The recent discovery of a carotenoid light-harvesting antenna in xanthorhodopsin, a retinal-based proton pump in Salinibacter ruber, made use of photoinhibition of respiration in whole cells to obtain action spectra [Balashov et al. Science 309, (2005) 2061-2064]. Here we provide further details of this phenomenon, and compare action spectra in three different systems where carotenoids have different functions or efficiencies of light-harvesting. The kinetics of light-induced inhibition of respiration in Salinibacter ruber was determined with single short flashes, and the photochemical cross section of the photoreaction was estimated. These measurements confirm that the xanthorhodopsin complex includes no more than a few, and most likely only one, carotenoid molecule, which is far less than the core complex antenna of photosynthetic bacteria. Although the total cross-section of light absorption in the purple bacterium Rhodospirillum rubrum greatly exceeds that in Salinibacter, the cross-sections are roughly equivalent in the shared wavelength range. We show further that despite interaction of bacterioruberin with archaerhodopsin, another retinal-based proton pump, there is no significant energy transfer from this carotenoid. This emphasizes the uniqueness of the salinixanthin-retinal interaction in xanthorhodopsin, and indicates that bacterioruberin in Halorubrum species has a structural or photoprotective rather than energetic role.  相似文献

The mature ovary of Gastrotheca riobambae presents large oocytes (3 mm in diameter) of pale yellow color. After ovulation and the beginning of embryonic incubation, the empty postovulatory follicles can be recognized in the ovary for about 30 days. The granulosa of these follicles never fills the follicular lumen and this cavity becomes filled with fluid during the first five days of incubation. Later, at 18 days of incubation, the lumen is mostly empty and contains few cells of the granulosa. Shrinkage results in the disappearance of the follicular cavity by approximately the thirtieth day of incubation. The thecae are thick and become separated by a space. This space is filled progressively with cells, blood capillaries and fluid. After the thirtieth to fortieth day of incubation, these follicles become regressive and disappear. The postovulatory follicles of Gastrotheca may correspond to corpora lutea. The evidence suggests that pouch vascularization and the formation of embryonic chambers of pouch tissue may be under ovarian control. In addition, the process of vitellogenesis is influenced by incubation, as most growth of the ovarian oocyte occurs after birth of the tadpoles. Follicular atresia is common and is similar to that of other frogs.  相似文献

Tissue banks around the world store human cartilage obtained from cadaveric donors for use in diverse reconstructive surgical procedures. To ensure this tissue is sterile at the time of distribution, tissues may be sterilized by ionizing radiation. In this work, we evaluate the physical changes in deep frozen costal cartilage (?70 °C) or costal cartilage preserved in high concentrations of glycerol (>98 %) followed by a terminal sterilization process using ionizing radiation, at 3 different doses (15, 25 and 50 kGy). Tension and compression tests were carried out to determine the mechanical changes related both to the different preservation methods and irradiation doses. For both methods of preservation, tension strength was increased by about 24 %, when cartilage tissue was irradiated with 15 kGy. Deep frozen samples, when irradiated with 25 or 50 kGy, had a decrease in their mechanical performance, albeit to a lesser extent than when tissues were preserved in high concentration of glycerol and equally irradiated. In conclusion, processing in high concentration of glycerol did not increase tissue protection against radiation damage; while cartilage preserved in high concentrations of glycerol withstands radiation up to 25 kGy, deep frozen human costal cartilage may be sterilized with a doses up to 50 kGy without significant mechanical impact.  相似文献

Seed dispersal is one of the most important steps in the plant life cycle. However, there is, generally, a lack of fieldworks focused on wind dispersal and especially on semachorous dispersal (seeds spread when the fruits are shaken by wind and other vectors, such as animals), including boleochorous dispersal. Therefore, we aimed to determine how different types of wind and animals affected seed dispersal under natural conditions in the widespread species Scrophularia canina. We evaluated the effects of wind gusts (simulating them using a leaf blower) and wild animals (using differently sized dogs) on seed dispersal in a population located in south-western Europe. We found that S. canina is a semachorous species, and its spatial seed dispersal was affected by wind gust speed and direction, plant structure and vector type. The results also revealed the presence of xerochasy, individual anisotropy with strong winds, and primary short-distance dispersal associated with successional processes independent of the vector. Additionally, there was a masking effect of plant structure on the seed shadow outline. It is essential to conduct fieldworks to reveal what actually happens in nature, taking into account the characteristics determining seed dispersal. In addition, in these works it is important to find out what factors affect seed distributions of anemochorous and semachorous species.  相似文献

Summary The amino acid contents of edible insects from different provinces of Mexico and reference proteins were analysed by reversed-phase high-performance liquid chromatography and ion exchange chromatography.
