We collected surface samples in Franklin Bay (Western Arctic) from ice-covered to ice-free conditions, to determine seasonal changes in the identity and in situ activity of the prokaryotic assemblages. Catalysed reported fluorescence in situ hybridization was used to quantify the abundance of different groups, and combined with microautoradiography to determine the fraction of active cells taking up three substrates: glucose, amino acids and ATP. In surface waters, Archaea accounted for 16% of the total cell count in winter, but decreased to almost undetectable levels in summer, when Bacteria made up 97% of the total cell count. Alphaproteobacteria were the most abundant group followed by Bacteroidetes (average of 34% and 14% of total cell counts respectively). Some bacterial groups appearing in low abundances (< 10% of total cell counts), such as Betaproteobacteria , Roseobacter and Gammaproteobacteria , showed a high percentage of active cells. By contrast, more abundant groups, such as SAR11 or Bacteroidetes , had a lower percentage of active cells in the uptake of the substrates tested. Archaea showed low heterotrophic activity throughout the year. In comparison with temperate oceans, the percentage of active Bacteria in the uptake of the substrates was relatively high, even during the winter season.  相似文献

We present an agent-based model inspired by the Evolutionary Minority Game (EMG), albeit strongly adapted, to the case of competition for limited resources in ecology. The agents in this game become able, after some time, to predict the a priori best option as a result of an evolution-driven learning process. We show that a self-segregated social structure can emerge from this process, i.e., extreme learning strategies are always favoured while intermediate learning strategies tend to die out. This result may contribute to understanding some levels of organization and cooperative behaviour in ecological and social systems. We use the ideas and results reported here to discuss an issue of current interest in ecology: the mistimings in egg laying observed for some species of bird as a consequence of their slower rate of adaptation to climate change in comparison with that shown by their prey. Our model supports the hypothesis that habitat-specific constraints could explain why different populations are adapting differently to this situation, in agreement with recent experiments.  相似文献

We studied influence of the Argentine ant's Linepithema humile occurrence on seed dispersal of Rhamnus alaternus (Rhamnaceae). Rhamnus alaternus is a fleshy fruit plant dispersed primarily by birds; the seeds have an elaiosome attractive to ants. The observations were made in two study plots of Mediterranean cork-oak forests (one invaded and the other not invaded by L. humile ) over two years. For R. alaternus , presence of L. humile was associated with of the following: reduction of seed transport to ant nests (80.2% of seed offered were removed in non-invaded vs 7.1% in invaded area), a shorter seed dispersal distance (mean=90.2 cm in non-invaded vs 1.1 cm in invaded area), increase in seed retention time on the soil surface (median time in non-invaded was 50 vs 209.2 min in invaded area) and increased vulnerability to predation. In addition there was lower probability of seedling emergence, due to little elaiosome removal from the seeds (82.0% of emergence for seeds without elaiosome and 57.3% for seeds with elaiosome). These results, similar to findings obtained in South African fynbos, confirm that the Argentine ant invasion can strongly affect ecosystem processes in the Mediterranean biome.  相似文献

A flow cytometric protocol to detect and enumerate heterotrophic nanoflagellates (HNF) in enriched waters is reported. At present, the cytometric protocols that allow accurate quantification of bacterioplankton cannot be used to quantify protozoa for the following reasons: i) the background produced by the bacterial acquisitions does not allow the discrimination of protozoa at low abundance, ii) since the final protozoan fluorescence is much higher than the bacterioplankton fluorescence (more than 35 fold) the protozoa acquisitions lie outside the range. With an increase in the fluorescence threshold and a reduction of the fluorescence detector voltage, low fluorescence particles (bacteria) are beneath the detection limits and only higher fluorescence particles (most of them heterotrophic nanoflagellates) are detected. The main limitation for the application of the cytometric protocol developed is that a ratio of bacteria/HNF below 1000 is needed. At higher ratios, the background of larger cells of bacterioplankton makes it difficult to discriminate protozoa. The proposed protocol has been validated by epifluorescence microscopy analyzing both a mixed community and two single species of HFN: Rhynchomonas nasuta and Jakoba libera. Taking into account the required bacteria/HNF ratio cited above, the results provide evidence that the flow cytometric protocol reported here is valid for counting mixed communities of HNF in enriched seawater and in experimental micro or mesocosms. In the case of single species of HNF previous knowledge of the biological characteristics of the protist and how they can affect the effectiveness of the flow cytometric count is necessary.  相似文献

