The effects of the expression of the Na+/H+ exchanger regulatory factor-1 (NHERF1) on the distribution, dynamics, and signaling properties of the PTH type 1 receptor (PTH1R) were studied in rat osteosarcoma cells ROS 17/2.8. NHERF1 had a dramatic effect on the subcellular distribution of PTH1R, promoting a substantial relocation of the receptor to regions of the plasma membrane located in very close proximity to cytoskeletal fibers. Direct interactions of NHERF1 with the PTH1R and the cytoskeleton were required for these effects, because they were abolished by 1) PTH1R mutations that impair NHERF1 binding, and 2) NHERF1 mutations that impair binding to the PTH1R or the cytoskeleton. NHERF1 reduced significantly the diffusion of the PTH1R by a mechanism that was also dependent on a direct association of NHERF1 with the PTH1R and the cytoskeleton. NHERF1 increased ligand-dependent production of cAMP and induced ligand-dependent rises in intracellular calcium. These effects on calcium were due to increased calcium uptake, as they were blocked by calcium channel inhibitors and by the addition of EGTA to the medium. These calcium effects were abolished by protein kinase A inhibition but phospholipase C inhibition was without effect. Based on these analyses, we propose that, in ROS cells, the presence of NHERF1 induces PTH-dependent calcium signaling by a cAMP-mediated mechanism that involves local protein kinase A-dependent activation of calcium channels. 相似文献

Cellular therapy is a potential approach to improve the regenerative capacity of damaged or diseased skeletal muscle. However, its clinical use has often been limited by impaired donor cell survival, proliferation and differentiation following transplantation. Additionally, functional improvements after transplantation are all-too-often negligible. Because the host microenvironment plays an important role in the fate of transplanted cells, methods to modulate the microenvironment and guide donor cell behavior are warranted. The purpose of this study was to investigate whether the use of neuromuscular electrical stimulation (NMES) for 1 or 4 weeks following muscle-derived stem cell (MDSC) transplantation into dystrophic skeletal muscle can modulate the fate of donor cells and enhance their contribution to muscle regeneration and functional improvements. Animals submitted to 4 weeks of NMES after transplantation demonstrated a 2-fold increase in the number of dystrophin+ myofibers as compared to control transplanted muscles. These findings were concomitant with an increased vascularity in the MDSC+NMES group when compared to non-stimulated counterparts. Additionally, animals subjected to NMES (with or without MDSC transplantation) presented an increased maximal specific tetanic force when compared to controls. Although cell transplantation and/or the use of NMES resulted in no changes in fatigue resistance, the combination of both MDSC transplantation and NMES resulted in a faster recovery from fatigue, when compared to non-injected and non-stimulated counterparts. We conclude that NMES is a viable method to improve MDSC engraftment, enhance dystrophic muscle strength, and, in combination with MDSC transplantation, improve recovery from fatigue. These findings suggest that NMES may be a clinically-relevant adjunct approach for cell transplantation into skeletal muscle. 相似文献

Alphaviruses are small, spherical, enveloped, positive-sense, single-stranded, RNA viruses responsible for considerable human and animal disease. Using microinjection of preassembled cores as a tool, a system has been established to study the assembly and budding process of Sindbis virus, the type member of the alphaviruses. We demonstrate the release of infectious virus-like particles from cells expressing Sindbis virus envelope glycoproteins following microinjection of Sindbis virus nucleocapsids purified from the cytoplasm of infected cells. Furthermore, it is shown that nucleocapsids assembled in vitro mimic those isolated in the cytoplasm of infected cells with respect to their ability to be incorporated into enveloped virions following microinjection. This system allows for the study of the alphavirus budding process independent of an authentic infection and provides a platform to study viral and host requirements for budding. 相似文献

