In this study, the effect of pharmacological inhibition of catecholaminergic activity on hCG-induced spawning was evaluated and correlated with tyrosine hydroxylase (TH) activity, the rate-limiting enzyme in catecholamine biosynthesis. Gravid female H. fossilis collected in both prespawning and spawning phases were given alpha-methylparatyrosine (alpha-MPT: 250 microg/g body weight, ip, an irreversible inhibitor of TH) and human chorionic gonadotropin (hCG: 100 IU/fish, ip) alone or in combination. The fish were sampled at different intervals for measuring hypothalamic and ovarian TH activity and checking spawning response. The administration of hCG resulted in ovulation and spawning in both phases with a higher response in the spawning phase. The administration of alpha-MPT did not induce any response, like the control fish. In the hCG + alpha-MPT groups, the spawning response of hCG was significantly inhibited and delayed by the inhibitor. The spawning response of hCG was accompanied by a significant increase in both hypothalamic and ovarian TH activity at 6 and 12 h of the injection. However, at 24hr the activity decreased except in the spawning phase. The alpha-MPT treatment inhibited TH activity significantly in a duration-dependent manner. In the hCG + alpha-MPT groups, enzyme activity was inhibited at all duration. The results indicate the involvement of catecholamines during the hCG-induced spawning and the specific functional nature of the involvement needs further investigation. 相似文献

Bis-bidentate Schiff base ligand L and its two mononuclear complexes [CuL(CH3CN)2]ClO4 (1) and [CuL(PPh3)2]ClO4 (2) have been prepared and thoroughly characterized by elemental analyses, IR, UV-Vis, NMR spectroscopy and X-ray diffraction analysis. In both the complexes the metal ion auxiliaries adopt tetrahedral coordination environment. Their reactivity, electrochemical and photophysical behavior have been studied. Complex 1 shows reversible CuII/I couple with potential 0.74 V versus Ag/AgCl in CH2Cl2. Invasive plants and urban run‐off constrain efforts to restore sedge meadow wetlands. We asked if native graminoids can self‐restore following the removal of Typha × glauca (hybrid cattail), and if not, what limits their recovery? After we harvested Typha and depleted its rhizome starch reserves, Carex spp. expanded vegetatively (approximately 1 m over 2 years) but not by recruiting seedlings. A seedling emergence experiment showed that seed banks were depleted where Typha had eliminated the sedge meadow over a decade ago (based on aerial photo analysis). Carex seedling emergence was 75–90% lower where Carex was absent than where it remained in the plant community, and at least 17 species that were abundant 30 years ago were absent from the seed bank and extant vegetation. By varying hydroperiod, we showed that prolonged flooding prevented emergence of Carex seedlings and that a fluctuating hydroperiod reduced emergence and ultimately killed all Carex seedlings. In contrast, Typha seedlings emerged and survived regardless of hydroperiod. Thus, slow vegetative expansion by Carex, depauperate seed banks, and altered hydroperiods all constrain self‐restoration. To compensate for multiple constraints on self‐restoration, we recommend a long‐term management approach that capitalizes on flooding and the capacity of Carex spp. to regrow vegetatively. We suggest annually harvesting swaths of Typha at the edges of clones, before or during flood events, to allow gradual, vegetative self‐restoration of Carex spp. 相似文献

active fractions corresponding to the peaks E and F were eluted at the retention times of 16.8 and 18 min, respectively. Fourier
transform infrared spectroscopy and matrix assisted laser desorption ionization time of flight (MALDI-ToF) mass spectral analysis
showed that the produced lipopeptides belonged to surfactin family. In the MALDI-ToF spectrum, the major molecular mass of
the HPLC purified isoforms were identified at m/z 1,044 and 1,058 Da, respectively. In addition, this marine strain also produced new variants of surfactins with molecular
weights ranging from m/z 1,066 to 1,098 Da, which have not been reported earlier. Both surface-active fractions were found to have potent antimicrobial

