Danshen, in particular its derivative tanshinone IIA (TS), is a promising compound in the treatment of cardiovascular diseases and has been used for many years in traditional Chinese medicine. Although many actions of TS have been researched, its vasodilator effects in pregnancy remain unknown. There have been a few studies that have shown the ability of TS to reduce blood pressure in women with hypertensive pregnancies; however, there are no studies which have examined the vascular effects of TS in the pregnant state in either normal or complicated pregnancies. Our aim was to determine the vasoactive role of TS in multiple arteries during pregnancy including: rat resistance (mesenteric and uterine) and conduit (carotid) arteries. Further, we aimed to assess the ability of TS to improve uterine blood flow in a rodent model of intrauterine growth restriction. Wire myography was used to assess vascular responses to the water-soluble derivative, sodium tanshinone IIA sulphonate (STS) or to the endothelium-dependent vasodilator, methylcholine. At mid-pregnancy, STS caused direct vasodilation of rat resistance (pEC50 mesenteric: 4.47±0.05 and uterine: 3.65±0.10) but not conduit (carotid) arteries. In late pregnancy, human myometrial arteries responded with a similar sensitivity to STS (pEC50 myometrial: 3.26±0.13). STS treatment for the last third of pregnancy in eNOS-/- mice increased uterine artery responses to methylcholine (Emax eNOS-/-: 55.2±9.2% vs. eNOS-/- treated: 75.7±8.9%, p<0.0001). The promising vascular effects, however, did not lead to improved uterine or umbilical blood flow in vivo, nor to improved fetal biometrics; body weight and crown-rump length. Further, STS treatment increased the uterine artery resistance index and decreased offspring body weight in control mice. Further research would be required to determine the safety and efficacy of use of STS in pregnancy. 相似文献

Drought is a natural phenomenon experienced by many intermittent and also seasonal lotic systems. It has diverse effects on the structure and distribution of biological communities through habitat transition from wetted to terrestrial conditions. The Njoro River, a tropical stream, was drought‐stressed between late 1999 and mid 2000, providing an opportunity to sample and describe the distributional patterns of diatoms and Limnodrilus oligochaetes in the vertical sediment profile. The dispersion of Limnodrilus oligochaetes with sediment depth profile varied from quasi‐random (i.e. exponent k of the negative binomial distribution >2.0 or <0) at the surface to strong aggregation (0 < k < 1.0) in the deeper sediments. Diatoms were heterogenous, with most species contributing less than 1% of all the diatoms collected from the riverbed. Contagious dispersion was a common feature among the diatom species. The distribution of Fragilaria ulna was largely quasi‐random in all sites, with Nitzschia amphibia and Cocconeis placentula demonstrating quasi‐random distribution in the Kerma vertical sediment profile. Escape from stranding to deeper sediment strata as the drought progressed was not a universal response among the diatom species. Our results showed that drought‐stress altered the structure of biological assemblages and also emphasized the need for the management of tropical lotic systems and their catchments for flow permanence. As agents of mortality, viruses and nanoflagellates impact on picoplankton populations. We examined the differences in interactions between these compartments in two French Atlantic bays. Microbes, considered here as central actors of the planktonic food web, were first monitored seasonally in Arcachon (2005) and Marennes‐Oléron (2006) bays. Their dynamics were evaluated to categorize trophic periods using the models of Legendre and Rassoulzadegan as a reference framework. Microbial interactions were then compared through 48 h batch culture experiments performed during the phytoplankton spring bloom, identified as herbivorous in Marennes and multivorous in Arcachon. Marennes was spatially homogeneous compared with Arcachon. The former was potentially more productive, featuring a large number of heterotrophic pathways, while autotrophic mechanisms dominated in Arcachon. A link was found between viruses and phytoplankton in Marennes, suggesting a role of virus in the regulation of autotroph biomass. Moreover, the virus–bacteria relation was weaker in Marennes, with a bacterial lysis potential of 2.6% compared with 39% in Arcachon. The batch experiments (based on size‐fractionation and viral enrichment) revealed different microbial interactions that corresponded to the spring‐bloom trophic interactions in each bay. In Arcachon, where there is a multivorous web, flagellate predation and viral lysis acted in an opposite way on picophytoplankton. When together they both reduced viral production. Conversely, in Marennes (herbivorous web), flagellates and viruses together increased viral production. Differences in the composition of the bacterial community composition explained the combined flagellate‐virus effects on viral production in the two bays. 相似文献

Cholera remains epidemic and endemic in the world, causing thousands of deaths annually in locations lacking adequate sanitation and water infrastructure. Yet, its dynamics are still not fully understood. In this paper, we simplify and improve Jensen et al.’s model (PNAS 103:4652–4657, 2006) by incorporating a Minimum Infection Dose (MID) into the incidence term. We perform local stability analysis and provide bifurcation diagrams of the bacterial carrying capacity with or without shedding. Choosing parameters such that the endemic or epidemic equilibrium is unstable (as it is the case in reality), we observe numerically that for the bacterial carrying capacity (K) less than the MID (c), oscillating trajectories exist only in the microbial scale, whereas for \(K>c\) , they exist in both the microbial and population scales. In both cases, increasing pathogen shed rate \(\xi \) increases the amplitude of the trajectories and the period of the trajectories for those that are periodic. Our findings highlight the importance of the relationship among the shedding rates, K, MID, the maximum bacterial growth rate (r) and the features of the disease outbreak. In addition, we identified a region in the parameter space of our model that leads to chaotic behaviour. This could be used to explain the irregularity in the seasonal patterns of outbreaks amongst different countries, especially if the positive relationship between bacterial proliferation and temperature is considered. 相似文献
