Stimulation of native microbial populations in soil by the addition of small amounts of secondary carbon sources (cosubstrates) and its effect on the degradation and theoretical mineralization of DDT [l,l,l-trichloro-2,2-bis(p-chlorophenyl)ethane] and its main metabolites, DDD and DDE, were evaluated. Microbial activity in soil polluted with DDT, DDE and DDD was increased by the presence of phenol, hexane and toluene as cosubstrates. The consumption of DDT was increased from 23 % in a control (without cosubstrate) to 67, 59 and 56 % in the presence of phenol, hexane and toluene, respectively. DDE was completely removed in all cases, and DDD removal was enhanced from 67 % in the control to ~86 % with all substrates tested, except for acetic acid and glucose substrates. In the latter cases, DDD removal was either inhibited or unchanged from the control. The optimal amount of added cosubstrate was observed to be between 0.64 and 2.6 mg C $ {\text{g}}^{ - 1}_{\text{dry soil}} $ . The CO2 produced was higher than the theoretical amount for complete cosubstrate mineralization indicating possible mineralization of DDT and its metabolites. Bacterial communities were evaluated by denaturing gradient gel electrophoresis, which indicated that native soil and the untreated control presented a low bacterial diversity. The detected bacteria were related to soil microorganisms and microorganisms with known biodegradative potential. In the presence of toluene a bacterium related to Azoarcus, a genus that includes species capable of growing at the expense of aromatic compounds such as toluene and halobenzoates under denitrifying conditions, was detected.  相似文献

Tumor necrosis factor-like weak inducer of apoptosis (TWEAK) regulates apoptosis, proliferation and inflammation in renal epithelial cells and plays a role in acute kidney injury. However, there is little information on the chronic effects of TWEAK. We hypothesized that TWEAK may influence renal fibrosis and regulate kidney fibroblast biology, in part, through Ras pathway.We studied a chronic model of experimental unilateral ureteral obstruction in wild type and TWEAK deficient mice, and a murine model of systemic TWEAK overexpression. TWEAK actions were also explored in cultured renal and embryonic fibroblasts.TWEAK and TWEAK receptor expression was increased in the obstructed kidneys. The absence of TWEAK decreased early kidney tubular damage, inflammatory infiltrates and myofibroblast number. TWEAK deficient mice had decreased renal fibrosis 21 days after obstruction, as assessed by extracellular matrix staining. In mice without prior underlying kidney disease, systemic overexpression of TWEAK induced kidney inflammation and fibrosis. In cultured fibroblasts, TWEAK induced proliferation through activation of the Ras/ERK pathway. TWEAK also activated nuclear factor κB (NFκB)-dependent inflammatory chemokine production in murine renal fibroblasts.In conclusion, lack of TWEAK reduces renal fibrosis in a model of persistent kidney insult and overexpression of TWEAK led to renal fibrosis. TWEAK actions on renal fibroblasts may contribute to the in vivo observations, as TWEAK promotes inflammatory activity and proliferation in fibroblast cultures.  相似文献

The aim of this study was to evaluate the effect of different concentrations of soybean lecithin (SL) in extenders for sperm goat cryopreservation. Sexually mature male Saanen goats (n = 4) were used, and the ejaculates were obtained using an artificial vagina method. The semen samples were pooled and diluted in a skim milk-based extender (control group; CG) or Tris extender supplemented with SL at different concentrations (G1 = 0.04%, SL G2 = 0.08% SL and G3 = 0.16%) for a final concentration of 240 × 106 spermatozoa/mL. The semen samples were packed in straws (0.25 mL), frozen using an automated system and stored in liquid nitrogen (?196 °C). After thawing (37 °C/30 s), the samples were evaluated for sperm quality parameters, including sperm motility, membrane integrity, acrosome integrity and mitochondrial activity. No significant difference was observed among the experimental and control groups for all of the parameters (P > 0.05). However, even though the control group presented a significantly lower mitochondrial membrane potential compared to fresh semen (P < 0.05), the same did not occur for the extender supplemented with soybean lecithin, that is, it did not differ from fresh sperm (P > 0.05). The extender containing soybean lecithin at different concentrations preserved the sperm quality parameters in a manner similar to the conventional skim milk-based extender. Thus, it is concluded that an extender containing soybean lecithin as the lipoprotein source can be used for freezing goat semen.  相似文献

This work analyzed the diurnal variation of general activity and plasmatic levels of melatonin and corticosterone in pigeons submitted to a 12:00:12:00 h light-dark cycle (lights on at 6:00 a.m.) or to constant light. In both conditions pigeons were observed in 5-min sessions at times 03:00, 06:00, 09:00, 12:00, 15:00, 18:00, 21:00 and 24:00 h during two successive days. Behavior was video taped in the home cages for posterior categorization and quantification. Radioimmunoassays were used to evaluate plasmatic levels of melatonin and corticosterone. Blood samples were obtained at the times of behavioral observation. In the light-dark condition the results showed day-night variation of general activity (p &lt; 0.001) and a robust diurnal rhythm of plasmatic melatonin (p &lt; 0.001). Both of these variations as well as the oscillatory secretion of corticosterone disappeared under constant light condition. The parallel changes in general activity and blunting of melatonin rhythm secretion in constant light condition agree with previous evidences that melatonin may regulate behavioral oscillations in the pigeon. The present data are related to the proposition that the timing system in pigeons may involve neuroendocrine relations characterized by interactions between blood born signalization by melatonin and corticosterone.  相似文献

