Since the 1980s, cetacean morbillivirus (CeMV) has caused mass mortality events worldwide. However, no epizootics had been recorded in the South Atlantic, until an unusual mortality event (UME) linked to Guiana dolphin cetacean morbillivirus (GD-CeMV) began in Ilha Grande Bay, southeastern Brazil, in November 2017. In a five-month period, the UME spread to neighboring Sepetiba Bay and accounted for the death of at least 277 Guiana dolphins (Sotalia guianensis). Prevalence of morbillivirus positive dolphins, as estimated from RT-PCR diagnostics, was 92.3% (24/26) in Ilha Grande Bay and 91.9% (57/62) in Sepetiba Bay. Females had higher mortality rates during the UME (1.5:1), in contrast with historical mortality data from both bays that showed a 2:1 male to female death ratio. Calf mortality rates also increased in both bays. These results suggest that females and calves were more vulnerable to morbilliviral infection. Herein, we discuss possible explanations for such sex-biased death pattern during the UME and their implication for the conservation of endangered Guiana dolphins. We also speculate about the origin and spread of morbillivirus in the South Atlantic Ocean.  相似文献

Low Reynolds number flows (Re<1) in the human pulmonary acinus are often difficult to assess due to the submillimeter dimensions and accessibility of the region. In the present computational study, we simulated three-dimensional alveolar flows in an alveolated duct at each generation of the pulmonary acinar tree using recent morphometric data. Rhythmic lung expansion and contraction motion was modeled using moving wall boundary conditions to simulate realistic sedentary tidal breathing. The resulting alveolar flow patterns are largely time independent and governed by the ratio of the alveolar to ductal flow rates, Qa/Qd. This ratio depends uniquely on geometrical configuration such that alveolar flow patterns may be entirely determined by the location of the alveoli along the acinar tree. NRAGE (also known as Maged1, Dlxin) is a member of the MAGE gene family that may play a role in the neuronal apoptosis that is regulated by the p75 neurotrophin receptor (p75NTR). To test this hypothesis in vivo, we generated NRAGE knockout mice and found that NRAGE deletion caused a defect in developmental apoptosis of sympathetic neurons of the superior cervical ganglia, similar to that observed in p75NTR knockout mice. Primary sympathetic neurons derived from NRAGE knockout mice were resistant to apoptosis induced by brain-derived neurotrophic factor (BDNF), a pro-apoptotic p75NTR ligand, and NRAGE-deficient sympathetic neurons show attenuated BDNF-dependent JNK activation. Hair follicle catagen is an apoptosis-like process that is dependent on p75NTR signaling; we show that NRAGE and p75NTR show regulated co-expression in the hair follicle and that identical defects in hair follicle catagen are present in NRAGE and p75NTR knockout mice. Interestingly, NRAGE knockout mice have severe defects in motoneuron apoptosis that are not observed in p75NTR knockout animals, raising the possibility that NRAGE may facilitate apoptosis induced by receptors other than p75NTR. Together, these studies demonstrate that NRAGE plays an important role in apoptotic-signaling in vivo.  相似文献

Background and Aims

Sustainable agriculture requires the identification of new, environmentally responsible strategies of crop protection. Modelling of pathosystems can allow a better understanding of the major interactions inside these dynamic systems and may lead to innovative protection strategies. In particular, functional–structural plant models (FSPMs) have been identified as a means to optimize the use of architecture-related traits. A current limitation lies in the inherent complexity of this type of modelling, and thus the purpose of this paper is to provide a framework to both extend and simplify the modelling of pathosystems using FSPMs.

Methods

Different entities and interactions occurring in pathosystems were formalized in a conceptual model. A framework based on these concepts was then implemented within the open-source OpenAlea modelling platform, using the platform''s general strategy of modelling plant–environment interactions and extending it to handle plant interactions with pathogens. New developments include a generic data structure for representing lesions and dispersal units, and a series of generic protocols to communicate with objects representing the canopy and its microenvironment in the OpenAlea platform. Another development is the addition of a library of elementary models involved in pathosystem modelling. Several plant and physical models are already available in OpenAlea and can be combined in models of pathosystems using this framework approach.

Key Results

Two contrasting pathosystems are implemented using the framework and illustrate its generic utility. Simulations demonstrate the framework''s ability to simulate multiscaled interactions within pathosystems, and also show that models are modular components within the framework and can be extended. This is illustrated by testing the impact of canopy architectural traits on fungal dispersal.