After initial small-scale experiments, a 25.0 l rotating disk bioreactor (RDBR) was investigated for the cultivation of a
biofilm-forming salt-tolerant Streptomyces sp. MS1/7, producing an antimicrobial compound. Peak activity attainment rate, PAAR (ratio of the peak antimicrobial activity,
PAMA and the time taken to attain PAMA) was determined. Of the three pH values examined (8.0, 9.0 and 10.0) maximum PAAR (1.82 mm/h)
was attained at pH 9.0. Three aeration rates (9.0, 6.0 and 3.0 l/min) were considered at three levels (25, 50 and 75%) of
disk submergence. At the highest aeration rate and 50% submergence level, PAMA (41 mm), PAAR (1.86 mm/h) and biofilm density
(BD, 0.91 g/ml) attained their highest values. At any given aeration rate, PAMA was always higher at 50% submergence level.
This supports our earlier premise that ideal intertidal conditions, 12 h periods of immersion and emersion, promote maximum

Microbial enzymatic reduction of a toxic form of chromium [Cr(VI)] has been considered as an effective method for bioremediation of this metal. This study reports on the in vitro reduction of Cr(VI) using cell-free extracts from a Cr(VI) reducing Bacillus firmus KUCr1 strain. Chromium reductase was found to be constitutive and its activity was observed both in soluble cell fractions (S12 and S150 and membrane cell fraction (P150). The reductase activity of S12 fraction was found to be optimal at 40 microM Cr(VI) with enzyme concentration equivalent to 0.493 mg protein/ml. Enzyme activity was dependent on NADH or NADPH as electron donor; optimal temperature and pH for better enzyme activity were 70 degrees C and 5.6, respectively. The Km value of the reductase was 58.33 microM chromate having a V(max) of 11.42 microM/min/mg protein. The metabolic inhibitor like sodium azide inhibited reductase activity of membrane fraction of the cell-free extract. Metal ions like Cu2+, Co2+, Ni2+ and As3+ stimulated the enzyme but others, such as Ag+, Hg2+, Zn2+, Mn2+, Cd2+ and Pb2+, inhibited Cr(VI) reductase activity. 相似文献

Platform technologies (PT) are techniques or tools that enable a range of scientific investigations and are critical to today''s advanced technology research environment. Once installed, they require specialized staff for their operations, who in turn, provide expertise to researchers in designing appropriate experiments. Through this pipeline, research outputs are raised to the benefit of the researcher and the host institution.1 Platform facilities provide access to instrumentation and expertise for a wide range of users beyond the host institution, including other academic and industry users. To maximize the return on these substantial public investments, this wider access needs to be supported. The question of support and the mechanisms through which this occurs need to be established based on a greater understanding of how PT facilities operate. This investigation was aimed at understanding if and how platform facilities across the Bio21 Cluster meet operating costs. Our investigation found: 74% of platforms surveyed do not recover 100% of direct operating costs and are heavily subsidized by their home institution, which has a vested interest in maintaining the technology platform; platform managers play a major role in establishing the costs and pricing of the facility, normally in a collaborative process with a management committee or institutional accountant; and most facilities have a three-tier pricing structure recognizing internal academic, external academic, and commercial clients. 相似文献

GBM (glioblastoma multiforme) is the most aggressive and invasive form of primary human brain cancer. We recently developed a novel brain cancer model in the inbred VM mouse strain that shares several characteristics with human GBM. Using bioluminescence imaging, we tested the efficacy of CR (calorie restriction) for its ability to reduce tumour size and invasion. CR targets glycolysis and rapid tumour cell growth in part by lowering circulating glucose levels. The VM-M3 tumour cells were implanted intracerebrally in the syngeneic VM mouse host. Approx. 12–15 days post-implantation, brains were removed and both ipsilateral and contralateral hemispheres were imaged to measure bioluminescence of invading tumour cells. CR significantly reduced the invasion of tumour cells from the implanted ipsilateral hemisphere into the contralateral hemisphere. The total percentage of Ki-67-stained cells within the primary tumour and the total number of blood vessels was also significantly lower in the CR-treated mice than in the mice fed ad libitum, suggesting that CR is anti-proliferative and anti-angiogenic. Our findings indicate that the VM-M3 GBM model is a valuable tool for studying brain tumour cell invasion and for evaluating potential therapeutic approaches for managing invasive brain cancer.
