Evolution is known to act on contemporary timescales and invasive organisms are often used to study rapid evolutionary changes of geno- and phenotypes under natural conditions. The ability and speed of local adaptation is discussed as a key character triggering successful invasions. Variation of body shape among populations of two highly invasive, sympatric Ponto-Caspian goby species (Teleostei: Gobiidae) with a comparable invasion history in the upper Danube River, i.e. Neogobius melanostomus and Ponticola kessleri, was assessed using geometric morphometric methods. Phenotypic variation established within less than 15 generations was evident in both species. It was mainly correlated with geographical location, but in N. melanostomus also with substrate type, an ecological variable reflecting habitat quality. The two species differed in their degree of intraspecific variation which was more pronounced in N. melanostomus, the numerically dominant invader in the upper Danube. Body shape variation significantly correlating with geographical rather than ecological variables suggests a heritable component and renders phenotypic plasticity as a lone explanation unlikely. Patterns of body shape similarity among upper Danubian goby populations suggest a stepping-stone rather than a continuous expansion model for both species, where multiple introductions, possibly from various origins, may have shaped differentiation. 相似文献

In the present study, we have developed a novel one-arm single chain Fab heterodimeric bispecific IgG (OAscFab-IgG) antibody format targeting the insulin-like growth factor receptor type I (IGF-1R) and the epidermal growth factor receptor (EGFR) with one binding site for each target antigen. The bispecific antibody XGFR is based on the “knob-into-hole” technology for heavy chain heterodimerization with one heavy chain consisting of a single chain Fab to prevent wrong pairing of light chains. XGFR was produced with high expression yields and showed simultaneous binding to IGF-1R and EGFR with high affinity. Due to monovalent binding of XGFR to IGF-1R, IGF-1R internalization was strongly reduced compared with the bivalent parental antibody, leading to enhanced Fc-mediated cellular cytotoxicity. To further increase immune effector functions triggered by XGFR, the Fc portion of the bispecific antibody was glycoengineered, which resulted in strong antibody-dependent cell-mediated cytotoxicity activity. XGFR-mediated inhibition of IGF-1R and EGFR phosphorylation as well as A549 tumor cell proliferation was highly effective and was comparable with a combined treatment with EGFR (GA201) and IGF-1R (R1507) antibodies. XGFR also demonstrated potent anti-tumor efficacy in multiple mouse xenograft tumor models with a complete growth inhibition of AsPC1 human pancreatic tumors and improved survival of SCID beige mice carrying A549 human lung tumors compared with treatment with antibodies targeting either IGF-1R or EGFR. In summary, we have applied rational antibody engineering technology to develop a heterodimeric OAscFab-IgG bispecific antibody, which combines potent signaling inhibition with antibody-dependent cell-mediated cytotoxicity induction and results in superior molecular properties over two established tetravalent bispecific formats. 相似文献

Exosomes are small vesicles (50–150 nm) of endocytic origin that are released by many different cell types. Exosomes in the tumor microenvironment may play a key role in facilitating cell-cell communication. Exosomes are reported to predominantly contain RNA and proteins. In this study, we investigated whether exosomes from pancreatic cancer cells and serum from patients with pancreatic ductal adenocarcinoma contain genomic DNA. Our results provide evidence that exosomes contain >10-kb fragments of double-stranded genomic DNA. Mutations in KRAS and p53 can be detected using genomic DNA from exosomes derived from pancreatic cancer cell lines and serum from patients with pancreatic cancer. In addition, using whole genome sequencing, we demonstrate that serum exosomes from patients with pancreatic cancer contain genomic DNA spanning all chromosomes. These results indicate that serum-derived exosomes can be used to determine genomic DNA mutations for cancer prediction, treatment, and therapy resistance. 相似文献

Winter air temperature variability is projected to increase in the temperate zone whereas snow cover is projected to decrease, leading to more variable soil temperatures. In a field experiment winter warming pulses were applied and aboveground biomass and root length of four plant species were quantified over two subsequent growing seasons in monocultures and mixtures of two species. The experiment was replicated at two sites, a colder upland site with more snow and a warmer, dryer lowland site. Aboveground biomass of Holcus lanatus declined (?29 %) in the growing season after the warming pulse treatment. Its competitor in the grassland mixture, Plantago lanceolata, profited from this decline by increased biomass production (+18 %). These effects disappeared in the second year. There was a strong decline in biomass for P. lanceolata at the lowland site in the second year. These two species also showed a decline in leaf carbohydrate content during the manipulation. Aboveground productivity and carbohydrate content of the heathland species was not affected by the treatment. The aboveground effects of the treatment did not differ significantly between the two sites, thereby implying some generality for different temperate ecosystems with little and significant amount of snowfall. Root length increased directly after the treatment for H. lanatus and for Calluna vulgaris with a peak at the end of the first growing season. The observed species-specific effects emphasize the ecological importance of winter temperature variability in the temperate zone and appear important for potential shifts in community composition and ecosystem productivity. 相似文献

Directed dispersal by animal vectors has been found to have large effects on the structure and dynamics of plant populations adapted to frugivory. Yet, empirical data are lacking on the potential of directed dispersal by rotational grazing of domestic animals to mediate gene flow across the landscape. Here, we investigated the potential effect of large‐flock shepherding on landscape‐scale genetic structure in the calcareous grassland plant Dianthus carthusianorum, whose seeds lack morphological adaptations to dispersal to animals or wind. We found a significant pattern of genetic structure differentiating population within grazed patches of three nonoverlapping shepherding systems and populations of ungrazed patches. Among ungrazed patches, we found a strong and significant effect of isolation by distance (r = 0.56). In contrast, genetic distance between grazed patches within the same herding system was unrelated to geographical distance but significantly related to distance along shepherding routes (r = 0.44). This latter effect of connectivity along shepherding routes suggests that gene flow is spatially restricted occurring mostly between adjacent populations. While this study used nuclear markers that integrate gene flow by pollen and seed, the significant difference in the genetic structure between ungrazed patches and patches connected by large‐flock shepherding indicates the potential of directed seed dispersal by sheep across the landscape. 相似文献

In this study, we retrospectively analysed a total of 605 clinical isolates from six West or Central African countries (Benin, Cameroon, Central African Republic, Guinea-Conakry, Niger and Senegal). Besides spoligotyping to assign isolates to ancient and modern mycobacterial lineages, we conducted phenotypic drug-susceptibility-testing for each isolate for the four first-line drugs. We showed that phylogenetically modern Mycobacterium tuberculosis strains are more likely associated with drug resistance than ancient strains and predict that the currently ongoing replacement of the endemic ancient by a modern mycobacterial population in West/Central Africa might result in increased drug resistance in the sub-region. 相似文献
