目的观察人乳腺细胞系及乳腺组织中SIAH2和P-ERK表达的变化,探讨乳腺癌中SIAH2与P-ERK的关联。方法应用免疫组织化学检测140例乳腺石蜡包埋组织中SIAH2和P-ERK的表达,Western blot检测人乳腺细胞系和23例乳腺浸润性导管癌及癌旁正常乳腺组织中SIAH2和P-ERK的蛋白表达情况,人乳腺癌细胞系表达SIAH2-siRNA后,P-ERK蛋白表达情况。结果乳腺癌中SIAH2和P-ERK阳性率与组织学分级相关(P<0.05),SIAH2和P-ERK呈正相关性。人乳腺癌细胞系以及乳腺癌组织中SIAH2和P-ERK蛋白表达量明显高于人乳腺正常上皮细胞系与癌旁正常乳腺组织中SIAH2和P-ERK蛋白表达量(P<0.05);表达SIAH2-siRNA的乳腺癌细胞系与对照组相比,P-ERK蛋白表达明显减少(P<0.05)。结论 SIAH2、P-ERK过表达与乳腺癌的组织学分级有关。在乳腺癌细胞系中抑制SIAH2表达后,P-ERK表达也减少,因此抑制SIAH2可以抑制ERK通路。 相似文献

Life history theory predicts that individuals will allocate resources to different traits so as to maximize overall fitness. Because conditions experienced during early development can have strong downstream effects on adult phenotype and fitness, we investigated how four species of synovigenic, larval-pupal parasitoids that vary sharply in their degree of specialization (niche breadth) and life history (Diachasmimorpha longicaudata, Doryctobracon crawfordi, Opius hirtus and Utetes anastrephae), allocate resources acquired during the larval stage towards adult reproduction. Parasitoid larvae developed in a single host species reared on four different substrates that differed in quality. We measured parasitoid egg load at the moment of emergence and at 24 h, egg numbers over time, egg size, and also adult size. We predicted that across species the most specialized would have a lower capacity to respond to changes in host substrate quality than wasps with a broad host range, and that within species, females that emerged from hosts that developed in better quality substrates would have the most resources to invest in reproduction. Consistent with our predictions, the more specialized parasitoids were less plastic in some responses to host diet than the more generalist. However, patterns of egg load and size were variable across species. In general, there was a remarkable degree of reproductive effort-allocation constancy within parasitoid species. This may reflect more “time-limited” rather than “egg-limited” foraging strategies where the most expensive component of reproductive success is to locate and handle patchily-distributed and fruit-sequestered hosts. If so, egg costs, independent of degree of specialization, are relatively trivial and sufficient resources are available in fly larvae stemming from all of the substrates tested. 相似文献

Effects of avian gut-passage on seed germination are important to assess the effectiveness of frugivores in woodland regeneration,
particularly in biodiversity hotspots that have a high incidence of avian frugivory. We examined the effect of avian gut-passage
on seed germination in contrast to seeds that remain uneaten in five shrub species in Mediterranean central Chile and sought
to determine the physiological mechanism(s) by which seed germinability is modified. Germination assays were conducted in
a glasshouse for five common shrub species of the sub-Andean matorral: Azara dentata (Flacourtiaceae), Schinus polygamus and Schinus molle (Anacardiaceae), Cestrum parqui (Solanaceae), and Maytenus boaria (Celastraceae). We estimated germinability (final percent germination), dormancy length (time from sowing to first germination),
mean length of dormancy of all germinated seeds, and contrasted germination rates of defecated versus manually extracted and
pulp-enclosed seeds. Avian gut-passage increased seed germinability in four of the five shrub species studied—primarily through
deinhibition via pulp removal. Minimum dormancy length was not modified by avian gut-passage for A. dentata, but was significantly shorter for S. molle and C. parqui. Mean dormancy length was significantly shorter in gut-passed seeds of A. dentata, S. molle and M. boaria. Avian gut-passage greatly enhanced the seed germination rates of three species, A. dentata, S. molle and C. parqui. We conclude that the positive effects of birds on seed germination facilitate the regeneration of sub-Andean shrublands,
and that bird declines due to landscape change may impair recovery rates of successional or restored areas due to dispersal

城市森林是城市中主要的生态功能景观,已经成为城市生态环境建设的主体。基于居民方便获取城市森林服务的要求,根据大型城市森林斑块、道路网和街区单元数据,利用ArcGIS9.2的Network Analyst工具,分析了广州市中心城区在2km步行距离内街区单元到达城市森林斑块的可达性以及城市森林斑块的承载力,并结合航片解译的树冠覆盖数据对具有不同可达性的街区单元进行了分析。分析结果表明:广州中心城区的外环高速以内的区域范围,分别有73%、46%和18%的的街区单元在步行2km、1km和0.5km距离内能够到达大型城市森林绿地斑块。从街区单元的行政归属看,以越秀区的街区可达性最好,而海珠区和原芳村区的街区可达性较差。研究范围内,0.5km内不能到达大型城市森林斑块且树冠覆盖超过30%的街区类型中,以公共管理单位类型为主,单位绿化对于树冠覆盖的贡献率大;而绝大多数以住宅类型为主的街区单元的树冠覆盖率低于10%,其中64%的街区树冠覆盖率低于5%。以街区单元为基本对象进行可达性度量,分析城市森林斑块现状分布和服务状况,并结合其树冠覆盖特征,可以明确需要重点关注区域的空间分布,可为广州市在较小尺度上的城市森林空间布局优化提供参考。 相似文献