Conclusions

This study provides a framework for modelling a large number of pathosystems using FSPMs. This structure can accommodate both previously developed models for individual aspects of pathosystems and new ones. Complex models are deconstructed into separate ‘knowledge sources’ originating from different specialist areas of expertise and these can be shared and reassembled into multidisciplinary models. The framework thus provides a beneficial tool for a potential diverse and dynamic research community.  相似文献

Potyvirus RNA contains at the 5' end a covalently linked virus-encoded protein VPg, which is required for virus infectivity. This role has been attributed to VPg interaction with the eukaryotic translation initiation factor eIF4E, a cap-binding protein. We characterized the dissociation constants for the interaction of the potato virus Y VPg with different plant eIF4Es and its isoforms and mapped the eIF(iso)4E attachment region on VPg. VPg/eIF4E interaction results in the inhibition of cell-free protein synthesis, and we show that it stems from the liberation of the cap moiety from the complex with eIF4E. Since VPg does not attach the cap, it appears that VPg induces changes in the eIF4E structure, diminishing its affinity to the cap. We show here that the initiation complex scaffold protein eIF(iso)4G increases VPg interaction with eIF(iso)4E. These data together suggest similar cap and VPg interactions with eIF4E and characterize VPg as a novel eIF4E-binding protein, which inhibits host protein synthesis at a very early stage of the initiation complex formation through the inhibition of cap attachment to the initiation factor eIF4E.  相似文献

In this study we quantified variability in foliage herbivory and pre-dispersal seed predation and its effects on plant performance and demography in populations of a rare and protected perennial herb, Paeonia officinalis. An individual-based survey was performed during four years in four populations, which contained plants in both open habitat and woodland. We detected marked spatial and temporal variation among and within populations in foliage herbivory (by insects) and pre-dispersal seed predation (by insects, rodents and Roe Deer). Foliage herbivory decreased with plant demographic stages in open habitats, from seedlings to reproductive individuals, but no significant trend was detected in woodland habitats. This may be due to different demographic origin of larger vegetative plants in this habitat. Depending on demographic stage, herbivory was higher in open habitats or not significantly different between habitats. This suggests differences in herbivore abundance in different habitats within sites. Pre-dispersal seed predation remained weak and did not depend on habitat. We did not detect any consequence of foliage herbivory on seedling mortality and individual growth in our study. Our results illustrate the need to investigate plant-herbivore interactions over several years in distinct populations in order to more accurately evaluate herbivore impact on plant population dynamics.  相似文献

NECDIN belongs to the type II Melanoma Associated Antigen Gene Expression gene family and is located in the Prader-Willi Syndrome (PWS) critical region. Necdin-deficient mice develop symptoms of PWS, including a sensory and motor deficit. However, the mechanisms underlying the motor deficit remain elusive. Here, we show that the genetic ablation of Necdin, whose expression is restricted to post-mitotic neurons in the spinal cord during development, leads to a loss of 31% of specified motoneurons. The increased neuronal loss occurs during the period of naturally-occurring cell death and is not confined to specific pools of motoneurons. To better understand the role of Necdin during the period of programmed cell death of motoneurons we used embryonic spinal cord explants and primary motoneuron cultures from Necdin-deficient mice. Interestingly, while Necdin-deficient motoneurons present the same survival response to neurotrophic factors, we demonstrate that deletion of Necdin leads to an increased susceptibility of motoneurons to neurotrophic factor deprivation. We show that by neutralizing TNFα this increased susceptibility of Necdin-deficient motoneurons to trophic factor deprivation can be reduced to the normal level. We propose that Necdin is implicated through the TNF-receptor 1 pathway in the developmental death of motoneurons.  相似文献

In observations by confocal or conventional fluorescence microscopy, important factors should be considered in order to obtain accurate images. One of them, such as the fluorescence bleaching from highest intensity to lowest signal of fluorescence is a common problem with several DNA fluorochromes and especially for DAPI stain. The fluorescence of DAPI fades rapidly when it is exposed to UV light, under optimal conditions of observation. Although the fading process can be retarded using a mounting medium with antifading reagents, the photochemical process underlying the fluorescence decay has not yet been fully explained. In addition, no relationship between fluorescence fading and nuclear DNA content has been tested. In order to test this relationship, we measured by means of image analysis the DAPI-fluorescence intensity in several cellular types (spermatozoa, erythrocytes and haemocytes) during their fluorescence bleaching. An algorithm specifically built in MATLAB software was used for this approach. The correlation coefficient between nuclear DNA content and DAPI-fluorescence fading was found equal to 99%. This study demonstrates the feasibility to measure nuclear DNA content by fluorescence fading quantification, as an alternative method concurrently with image analysis procedures.  相似文献

We studied the soil seed bank in a possible scenario of fire regime shift and asked: (1) Does high fire frequency impact the density of seeds stored, species richness and evenness? (2) Overall, does high fire frequency produce changes in the presence–absence and abundance of species? The study was implemented in a Mediterranean Basin ecosystem in plots with increasing fire frequency (unburned, burned once and burned twice in the last 66 years). The number of seeds increased with fire frequency for all life forms (shrub, scrub, perennial forb, annual forb and perennial graminoid). Species richness of annual forbs also increased. Evenness of shrubs diminished because the number of seeds in all the species decreased, except C. albidus, which increased. Overall, differences in the abundance of species were found, mainly by depleting shrubs and increasing forbs. There were no differences in the presence–absence data. In conclusion, high fire frequencies act as a filtering factor for species of a larger size and advanced maturity age. In contrast, life forms of small size and rapid onset of reproductive maturity can be enhanced. This community conversion from woody to herbaceous soil seed banks is fundamental to identify vegetation changes in future regimes of high fire frequency.  相似文献
