularly in W Finland, where Pleurozium schreberi was favoured. A major decline in S. fuscum was also recorded in C and E Finland. Many of the changes detected in this study are apparently related to intensified forest management; but solely on the basis of this study, its effects cannot be distinguished from those of other large‐scale environmental changes.  相似文献

The effect of whole-body cryotherapy (WBC) on rectal and skin temperatures was measured in healthy subjects before, during and after WBC exposure. WBC did not cause any significant change in rectal temperature. The lowest local skin temperatures were recorded in the forearm, 5.2 (2.8)°C, and in the calf, 5.3 (3.0)°C. WBC involves no risk for frostbites. After WBC, all skin temperatures recovered rapidly, indicating that the analgetic effects of WBC only occur during a limited period after the exposure.  相似文献

Osteopetrosis is a metabolic bone disease with increased skeletal density radiographically and increased risk of fracture. Experimental studies with rat osteopetrotic mutations have shown increased bone density and decreased bone strength. However, it is not known if this reduction in bone strength is only due to changes in structure and geometry or if the tissue properties of bone material itself are changed as well. We have evaluated bone tissue properties with nanoindentation in three osteopetrotic mutations in the rat (incisors-absent ia/ia, osteopetrosis op/op and toothless tl/tl) to test the hypothesis that reduced bone resorption in these mutations results in reduced tissue properties of bone material. No significant differences in elastic modulus or hardness were found between osteopetrotic mutants and their normal littermates (NLMs) in any of the three stocks. This indicates that the tissue properties of bone material are not changed significantly in osteopetrosis, even if the mechanical strength is decreased at the macroscopic level.  相似文献

This study compared the effect of repetitive work in thermoneutral and cold conditions on forearm muscle electromyogram (EMG) and fatigue. We hypothesize that cold and repetitive work together cause higher EMG activity and fatigue than repetitive work only, thus creating a higher risk for overuse injuries. Eight men performed six 20-min work bouts at 25 degrees C (W-25) and at 5 degrees C while exposed to systemic (C-5) and local cooling (LC-5). The work was wrist flexion-extension exercise at 10% maximal voluntary contraction. The EMG activity of the forearm flexors and extensors was higher during C-5 (31 and 30%, respectively) and LC-5 (25 and 28%, respectively) than during W-25 (P < 0.05). On the basis of fatigue index (calculated from changes in maximal flexor force and flexor EMG activity), the fatigue in the forearm flexors at the end of W-25 was 15%. The corresponding values at the end of C-5 and LC-5 were 37% (P < 0.05 in relation to W-25) and 20%, respectively. Thus repetitive work in the cold causes higher EMG activity and fatigue than repetitive work in thermoneutral conditions.  相似文献

Telomeres are highly conserved nucleoprotein structures which protect genome integrity. The length of telomeres is influenced by both genetic and environmental factors, but relatively little is known about how different hereditary and environmental factors interact in determining telomere length. We manipulated growth rates and timing of maturation by exposing full-sib nine-spined sticklebacks (Pungitius pungitius) to two different temperature treatments and quantified the effects of temperature treatments, sex, timing of maturation, growth rate and family (genetic influences) on telomere length. We did not find the overall effect of temperature treatment on the relative telomere length. However, we found that variation in telomere length was related to timing of maturation in a sex- and temperature-dependent manner. Telomere length was negatively related to age at maturation in elevated temperature and early maturing males and females differed in telomere length. Variation in growth rate did not explain any variation in telomere length. The broad sense heritability (h 2) of telomere length was estimated at h 2 = 0.31 ? 0.47, suggesting predominance of environmental over genetic determinants of telomere length variability. This study provides the first evidence that age at maturation together with factors associated with it are influencing telomere length in an ectotherm. Future studies are encouraged to identify the extent to which these results can be replicated in other ectotherms.  相似文献

Niche conservatism is the tendency of related species to retain ancestral tolerances after geographic separation. We used Ecological Niche Modelling and Principal Components Analysis of bioclimatic and habitat variables to describe the extent of the species niche, and degrees of bioclimatic–habitat niche conservatism within the mountain hare (L. timidus) clade. Mountain hare niche space was contrasted with that of the European hare (L. europaeus), to shed light on species interactions in contact zones throughout Europe. All five subspecies of mountain hare had quantifiably distinct niches. Fennoscandian (L.t. sylvaticus, L.t. timidus) and highland (L.t. scoticus, L.t. varronis) subspecies, however, were most similar, exhibiting greatest apparent niche conservatism. They inhabit tundra, boreal forest and uplands, and, hence are presumed most similar to the ancestral form. The Irish hare was distinct, being consistently distinguished from other mountain hares in both 2D and nth dimensional (4D) niche space. The ecological distinctiveness of the Irish hare provides further evidence that it is an Evolutionarily Significant Unit, particularly vulnerable to displacement by introduced European hares with which it competes and hybridises. Projections under global climate change suggest that, by 2070, bioclimatic space for invasive European hares in Ireland will expand (by 79%) but contract for endemic Irish hares (by 75%), further facilitating their replacement. The near complete species replacement of the heath hare (L.t. sylvaticus) in southern Sweden, where the European hare has also been introduced, may suggest a similar fate may be in store for the Irish hare.  相似文献

Most neurodegenerative diseases such as Alzheimer''s disease are proteinopathies linked to the toxicity of amyloid oligomers. Treatments to delay or cure these diseases are lacking. Using budding yeast, we report that the natural lipid tripentadecanoin induces expression of the nitric oxide oxidoreductase Yhb1 to prevent the formation of protein aggregates during aging and extends replicative lifespan. In mammals, tripentadecanoin induces expression of the Yhb1 orthologue, neuroglobin, to protect neurons against amyloid toxicity. Tripentadecanoin also rescues photoreceptors in a mouse model of retinal degeneration and retinal ganglion cells in a Rhesus monkey model of optic atrophy. Together, we propose that tripentadecanoin affects p‐bodies to induce neuroglobin expression and offers a potential treatment for proteinopathies and retinal neurodegeneration.  相似文献
