Rabbit kidneys were perfused at 10 °C with a solution containing gelatin polypeptides (Haemaccel), and glycerol was introduced, and then removed, using a technique that has previously been shown to result in viable kidneys. This involved increasing the concentration of glycerol in the perfusate from zero to a maximum of 3 , holding it at this level for 30 min, and then decreasing it at the same rate to < 0.1 . Measurements were made of the concentration of glycerol in cortex, cortico-medullary zone, and medulla at various stages of perfusion. During the experiments it was observed that vascular resistance increased dramatically toward the end of deglycerolization, and changes in regional perfusate flow were measured by the diffusable indicator method. It was found that renal tissue is effectively permeated by glycerol using this technique. The perfusate flow throughout all regions of the kidney was reduced during deglycerolization but the greatest effect was on cortico-medullary flow, which was found to be abnormally high during the initial stages of hypothermic perfusion, but was severely impaired when the glycerol was removed. The cryoprotectant was almost completely removed by the washout procedure adopted.  相似文献

It is known that insulin is adsorbed onto glass but it has been assumed that it is not adsorbed onto plastic. We find that tritium-labelled insulin is adsorbed onto all materials tried. The adsorption is reduced in the presence of other proteins and can be prevented altogether by coating the vessels with cetyl alcohol.  相似文献

The predominant ureolytic bacteria in the pig large intestine were determined while growing pigs were fed a basal diet or basal diet plus copper sulfate, Aureo SP250, or clinoptilolite. Fecal samples were collected from four pigs fed each diet at 3, 9, and 14 weeks and analyzed for total colony counts and percent ureolytic bacteria. Fecal urease activity, ammonia nitrogen, and identity of the ureolytic bacteria were determined at 14 weeks. Copper sulfate and Aureo SP250 reduced the number of ureolytic organisms, with a marked decrease occurring in the Streptococcus spp., which made up 74% of the ureolytic isolates from the pigs on the basal diet. Other ureolytic species detected at lower concentrations were Staphylococcus spp., Selenomonas ruminantium, Bacteroides multiacidus, and Eubacterium limosum. Copper sulfate also reduced fecal urease activity (P less than 0.10). Fecal ammonia concentrations were not different between pigs fed the various diets. These data suggest that the streptococci are the most numerous ureolytic species in the pig intestinal tract and are significantly reduced by copper sulfate and Aureo SP250; however, only copper sulfate reduced intestinal urease activity.  相似文献

Neutrophils undergo rapid morphological changes as well as metabolic perturbations when stimulated with certain phorbol esters. Stimulated cells initially exhibit pronounced projections emanating from the cell bodies, followed by rounding of the cells, reduction in granule number, and the appearance of intracellular vesicles. We show these vesicles to be derived, at least in part, from the plasmalemma. The experimental approach involved labeling stimulated and unstimulated cells with native ferritin and cationized ferritin, along with the cytochemical localization of ecto-5'-nucleotidase. The labeling patterns of the vesicles indicate that these structures are involved in both phorbol ester-stimulated adsorptive and fluid-phase endocytosis. Neutrophils stimulated with 12-O-tetradecanoyl-phorbol-13-acetate (TPA) exhibit two distinct rates of superoxide release in which the second, prolonged level is approximately 50% of the initial rate. All-trans-retinal, which we have recently shown to stimulate O2- release but not granule exocytosis or cell vesiculation, induces a single prolonged rate of maximal O2- release. Neutrophils treated with both all-trans-retinal and TPA exhibit only a single sustained rate of maximal O2- release similar to that observed with all-trans-retinal alone. Moreover, treatment of cells with all-trans-retinal blocks the vesiculation of neutrophils induced by TPA in a dose-dependent manner. This observation provides a possible explanation for the differences in the kinetics of superoxide release.  相似文献

A modification of the second-order model for biodegradation was derived, applied to an example data set, and shown to be superior for describing the anaerobic biodegradation of p-cresol by an enriched bacterial consortium. The modified model circumvents the no-growth assumption implicit in the use of the second-order rate equation, but still requires the assumption of first-order kinetics over the course of substrate depletion. Violation of the no-growth assumption is particularly important since overestimates of the pseudo-first-order rate coefficient lead to underestimates of the time required for the removal of a xenobiotic chemical from a contaminated environment. Our calculations show that the errors introduced into the pseudo-first-order rate coefficient (and the resulting estimates of the second-order rate coefficient) approach 100% if one doubling occurs in activity over the course of substrate depletion. For an exemplary data set, use of a first-order model resulted in a 100% overestimate of the first-order decay coefficient, which would in turn lead to a corresponding overestimate of the second-order rate coefficient. The modified model we describe is a potential alternative to the pseudo-first-order model for the routine estimation of second-order rate coefficients.  相似文献

Summary Fifty four microcosmic communities were assembled over 4 months from a 28-species source pool of phytoplankton using nine different invasion patterns each replicated six times. Three communities from each set of replicates then were invaded with a cladoceran that feeds on phytoplankton. All communities were then treated identically for an additional 4 months. In all nine invasion categories species richness was greater in predated communities. Predation opened communities to invasion by increasing the representation of infrequently sampled species at the expense of more common species. Invasion rate was four times more influential than predation and over eleven times more important than either invasion order or the timing pattern of interspecific arrivals in determining species richness in this system of communitites.  相似文献