Willi Hennig's (Beitr. Ent. 1960, 10, 15) Die Dipteren-Fauna von Neuseeland als systematisches und tiergeographisches Problem applied a phylogenetic approach to examine the distributional patterns exhibited by the Diptera of New Zealand. Hennig showed how phylogenetic trees may be used to infer dispersal, based on the progression and deviation rules, and also discussed the existence of vicariance patterns. The most important author who applied Hennig's phylogenetic biogeography was Lars Brundin, when analysing the phylogenetic relationships of two taxa of Chironomidae (Diptera) and using them to examine the biogeographic relationships of Australia, New Zealand, South America and South Africa. The relevance of Brundin's contribution was noted by several authors, as it began the cladistic or vicariance approach to biogeography, that implies the discovery of vicariance events shared by different monophyletic groups. Both phylogenetic and cladistic biogeography have a place in contemporary biogeography, the former for analysing taxon biogeography and the latter when addressing Earth or biota biogeography. The recent use of the term “phylogenetic biogeography” to refer to a posteriori methods of cladistic biogeography is erroneous and should be avoided. 相似文献

The potato cyst nematodes (PCNs) Globodera pallida and Globodera rostochiensis are important parasites of potato. PCNs undergo complex biotrophic interactions with their hosts that involve gene expression changes in both the nematode and the host plant. The aim of this study was to determine key genes that are differentially expressed in Globodera pallida life cycle stages and during the initiation of the feeding site in susceptible and partially resistant potato genotypes. For this purpose, two microarray experiments were designed: (i) a comparison of eggs, infective second‐stage juveniles (J2s) and sedentary parasitic‐stage J2s (SJ2); (ii) a comparison of SJ2s at 8 days after inoculation (DAI) in the susceptible cultivar (Desirée) and two partially resistant lines. The results showed differential expression of G. pallida genes during the stages studied, including previously characterized effectors. In addition, a large number of genes changed their expression between SJ2s in the susceptible cultivar and those infecting partially resistant lines; the number of genes with modified expression was lower when the two partially resistant lines were compared. Moreover, a histopathological study was performed at several time points (7, 14 and 30 DAI) and showed the similarities between both partially resistant lines with a delay and degeneration in the formation of the syncytia in comparison with the susceptible cultivar. Females at 30 DAI in partially resistant lines showed a delay in their development in comparison with those in the susceptible cultivar. 相似文献

Background
Eradication of bovine tuberculosis (bTB) through the application of test-and-cull programs is a declared goal of developed countries in which the disease is still endemic. Here, longitudinal data from more than 1,700 cattle herds tested during a 12?year-period in the eradication program in the region of Madrid, Spain, were analyzed to quantify the within-herd transmission coefficient (??) depending on the herd-type (beef/dairy/bullfighting). In addition, the probability to recover the officially bTB free (OTF) status in infected herds depending on the type of herd and the diagnostic strategy implemented was assessed using Cox proportional hazard models.Results
Overall, dairy herds showed higher ?? (median 4.7) than beef or bullfighting herds (2.3 and 2.2 respectively). Introduction of interferon-gamma (IFN-??) as an ancillary test produced an apparent increase in the ?? coefficient regardless of production type, likely due to an increase in diagnostic sensitivity. Time to recover OTF status was also significantly lower in dairy herds, and length of bTB episodes was significantly reduced when the IFN-?? was implemented to manage the outbreak.Conclusions
Our results suggest that bTB spreads more rapidly in dairy herds compared to other herd types, a likely cause being management and demographic-related factors. However, outbreaks in dairy herds can be controlled more rapidly than in typically extensive herd types. Finally, IFN-?? proved its usefulness to rapidly eradicate bTB at a herd-level. 相似文献949.

Holliday junction (HJ) resolution is a fundamental step for completion of homologous recombination. HJ resolving enzymes (resolvases) distort the junction structure upon binding and prior cleavage, raising the possibility that the reactivity of the enzyme can be affected by a particular geometry and topology at the junction. Here, we employed a DNA origami nano-scaffold in which each arm of a HJ was tethered through the base-pair hybridization, allowing us to make the junction core either flexible or inflexible by adjusting the length of the DNA arms. Both flexible and inflexible junctions bound to Bacillus subtilis RecU HJ resolvase, while only the flexible junction was efficiently resolved into two duplexes by this enzyme. This result indicates the importance of the structural malleability of the junction core for the reaction to proceed. Moreover, cleavage preferences of RecU-mediated reaction were addressed by analyzing morphology of the reaction products. 相似文献