Diketide N-acetylcysteamine (diketide NAC) thioester precursors were fed to 6-Deoxyerythronolide B synthase (DEBS) ketosynthase-1 inactivated (KS1 degree) Saccharopolyspora erythraea strains to produce 13-substituted erythromycin analogs. This direct feeding process potentially represents a simplified production process over the current analog production system. Titers of these analogs were observed to increase linearly with the diketide concentration up to a precursor-specific saturation level. However, the rate of product formation was lower and the rate of diketide consumption higher with S. erythraea than was previously observed with a recombinant strain of Streptomyces coelicolor. Several strategies were pursued to address the issue of these high diketide consumption rates: (1) elucidation of the locale of diketide degradation, (2) addition of beta-oxidation inhibitors to the cultures, and (3) addition of a sacrificial diketide enantiomer to occupy putative degradative enzymes. Additionally, repeated addition of diketide to an S. erythraea KS1 degrees culture indicated that the titer of these erythromycin analogs is also currently limited by a shorter production period than observed during erythromycin synthesis by the parent strain. These results indicate potential avenues for expanding the use of this precursor-directed system from the generation of limited quantities of erythromycin analogs to a large-scale production system for these compounds.  相似文献

The mountains of the southeast Iberian Peninsula harbor several narrowly endemic species belonging to Narcissus section Pseudonarcissi (N. alcaracensis, N. bugei, N. enemeritoi, N. longispathus, N. nevadensis, N. segurensis, and N. yepesii) that are protected by regional Spanish laws. Most of these trumpet daffodils show a very similar overall morphology, and the correct identification of species is difficult unless the geographical origins of the accessions are known. ISSR (Inter-Simple Sequence Repeat) and nuclear ribosomal ITS sequences were used to assess patterns of genetic discontinuities present among the members of this section from southeast Spain. Species-specific ISSR bands were found in N. bugei, N. longispathus and N. nevadensis, but were absent in the other species. The multivariate ordination of the 288 ISSR-genotyped accessions showed that only N. longispathus and N. bugei were clearly differentiated, while the other species formed an unresolved group. ITS sequences of N. bugei were highly divergent from those retrieved from the remaining species, which were placed in a single largely unresolved clade in phylogenetic analysis. In conclusion, ISSR and ITS markers support the taxonomic distinction of N. bugei and N. longispathus. However, N. alcaracensis, N. segurensis, N. yepesii, N. enemeritoi, and N. nevadensis could be merged within a single species (N. nevadensis for nomenclatural priority).  相似文献

Passage through the digestive tract exposes Salmonella enterica to high concentrations of bile salts, powerful detergents that disrupt biological membranes. Mutations in the wecD or wecA gene, both of which are involved in the synthesis of enterobacterial common antigen (ECA), render S. enterica serovar Typhimurium sensitive to the bile salt deoxycholate. Competitive infectivity analysis of wecD and wecA mutants in the mouse model indicates that ECA is an important virulence factor for oral infection. In contrast, lack of ECA causes only a slight decrease in Salmonella virulence during intraperitoneal infection. A tentative interpretation is that ECA may contribute to Salmonella virulence by protecting the pathogen from bile salts.  相似文献

The interaction between pathogens and their multicellular hosts is initiated by activation of pathogen recognition receptors (PRRs). These receptors, that include most notably members of the toll-like receptor (TLR) family, recognize specific pathogen-associated molecular patterns (PAMPs). TLR4 is a central part of the receptor complex that is involved in the activation of the immune system by lipopolysaccharide (LPS) through the specific recognition of its endotoxic moiety (Lipid A). This is a critical event that is essential for the immune response to Gram-negative bacteria as well as the etiology of endotoxic shock. Interestingly, compared to mammals, fish are resistant to endotoxic shock. This in vivo resistance concurs with in vitro studies demonstrating significantly lowered sensitivity of fish leukocytes to LPS activation. Further, our in vitro analyses demonstrate that in trout mononuclear phagocytes, LPS fails to induce antiviral genes, an event that occurs downstream of TLR4 and is required for the development of endotoxic shock. Finally, an in silico approach that includes mining of different piscine genomic and EST databases, reveals the presence in fish of all of the major TLR signaling elements except for the molecules specifically involved in TLR4-mediated endotoxin recognition and signaling in mammals. Collectively, our analysis questions the existence of TLR4-mediated cellular responses to LPS in fish. We further speculate that other receptors, in particular beta-2 integrins, may play a primary role in the activation of piscine leukocytes by LPS.  相似文献
