An insertion mutation was isolated that resulted in derepressed expression of the Bacillus subtilis dipeptide transport operon (dpp) during the exponential growth phase in rich medium. DNA flanking the site of insertion was found to encode an operon (codVWXY) of four potential open reading frames (ORFs). The deduced product of the codV ORF is similar to members of the λ Int family; CodW and CodX are homologous to HsIV and HsIU, two putative heat-shock proteins from Escherichia coli, and to LapC and LapA, two gene products of unknown function from Pasteurella haemolytica. CodX also shares homology with a family of ATPases, including CIpX, a regulatory subunit of the E. coli ClpP protease. CodY does not have any homologues in the databases. The insertion mutation and all previously isolated spontaneous cod mutations were found to map In codY. In-frame deletion mutations in each of the other cod genes revealed that only codY is required for repression of dpp in nutrient-rich medium. The cody mutations partially relieved amino acid repression of the histidine utilization (hut) operon but had no effect on regulation of certain other early stationary phase-induced genes, such as spoVG and gsiA.  相似文献

Abstract A spontaneous Thiosphaera pantotropha mutant (Tp9002) that is able to grow on methanol has been isolated. With hybridization experiments it has been demonstrated that mxaF , the gene encoding the large subunit of methanol dehydrogenase, is absent from T. pantotropha . In Tp9002, a dye-linked enzyme activity was found with a substrate specificity similar to that of the dye-linked ethanol dehydrogenase from Pseudomonas aeruginosa . The N-terminus of a 26-kDa cytochrome c , exclusively synthesized in Tp9002, is homologous to the N-terminus of the electron acceptor of ethanol dehydrogenase. These results suggest that in Tp9002 a dye-linked ethanol dehydrogenase is responsible for methanol oxidation, using a 26-kDa cytochrome c as electron acceptor.  相似文献

A method has been developed for growing Physarum polycephalum plasmodia that are 8 to 10 times larger than those obtained in the petri dish cultures used by Nygaard, Guttes, and Rusch. In the large-scale procedure, plasmodia were grown in metal trays on a membrane supported by filter paper on stainless-steel screen. Plasmodia were started from a ring of inoculum to allow inward and outward migration and were incubated on a rocker so that nutrient medium would flow back and forth, wetting the undersurface of the plasmodium. Rocker and petri dish cultures had similar growth characteristics: (i) the interphase time between mitoses I and II and between II and III was about 8 hr; (ii) ribonucleic acid and protein increased essentially logarithmically throughout the cell cycle; and (iii) deoxyribonucleic acid increased only during early interphase and it doubled in approximately 3 hr after each mitosis. Rocker cultures were not as nearly synchronous as petri dish cultures and had a range in metaphase time (at mitosis III) within individual plasmodia of 15 to 45 min, as compared with 5 to 10 min in petri dish cultures.  相似文献

Drepanopeziza ribis causes the leaf spot disease of blackcurrant ( Ribes nigrum ) and may lead to severe premature leaf-fall. Artificial inoculation studies were carried out to investigate infection of leaves by D. ribis conidia in relation to environmental conditions and leaf position (age) on cvs. Baldwin and Ben Hope in April and July 2007. All leaves on a number of selected extension shoots on potted three-year old plants were inoculated with conidia and then incubated under different conditions: 10, 17.5 and 25°C each with five wet periods (4, 8, 12, 24 and 30 h). Number of infected leaves was determined. The two cultivars differed significantly in their susceptibility to conidial infection: cv. Baldwin was much more susceptible than cv. Ben Hope. Older leaves on extension shoots were more susceptible to conidial infection than younger leaves. Increasing length of wetness duration led to increasing incidence of leaves infected, particularly when inoculated in July. However, the effects of temperature were inconclusive and generally very small in comparison with other factors. Field epidemics were monitored over three years (2005–07). Field data confirmed the main findings from controlled inoculation studies: severe disease was associated with very wet conditions and older leaves. Furthermore, they also suggested that significant disease increase only occurred from late July onwards.  相似文献

Cordimus, a new genus of cricetid rodent, is described from Neogene deposits on the islands of Curaçao and Bonaire, Dutch Antilles. The genus is characterized by strongly cuspidate molars, the presence of mesolophs in most upper molars and the absence of mesolophids in lower molars. Similarities with the early cricetid Copemys from the Miocene of North America coupled with apparent derived characters shared with the subfamily Sigmodontinae suggest that Cordimus may be close to the root of the sigmodontine lineage, a possibility that remains to be tested through explicit phylogenetic analysis. Three species are recognized on the basis of size and details of molar morphology. Cordimus hooijeri sp. nov. is described from Bonaire on the basis of Holocene owl pellet material that consists of dentaries and postcranial material only. This species is presumed to be extinct, but focused surveys are needed to confirm this hypothesis. Cordimus debuisonjei sp. nov. and Cordimus raton sp. nov. are described from deposits on Tafelberg Santa Barbara in Curaçao. Although the age of these deposits is not known, they are most likely of late Pliocene or early Pleistocene age. Both are represented by numerous isolated molars and some osteological material.  相似文献
