Summary Fetal bovine serum (FBS) or heat-inactivated FBS (56° C for 30 min, HFBS) caused a dose-dependent decrease in the transepithelial
electrical resistance of an epithelial monolayer (MDCK). A saturating concentration of HFBS (30%) caused an average fall of
25 ± 2% within 60 min. Upon removal of HFBS, the resistance returned to its starting value within 1 h. Flux studies with [3H]mannitol demonstrate that the fall in resistance is due to an increased permeability of the tight junctions. Thirty percent
heat inactivated sera from goat, newborn calf, calf, bovine, and horse caused falls ranging from 26 to 47%. In contrast with
the basolateral preference of human and bovine adult sera, fetal bovine and newborn calf sera elicit this response primarily
by interacting with the apical surface of the epithelium. HFBS-treated monolayers show a significant increase in the condensation
of F-actin at points where ≥3 cells meet. These results demonstrate that FBS and other sera used as nutritional supplements

In the production of biopharmaceuticals such as monoclonal antibodies (mAbs) and vaccines, the residual amounts of host-cell proteins (HCPs) are among the critical quality attributes. In addition to overall HCP levels, individual HCPs may elude purification, potentially causing issues in product stability or patient safety. Such HCP persistence has been attributed mainly to biophysical interactions between individual HCPs and the product, resin media, or residual chromatin particles. Based on measurements on process streams from seven mAb processes, we have found that HCPs in aggregates, not necessarily chromatin-derived, may play a significant role in the persistence of many HCPs. Such aggregates may also hinder accurate detection of HCPs using existing proteomics methods. The findings also highlight that certain HCPs may be difficult to remove because of their functional complementarity to the product; specifically, chaperones and other proteins involved in the unfolded protein response (UPR) are disproportionately present in the aggregates. The methods and findings described here expand our understanding of the origins and potential behavior of HCPs in cell-based biopharmaceutical processes and may be instrumental in improving existing techniques for HCP detection and clearance. 相似文献

Plants have evolved a broad array of defense mechanisms involved in disease resistance. These include synthesis of phytoalexin antibiotics and proteinase inhibitors, deposition of cell wall materials, and accumulation of hydrolytic enzymes such as chitinases. Resistance appears to depend on the ability of the host to recognize the pathogen rapidly and induce these defense responses in order to limit pathogen spread. Application of molecular technologies has yielded significant new information on mechanisms involved in pathogen recognition, signal transduction, and defense-related gene activation, and is leading to novel strategies for engineering enhanced disease resistance. We are using these approaches to analyze regulation of 3-hydroxy-3-methylglutaryl CoA reductase (HMGR), a key enzyme mediating the production of terpenoid defense compounds. This enzyme is encoded by four genes in tomato; hmg2 gene expression is specifically associated with responses to pathogen or defense elicitors. Transgenic plants containing DNA constructs that fuse the hmg2 promoter to a reporter gene have been used to analyze both tissue specificity and patterns of defense-related expression. Because this gene is rapidly induced in tissues directly surrounding the site of ingress by a variety of pathogens, it may serve as a valuable tool in engineering new disease-resistance mechanisms. 相似文献

Two regions involved in catechol biosynthesis (cbs) of Erwinia carotovora W3C105 were cloned by functional complementation of Escherichia coli mutants that were deficient in the biosynthesis of the catechol siderophore enterobactin (ent). A 4.3-kb region of genomic DNA of E. carotovora complemented the entB402 mutation of E. coli. A second genomic region of 12.8 kb complemented entD, entC147, entE405, and entA403 mutations of E. coli. Although functions encoded by catechol biosynthesis genes (cbsA, cbsB, cbsC, cbsD, and cbsE) of E. carotovora were interchangeable with those encoded by corresponding enterobactin biosynthesis genes (entA, entB, entC, entD, and entE), only cbsE hybridized to its functional counterpart (entE) in E. coli. The cbsEA region of E. carotovora W3C105 hybridized to genomic DNA of 21 diverse strains of E. carotovora but did not hybridize to that of a chrysobactin-producing strain of Erwinia chrysanthemi. Strains of E. carotovora fell into nine groups on the basis of sizes of restriction fragments that hybridized to the cbsEA region, indicating that catechol biosynthesis genes were highly polymorphic among strains of E. carotovora. 相似文献

