Therapeutic proteins are utilized in a variety of clinical applications, but side effects and rapid in vivo clearance still present hurdles. An approach that addresses both drawbacks is protein encapsulation within in a polymeric nanoparticle, which is effective but introduces the additional challenge of destabilizing the nanoparticle shell in clinically relevant locations. This study examined the effects of crosslinking self-assembled poly(l -lysine)-grafted-poly(ethylene glycol) nanoparticles with redox-responsive 3,3′-dithiobis(sulfosuccinimidyl propionate) (DTSSP) to achieve nanoparticle destabilization in a reductive environment. The polymer-protein nanoparticles (DTSSP NPs) were formed through electrostatic self-assembly and crosslinked with DTSSP, which contains a glutathione-reducible disulfide. As glutathione is upregulated in various cancers, DTSSP NPs could display destabilization within cancer cells. A library of DTSSP NPs was formed with varying copolymer to protein (C:P) and crosslinker to protein (X:P) mass ratios and characterized by size and encapsulation efficiency. DTSSP NPs with a 7:1 C:P ratio and 2:1 X:P ratio were further characterized by stability in the presence proteases and reducing agents. DTSSP NPs fully encapsulated the model protein and displayed 81% protein release when incubated with 5 mM dithiothreitol for 12 hr. This study contributes to understanding stimulus-responsive crosslinking of polymeric nanoparticles and could be foundational to clinical administration of therapeutic proteins.  相似文献

ABSTRACT Telemetry data have been widely used to quantify wildlife habitat relationships despite the fact that these data are inherently imprecise. All telemetry data have positional error, and failure to account for that error can lead to incorrect predictions of wildlife resource use. Several techniques have been used to account for positional error in wildlife studies. These techniques have been described in the literature, but their ability to accurately characterize wildlife resource use has never been tested. We evaluated the performance of techniques commonly used for incorporating telemetry error into studies of wildlife resource use. Our evaluation was based on imprecise telemetry data (mean telemetry error = 174 m, SD = 130 m) typical of field-based studies. We tested 5 techniques in 10 virtual environments and in one real-world environment for categorical (i.e., habitat types) and continuous (i.e., distances or elevations) rasters. Technique accuracy varied by patch size for the categorical rasters, with higher accuracy as patch size increased. At the smallest patch size (1 ha), the technique that ignores error performed best on categorical data (0.31 and 0.30 accuracy for virtual and real data, respectively); however, as patch size increased the bivariate-weighted technique performed better (0.56 accuracy at patch sizes >31 ha) and achieved complete accuracy (i.e., 1.00 accuracy) at smaller patch sizes (472 ha and 1,522 ha for virtual and real data, respectively) than any other technique. We quantified the accuracy of the continuous covariates using the mean absolute difference (MAD) in covariate value between true and estimated locations. We found that average MAD varied between 104 m (ignore telemetry error) and 140 m (rescale the covariate data) for our continuous covariate surfaces across virtual and real data sets. Techniques that rescale continuous covariate data or use a zonal mean on values within a telemetry error polygon were significantly less accurate than other techniques. Although the technique that ignored telemetry error performed best on categorical rasters with smaller average patch sizes (i.e., ≤31 ha) and on continuous rasters in our study, accuracy was so low that the utility of using point-based approaches for quantifying resource use is questionable when telemetry data are imprecise, particularly for small-patch habitat relationships.  相似文献

Species adapt to the selective pressures of novel environments. Here, Gleditsch and Sperry tested whether four nonnative frugivorous bird species on the Hawaiian island of O'ahu diverged morphologically from their ancestral populations. They found that all four species significantly diverged from populations in their native ranges, with a general trend of smaller body size and larger bills. These differences were likely due to a combination of adaptive and nonadaptive processes.  相似文献