Abstract

As part of studies on the interactive effects of light and endocrine factors on hepatic mixed‐function oxidases, the aminopyrine demethylase activity of liver microsomes and the serum concentrations of corticosterone and testosterone were measured in adult male rats (sham‐operated, castrated or adrenalectomized) exposed for 21 days to one of the following types of environmental lighting: (1) Normal light cyle (L from 09.00 to 21.00 h); (2) Reverse light cycle; (3) Constant light (LL) or (4) Constant darkness (DD). One half of the animals in each of the 12 groups was killed at 06.00 h and the other at 18.00 h on the last day of treatment. A 3‐way analysis of variance, multiple comparisons and correlations allowed the following conclusions: (1) The type of lighting had the most significant effect on demethylase activity, its highest values corresponding to periods of darkness (particularly in the rats exposed to a reverse light cycle) and its lowest to LL. (2) Castrated rats as a group had the lowest demethylase activity. (3) Time of death was a significant factor, particularly for the rats in a reverse light cycle (higher activity at 18.00 than at 06.00 h). (4) There was a significant and positive correlation between aminopyrine demethylase and the rates of 7α‐hydroxylation of 3β‐hydroxy‐5‐androsten‐17‐one previously reported for the same animals. (5) There was no significant correlation between demethylase activity and serum corticosterone or testosterone.  相似文献

Circadian rhythms in the emergence of S. mansoni cercariae from Biomphalaria tenagophila and B. glabrata, snail hosts of schistosomosis in Brazil, were investigated. A total of 35 specimens of B. tenagophila (São Paulo, Brazil) and 12 B. glabrata (Minas Gerais, Brazil) exposed individually to five miracidia of Schistosoma mansoni originated from the same biotope as their snail hosts, were tested. Observations were carried out at outdoors, with the quantification of cercarial emergence at 3h intervals during three consecutive days in November 1989 and in May 1990. Cercarial emergence was essentially diurnal (from 06.00-18.00h) in both species. Circadian rhythms were detected by the Single Cosinor Method among 74.3% of B. tenagophila and 91.7% of B. glabrata snails. The acrophases corresponding to individual snails were between 11.37 e 17.54h in B. tenagophila and between 14.15 and 16.29h in B. glabrata. These findings confirm our preliminary observations in B. tenagophila and are in accordance to those of other authors in regard to B. glabrata. The acrophases of individual snails were similar within each species, thus indicating that at populacional level cercarial emergence was concentrated in particular times of the day. Group acrophases for each species varied from 13.22 to 15.22h and were not significantly different between B. tenagophila and B. glabrata. Cercariae emerging from B. tenagophila snails seemed to be more sensitive to environmental temperature than those emerging from B. glabrata, at least in the temperature range prevailing along the tests. Further chronobiological studies on host-parasite interactions are encouraged to improve our knowledge about temporal aspects of schistosomosis transmission.  相似文献

Plant pathologists need to manage plant diseases at low incidence levels. This needs to be performed efficiently in terms of precision, cost and time because most plant infections spread rapidly to other plants. Adaptive cluster sampling with a data‐driven stopping rule (ACS*) was proposed to control the final sample size and improve efficiency of the ordinary adaptive cluster sampling (ACS) when prior knowledge of population structure is not known. This study seeks to apply the ACS* design to plant diseases at various levels of clustering and incidences levels. Results from simulation study show that the ACS* is as efficient as the ordinary ACS design at low levels of disease incidence with highly clustered diseased plants and is an efficient design compared with simple random sampling (SRS) and ordinary ACS for some highly to less clustered diseased plants with moderate to higher levels of disease incidence.  相似文献

The mathematical and statistical advances in fitting stock assessment models enabled the emergence of the paradigm of “integrated analysis”, which fits all available data to a single model of population dynamics that traditionally has total catch as the only forcing function of the system. This approach, however, allowed us to include, in a flexible way, the effect of hydrological regime as an additional forcing function. We tried to achieve this flexibility by making the annual recruitment rates and spawning biomass adjustable to the attributes of the hydrological cycle data. Our models showed that these attributes are influential in the population dynamics of Brycon hilarii of the Northern Pantanal, and their inclusion in the models allowed best partial fits (which considered fits only to the data components length- and age-compositions, CPUE of juveniles and adults) than the Base-case (without hydrological attributes). The best partial fits where obtained when the attributes “delay of floods” and “intensity of floods” were forcing the spawning biomass and the annual recruitment respectively, indicating that these characteristics of the population may be influenced by specific attributes of the water level. The use of integrated modeling contributed with the advancement of population ecology knowledge of rheophilic fish. It is recommended that freshwater fisheries management be integrated into the hydrology management.  相似文献