About 150 species belonging to 14 families are mentioned in the scattered literature dealing with the land molluscs of Chile. However, the records of the Streptaxidae and Haplotrematidae are dubious and all those for the Limacidae, Zonitidae and Helicidae correspond to introduced species. The remaining 9 families are represented by endemic genera and subgenera present either in the oceanic islands (Fernandezia, Tornatellina, Ambrosiella) or in continental Chile (Bostryx (Ataxus), Chiliborus). A few are limited in their distribution to the southern part of South America (Austrodiscus, Stephadiscus, Macrocyclis) or are found both in the islands and along the continent (Amphiodoxa, Stephanoda). The majority of the other genera have a wider distribution in the Indo‐Pacific (Punctum, Ptychodon, Champa), in South America, the Americas in general and/or in the other parts of the world. The distribution of molluscan genera and species in continental Chile is discussed with regard to the biogeographic units proposed by various authors for plants and animals. Their probable relationships with the faunas of neighbouring countries and other biogeographic regions is briefly outlined. A taxonomic list of the species indicating the original reference, general distribution and the uncertain position of some of them within the different genera is given. Spatial or temporal differences in environmental variables, such as temperature, are ubiquitous in nature and impose stress
on organisms. This is especially true for organisms that are isothermal with the environment, such as insects. Understanding
the means by which insects respond to temperature and how they will react to novel changes in environmental temperature is
important for understanding the adaptive capacity of populations and to predict future trajectories of evolutionary change.
The organismal response to heat has been identified as an important environmental variable for insects that can dramatically
influence life history characters and geographic range. In the current study we surveyed the amount of variation in heat tolerance
among Drosophila melanogaster populations collected at diverse sites along a latitudinal gradient in Argentina (24°–38°S). This is the first study to quantify
heat tolerance in South American populations and our work demonstrates that most of the populations surveyed have abundant
within-population phenotypic variation, while still exhibiting significant variation among populations. The one exception
was the most heat tolerant population that comes from a climate exhibiting the warmest annual mean temperature. All together
our results suggest there is abundant genetic variation for heat-tolerance phenotypes within and among natural populations
of Drosophila and this variation has likely been shaped by environmental temperature. 相似文献

Background
It has traditionally been thought that the pathological accumulation of tau in Alzheimer's disease and other tauopathies facilitates neurodegeneration, which in turn leads to cognitive impairment. However, recent evidence suggests that tau tangles are not the entity responsible for memory loss, rather it is an intermediate tau species that disrupts neuronal function. Thus, efforts to discover therapeutics for tauopathies emphasize soluble tau reductions as well as neuroprotection.Results
Here, we found that neuroprotection alone caused by methylene blue (MB), the parent compound of the anti-tau phenothiaziazine drug, Rember?, was insufficient to rescue cognition in a mouse model of the human tauopathy, progressive supranuclear palsy (PSP) and fronto-temporal dementia with parkinsonism linked to chromosome 17 (FTDP17): Only when levels of soluble tau protein were concomitantly reduced by a very high concentration of MB, was cognitive improvement observed. Thus, neurodegeneration can be decoupled from tau accumulation, but phenotypic improvement is only possible when soluble tau levels are also reduced.Conclusions
Neuroprotection alone is not sufficient to rescue tau-induced memory loss in a transgenic mouse model. Development of neuroprotective agents is an area of intense investigation in the tauopathy drug discovery field. This may ultimately be an unsuccessful approach if soluble toxic tau intermediates are not also reduced. Thus, MB and related compounds, despite their pleiotropic nature, may be the proverbial "magic bullet" because they not only are neuroprotective, but are also able to facilitate soluble tau clearance. Moreover, this shows that neuroprotection is possible without reducing tau levels. This indicates that there is a definitive molecular link between tau and cell death cascades that can be disrupted.140.
d-Penicillamine (DPA) is effective in the treatment of Wilson’s disease, whereas zinc salts are also used as a therapy for
this disorder of copper transport. Recently, it has been shown that the copper chelators 1,4,7,11-tetraazaundecane tetrahydrochloride
(TAUD) and tetraethylenepentamine pentahydrochloride (TETREN) could be useful for copper mobilization in rats. Because these
agents could be potential clinical alternatives to DPA for patients with Wilson’s disease who are intolerant to this drug,
we examined whether oral administration of TAUD and TETREN could be effective in mobilizing copper in experimental copper-overloaded
rats. The efficacy of a combined administration of zinc and DPA, TAUD, or TETREN was also assessed. Rats were copper loaded
with 0.125% copper acetate in water for 12 wk. After this period, DPA, TAUD, and TETREN were administered by gavage at 0.67
mmol/kg/d for 5 d, and zinc was given at 2.5 mg Zn/kg/d. Twelve weeks of copper loading resulted in a 32-fold increase in
total hepatic copper. TETREN was the most effective chelator in increasing the urinary excretion of copper. However, it did
not reduce significantly the hepatic copper levels. In turn, combined administration of zinc and chelating agents significantly
reduced the amount of copper found in the feces. Although TAUD and TETREN showed a similar or higher efficacy to DPA in mobilizing
copper, concurrent treatment of chelating agents and zinc salts should be discarded according to the current results. 相似文献