A series of 4-day manipulations of zooplankton biomass and nutrientavailability was performed in enclosures in three lakes to determinespecies-specific algal responses to herbivory and nutrient enrichment.Algal performance in enclosures was compared to the relationshipsbetween weekly algal growth rates and the zooplankton in situ.When in situ growth rates were significant functions of zooplanktonbiomass, the responses were generally consistent with responsesin the enclosure experiments. The importance of both nutrientsand zooplankton in mediating algal growth was demonstrated bynumerous observations: strong algal community response to enrichment,unimodal or positive responses of certain algal taxa to zooplanktonbiomass, differences in degree of nutrient limitation amongthe algal response types, lack of nutrient limitation of non-grazedalgal taxa and a preponderance of taxa with no net responseto increasing zooplankton biomass. Variation in the zooplanktoncommunity may be the largest source of variability in nutrientsupply rate during summer in stratified lakes, and causes substationalvariability in the algae. Electron paramagnetic resonance (EPR) studies of the nitrosyl adduct of ferrous lactoperoxidase (LPO) confirm that the fifth axial ligand in LPO is bound to the iron via a nitrogen atom. Complete reduction of the ferric LPO sample is required in order to observe the nine-line hyperfine splitting in the ferrous LPO/NO EPR spectrum. The ferrous LPO/NO complex does not exhibit a pH or buffer system dependence when examined by EPR. Interconversion of the ferrous LPO/NO complex and the ferric LPO/NO2- complex is achieved by addition of the appropriate oxidizing or reducing agent. Characterization of the low-spin LPO/NO2- complex by EPR and visible spectroscopy is reported. The pH dependence of the EPR spectra of ferric LPO and ferric LPO/CN- suggests that a high-spin anisotropic LPO complex is formed at high pH and an acid-alkaline transition of the protein conformation near the heme site does occur in LPO/CN-. The effect of tris(hydroxymethyl)aminomethane buffer on the LPO EPR spectrum is also examined.  相似文献

Identification of gene function has often relied on isolation of mutant cells in which expression of the gene was inactivated. Gene targeting by homologous recombination in tissue culture now may provide a technology to rapidly and directly produce such mutant mammalian cells. We demonstrate that selection of embryonic stem and pre-B cell lines for expression of a promoterless construct containing murine N-myc genomic sequences fused to a gene encoding neomycin resistance allows highly efficient recovery of variants in which the endogenous N-myc gene is disrupted. The high frequency of N-myc gene disruption by this method should permit targeted disruption of both allelic N-myc copies in various cell lines to study N-myc function.  相似文献

Summary In this preliminary study, non-invasive infrared thermography has been used to visualize individual sweat pores and whole body skin temperature patterns in subjects with X-linked hypohidrotic ectodermal dysplasia (XHED) and normal controls. The findings in eight obligate heterozygotes and four affected males were compared to six normal female controls and to six non-manifesting females at risk for carrier status. Sweat secretion from individual pores in circumscribed areas was imaged using a high spatial resolution SPRITE infrared detector system working in the 8–14 m band. In seven out of eight obligate heterozygotes, skin areas devoid of active sweat glands were found on the face, the hands or the trunk. Tear front movement over the cornea was also visualized and abnormal patterns were identified in obligate heterozygotes. Whole body skin temperature patterns, obtained with an Agema 780 Medical Thermovision system, identified abnormal skin temperature distributions, including characteristic aberrant cas-cade back patterns, in obligate carriers. Two out of six at risk females had skin temperature patterns comparable with obligate heterozygotes and we have tentatively concluded that they are carriers. Thermal imaging may be used for the examination of at risk non-manifesting females in families with a single affected male. The results of this study suggest that the random X-inactivation in females with XHED, as well as producing relatively large skin areas with sweat pore aplasia, is also associated with abnormal temperature patterns that are consistent with altered peripheral vascular perfusion.  相似文献

Clenbuterol, as other sympathomimetic drugs, relaxes the myometrium, thus causing a short-term inhibition of labor and the delay of parturition. This study has examined the influence of clenbuterol on the release of prostaglandin F2 alpha (PGF2 alpha) induced by oxytocin alone or with estradiol-17 beta. Five bilaterally ovariectomized heifers, primed with progesterone for 14 days, were used in two experiments. In the first they received two i.v. injections of oxytocin 6h apart, with and without an i.v. injection of clenbuterol before the second oxytocin injection; the second experiment was similar to the first except that the animals were given estradiol-17 beta 30 min after the first oxytocin injection. Frequent blood samples were taken for the measurement of 13,14-dihydro-15-keto-PGF2 alpha by radioimmunoassay. The data show that clenbuterol does not influence PGF2 alpha release in response to oxytocin alone or with estradiol-17 beta, and it does not inhibit the basal release of PGF2 alpha. This suggests that clenbuterol does not act on the endometrium to alter the secretion of PGF2 alpha in the non-pregnant cow.  相似文献

Mature Holstein heifers were given either a priming dose of follicle-stimulating hormone-pituitary (FSH-P, 10 mg) or saline on Day 2 of the estrous cycle, or no pretreatment. All animals were subsequently given a decreasing dose superovulatory treatment of FSH-P beginning between Days 8 and 14, coupled with an injection of prostaglandin F2a to induce luteolysis. Pretreatment with FSH-P had no effect on the total superovulatory response or on the number of transferable embryos collected at Day 7 of gestation. Comparison of the results of our study with previous reports in the literature may suggest that FSH-priming early in the cycle may be advantageous in promoting superovulation only when the superovulatory response of the population of animals is otherwise weak.  相似文献