Crystals of the channel-forming domain of colicin E1 from E. coli were grown by vapor diffusion at pH 6.4 and higher pH values. Cleavage of the colicin molecule with trypsin or thermolysin produced two of the pore-forming polypeptides used in these experiments. The third polypeptide was purified from a constructed plasmid that overexpresses only the C-terminal domain of colicin E1. Polypeptide crystals are tetragonal with space group I4, have one monomer in the asymmetric unit, and diffract to 2.2–2.4 Å. Unit cell parameters for the tryptic and thermolytic polypeptides are a = 102.9 Å and c = 35.6 Å. Crystals of the overexpressed polypeptide have unit cell parameters of a =87.2 Å and c =59.1 Å. The crystals were characterized by precession photography, and native data sets of each channel-forming fragment were collected on a Siemens-Nicolet area detector. The crystallization and characterization of these polypeptides are the first steps in the structure determination of the channel-forming domain of colicin E1. © 1994 Wiley-Liss, Inc. 相似文献

The transition of the colicin E1 channel polypeptide from a water-soluble to membrane-bound state occurs in vitro at acid pH values that are associated with an unfolded channel structure whose properties qualitatively resemble those of a "molten globule," or "compact unfolded," intermediate state. The role of such a state for activity was tested by comparing the pH dependence of channel-induced solute efflux and the amplitude of the near-UV CD spectrum. The requirement of a partly unfolded state for activity was shown by the coincidence of the onset of channel activity measured for 4 different lipid compositions with the decrease in near-UV CD amplitude as a function of pH. Tertiary constraints on the 3 tryptophans of the colicin channel, assayed by the amplitude of the near-UV CD spectrum, are retained over the pH range 3-4 where channel activity could be measured and, as well, at pH 2. In addition, the tryptophan fluorescence emission spectrum is virtually unchanged over the pH range 2-6. The temperature independence of the near-UV spectrum at pH 3-6 up to 70 degrees C implies that the colicin E1 channel polypeptide is more stable than that of colicin A. A transition between 53 and 58 degrees C in the amplitude of the near-UV CD is consistent with preservation of part of the hydrophobic core in a destabilized state at pH 2. Thus, the unfolded state associated with colicin activity at acidic pH has the properties of a "compact unfolded" state, having some, but not all of the properties of a "molten globule."(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

The degradation of trichloroethylene (TCE, Cl2C=CHCl) and tetrachloroethylene (PCE, Cl2C=CCI2) in a gas stream from a soil vapor extraction (SVE) well was demonstrated with an annular photocatalytic reactor packed with porous TiO2 pellets in a field trial at the Savannah River Site in Aiken, SC. The TiO2 pellets were prepared using a sol‐gel method. The experiments were performed at 55 to 60°C using space times of 108 to 1010 g ? s ? mol‐1 for TCE and PCE. Chloroform (CHCl3) and carbon tetrachloride (CCl4) were detected as minor products from side reactions. On a molar basis, the amounts CCl4 and CHCl3 produced were about 2 and 0.2% of the reactants, respectively. 相似文献

Various types of noncontingent feedback have been used as control procedures in EMG training; however, their effects on such training have received little attention. Experiment 1 in the present study examined the effects of noncontingent feedback on EMG training, and Experiment 2 assessed the effects of feedback characteristics on EMG responses. In Experiment 1, three noncontingent feedback groups (yoked control, randomly fluctuating tones, and decreasing tones) and one contingent group underwent 20 minutes of training for frontal EMG decreases. Procedures in Experiment 2 were identical to those in Experiment 1 except that subjects were instructed merely to listen to the feedback tones. Results of Experiment 1 indicated that contingent and noncontingent fluctuating feedback groups achieved significantly lower EMG levels than noncontingent decreasing and yoked control groups. In Experiment 2, however, no differences in EMG activity were found among groups. In both experiments, groups did not differ in terms of subjective variables such as frustration, suspiciousness about the tone, or length of time attending to the tone. Results of these two experiments suggest that differences in EMG responses to various types of noncontingent feedback result from interactions between characteristics of the feedback stimulus and instructions to decrease the stimulus.This research was supported by Ohio University Research Grants No. 9147 and No. 9155 to the first author. 相似文献
