The synchronization of different γ-rhythms arising in different brain areas has been implicated in various cognitive functions. Here, we focus on the effect of the ubiquitous neuronal heterogeneity on the synchronization of ING (interneuronal network gamma) and PING (pyramidal-interneuronal network gamma) rhythms. The synchronization properties of rhythms depends on the response of their collective phase to external input. We therefore determine the macroscopic phase-response curve for finite-amplitude perturbations (fmPRC) of ING- and PING-rhythms in all-to-all coupled networks comprised of linear (IF) or quadratic (QIF) integrate-and-fire neurons. For the QIF networks we complement the direct simulations with the adjoint method to determine the infinitesimal macroscopic PRC (imPRC) within the exact mean-field theory. We show that the intrinsic neuronal heterogeneity can qualitatively modify the fmPRC and the imPRC. Both PRCs can be biphasic and change sign (type II), even though the phase-response curve for the individual neurons is strictly non-negative (type I). Thus, for ING rhythms, say, external inhibition to the inhibitory cells can, in fact, advance the collective oscillation of the network, even though the same inhibition would lead to a delay when applied to uncoupled neurons. This paradoxical advance arises when the external inhibition modifies the internal dynamics of the network by reducing the number of spikes of inhibitory neurons; the advance resulting from this disinhibition outweighs the immediate delay caused by the external inhibition. These results explain how intrinsic heterogeneity allows ING- and PING-rhythms to become synchronized with a periodic forcing or another rhythm for a wider range in the mismatch of their frequencies. Our results identify a potential function of neuronal heterogeneity in the synchronization of coupled γ-rhythms, which may play a role in neural information transfer via communication through coherence. 相似文献

N 6‐methyladenosine (m6A) is a chemical modification present in multiple RNA species and is most abundant in mRNAs. Studies on m6A reveal its comprehensive roles in almost every aspect of mRNA metabolism, as well as in a variety of physiological processes. Although some recent discoveries indicate that m6A can affect the life cycles of numerous viruses as well as the cellular antiviral immune response, the roles of m6A modification in type I interferon (IFN‐I) signaling are still largely unknown. Here, we reveal that WT1‐associated protein (WTAP), one of the m6A “writers”, is degraded via the ubiquitination‐proteasome pathway upon activation of IFN‐I signaling. With the degradation of WTAP, the m6A levels of IFN‐regulatory factor 3 (IRF3) and interferon alpha/beta receptor subunit 1 (IFNAR1) mRNAs are reduced, leading to translational suppression of IRF3 and instability of IFNAR1 mRNA. Thus, the WTAP‐IRF3/IFNAR1 axis may serve as negative feedback pathway to fine‐tune the activation of IFN‐I signaling, which highlights the roles of m6A in the antiviral response by dictating the fate of mRNAs associated with IFN‐I signaling. 相似文献

The aim of this study was to identify novel prognostic mRNA and microRNA (miRNA) biomarkers for hepatocellular carcinoma (HCC) using methods in systems biology. Differentially expressed mRNAs, miRNAs, and long non-coding RNAs (lncRNAs) were compared between HCC tumor tissues and normal liver tissues in The Cancer Genome Atlas (TCGA) database. Subsequently, a prognosis-associated mRNA co-expression network, an mRNA–miRNA regulatory network, and an mRNA–miRNA–lncRNA regulatory network were constructed to identify prognostic biomarkers for HCC through Cox survival analysis. Seven prognosis-associated mRNA co-expression modules were obtained by analyzing these differentially expressed mRNAs. An expression module including 120 mRNAs was significantly correlated with HCC patient survival. Combined with patient survival data, several mRNAs and miRNAs, including CHST4, SLC22A8, STC2, hsa-miR-326, and hsa-miR-21 were identified from the network to predict HCC patient prognosis. Clinical significance was investigated using tissue microarray analysis of samples from 258 patients with HCC. Functional annotation of hsa-miR-326 and hsa-miR-21-5p indicated specific associations with several cancer-related pathways. The present study provides a bioinformatics method for biomarker screening, leading to the identification of an integrated mRNA–miRNA–lncRNA regulatory network and their co-expression patterns in relation to predicting HCC patient survival. 相似文献

Multiple Sclerosis (MS) is the most common progressive and disabling neurological condition affecting young adults in the world today. From a genetic point of view, MS is a complex disorder resulting from the combination of genetic and non-genetic factors. We aimed to identify previously unidentified loci conducting a new GWAS of Multiple Sclerosis (MS) in a sample of 296 MS cases and 801 controls from the Spanish population. Meta-analysis of our data in combination with previous GWAS was done. A total of 17 GWAS-significant SNPs, corresponding to three different loci were identified:HLA, IL2RA, and 5p13.1. All three have been previously reported as GWAS-significant. We confirmed our observation in 5p13.1 for rs9292777 using two additional independent Spanish samples to make a total of 4912 MS cases and 7498 controls (ORpooled = 0.84; 95%CI: 0.80-0.89; p = 1.36 × 10-9). This SNP differs from the one reported within this locus in a recent GWAS. Although it is unclear whether both signals are tapping the same genetic association, it seems clear that this locus plays an important role in the pathogenesis of MS. 相似文献
