The dwarf ucu (ultracurvata) mutants of Arabidopsis display vegetative leaves that are spirally rolled downwards and show reduced expansion along the longitudinal axis. We have previously determined that the UCU1 gene encodes a SHAGGY/GSK3-like kinase that participates in the signaling pathways of auxins and brassinosteroids. Here, we describe four recessive alleles of the UCU2 gene, whose homozygotes display helical rotation of several organs in addition to other phenotypic traits shared with ucu1 mutants. Following a map-based strategy, we identified the UCU2 gene, which was found to encode a peptidyl-prolyl cis/trans-isomerase of the FK506-binding protein family, whose homologs in metazoans are involved in cell signaling and protein trafficking. Physiological and double mutant analyses suggest that UCU2 is required for growth and development and participates in auxin and brassinosteroid signaling. 相似文献

Summary. Along the past recent years, two-photon excitation (TPE) microscopy has moved from the realms of technical curiosity to be a standard application in many advanced cell biology laboratories. The growing body of literature covered in this review points out the obvious advantages of TPE over any other imaging method based on fluorescence, clearly improving signal-to-noise ratio and thick-tissue penetration and showing added potential for vital imaging. Like any new technology that has to gain its own space, TPE microscopy is still going through the growing pains in which reproducible protocols, probes, and applications are scarce. Yet, the published reports and unpublished results covered in this review point out that TPE can eventually accommodate most available protocols and probes, most of the times with evident advantages. Further, the potential for plant sciences is obvious, as plant cells possess many absorbing molecules and structures and are routinely more opaque than tissues of other organisms. Abstract: A total of 175 spotted snapper Lutjanus guttatus were collected monthly in the Los Cobanos and Puerto La Libertad, El Salvador, from January to December 2000 to determine its feeding habits. The fishes studied ranged 9.8 - 58.0 cm in total length. Were collected using push-net and hook and line. This snapper is a bentonic opportunistic carnivorous predator. The total biomass of the stomach contents was 260.5 g. Crustaceans (Squillidae, Portunidae, Dynomenidae. Penaeidae, Sicyoniidae, Callianassidae), were the most abundant group: they accounted for 50.4% of the total biomass. Numerically, Portunus asper was the most abundant prey. Ontogenic differences were observed in the diet. In juveniles (16 cm TL). at any time of the year, the most frequent and abundant components were crustaceans and in adults were crustaceans, fishes and mollusks. The relative importance of different components of the diet was assessed with two indexes that combine. in different ways. percentage frequency of occurrence, percentage number and percentage weight of prey categories. This paper describes the Punta Cocles reef (Limón, Costa Rica). Data were obtained by sampling nine transects along the coast and observations done by skin diving between September and November of 2002. This reef consist of 10.5 hectares, where 13 species of corals, 39 of macroalgae, two of seagrasses, two of zoantids, one anemone, one corallimorpharian, and one sponge were identified. Life coral coverage (16%), was higher than in other years (5% for 1985, and 13.2% for 1995), and death coral coverage was very low (0.2%). Macroalgae have the highest coverage (59%), particularly brown algae with a patchy distribution of Sargassum and Padina. Laurencia brongniartii (Rhodophyta) is added to the list of marine flora of Costa Rica. The Punta Cocles reef works as a refuge for organisms, because there are no towns or river mouths nearby, and because of the coast formation. The refuge character is enhanced by the environmental conscience of the people that live close to the reef and help to protect the environment. 相似文献

We describe the female and male of the Neotropical fungus-growing ant Mycerarotes carinatus, hitherto only known from type locality, Rio de Janeiro state, Brazil, based on samples of workers. The sexual forms were obtained from a nest maintained in the laboratory. The samples found in Minas Gerais state, expand the geographic distribution of the species. Trypanosoma evansi contains protein kinases capable of phosphorylating endogenous substrates with apparent molecular masses in the range between 20 and 205 kDa. The major phosphopolypeptide band, pp55, was predominantly localized in the particulate fraction. Anti-alpha and anti-beta tubulin monoclonal antibodies recognized pp55 by Western blot analyses, suggesting that this band corresponds to phosphorylated tubulin. Inhibition experiments in the presence of emodin, heparin, and 2,3-bisphosphoglycerate indicated that the parasite tubulin kinase was a casein kinase 2 (CK2)-like activity. GTP, which can be utilized instead of ATP by CK2, stimulated rather than inactivated the phosphorylation of tubulin in the parasite homogenate and particulate fraction. However, GTP inhibited the cytosolic CK2 responsible for phosphorylating soluble tubulin and other soluble substrates. Casein and two selective peptide substrates, P1 (RRKDLHDDEEDEAMSITA) for casein kinase (CK1) and P2 (RRRADDSDDDDD) for CK2, were recognized as substrates in T. evansi. While the enzymes present in the soluble fraction predominantly phosphorylated P1, P2 was preferentially labeled in the particulate fractions. These results demonstrated the existence of CK1-like and CK2-like activities primarily located in the parasite cytosolic and membranous fractions, respectively. Histone II-A and kemptide (LRRASVA) also behaved as suitable substrates, implying the existence of other Ser/Thr kinases in T. evansi. Cyclic AMP only increased the phosphorylation of histone II-A and kemptide in the cytosol, demonstrating the existence of soluble cAMP-dependent protein kinase-like activities in T. evansi. However, no endogenous substrates for this enzyme were identified in this fraction. Further evidences were obtained by using PKI (6-22), a reported inhibitor of the catalytic subunit of mammalian cAMP-dependent protein kinases, which specifically hindered the cAMP-dependent phosphorylation of histone II-A and kemptide in the parasite soluble fraction. Since the sum of the values obtained in the parasite cytosolic and particulate fractions were always higher than the values observed in the total T. evansi lysate, the kinase activities examined here appeared to be inhibited in the original extract. 相似文献
