Abstract

The carboxamide functionality of tiazofurin 1a has been modified to produce the following analogs: carboximidates 5a,b, carboxamidines 6, 10, tetrahydropyrimidine 7, N-glycine 8 and N-glutamine 9. These structural modifications abolished the in vitro antiviral (RNA) activity exhibited by tiazofurin against the flaviviruses (yellow fever and Japanese encephalitis viruses), bunyavirus (Punta Toro virus) and togavirus (Venezuelan equine encephalomyelitis virus). Only carboximidates 5a,b retained marginal activity against bunyaviruses.  相似文献

Isolation of Legionella from water samples using various culture methods   总被引:3,自引:2,他引:1  
The efficacy of a non-selective medium and two selective media were compared for the isolation of legionellas from water samples. The effect of acid wash treatment for decontamination of the water samples on the isolation frequency of legionellas was also studied. The 236 samples were taken from cooling, humidifying and drinking water systems; 21% were legionella-positive when inoculated directly on modified Wadowsky-Yee (MWY) medium and 26% were positive when concentrated (x 200) before cultivation on MWY or CCVC media. Inoculation on MWY medium after concentration followed by decontamination by the acid-wash technique gave the highest isolation frequency (31%). The lowest frequency (8%) was found with the non-selective BCYEα medium. An isolation frequency of 28% was achieved with the BCYEα medium after concentration and acid-wash treatment of the samples. Forty per cent of the samples were positive for legionellas when the results from all the culture methods were combined. Not all the legionella-positive samples were identified by a single culture method. Ninety-three of the 95 positive samples were detected with the two best combinations of three culture methods. The best culture method for detecting legionellas depended on the source of the water sample. Some water quality characteristics, like temperature and organic matter content, affected the isolation frequency of Legionella spp.  相似文献

The meiofauna was studied from springs in Finland which wascompletely ice-covered during the last glacial epoch. Of the31springs that were examined, 15 were in areas considered to bein anatural state and 16 had catchment areas subject toanthropogenicdisturbance from one of four sources: agriculture,inhabitation,gravel extraction or winter road de-icing with NaCl. The taxarecorded were a mixture of those already described fromepigeicwaters or groundwaters and some which may be regarded ascharacteristic of springs Statistical analysis revealeddifferencesin the meiofaunal taxa in relation to the water quality orcatchment area variables. Of the major meiofaunal groups thenematodes favoured springs influenced by agriculture,oligochaeteswere most tolerant of road de-icing, cladocerans and copepodspreferred springs in a natural state, ostracods couldwithstand theinfluence of road de-icing and gravel extraction, whilstdipteranlarvae could withstand the influence of agriculture.Oligochaetescomprised 13.4% of meiofaunal numbers. The most abundantspecieswas Aeolosoma quaternarium, which was most numerous insprings in a natural state, while Dero digitatapreferredeutrophic conditions. Harpacticoids comprised 14.1% ofindividualsand clearly preferred a natural state, Bryocamptusechinatusand Moraria brevipes being the most abundant species.Ostracods, of which the most abundant species was Potamocyprispallida, were the most numerous animal group, constituting21% ofthe meiofaunal population, and their numbers were greatest insprings influenced by road de-icing or gravel extraction.Dipteranlarvae, of which the majority were chironomids, comprised14.9% ofindividuals. The ostracods Potamocypris pallida andthreespecies of Candona and the harpacticoid Bryocamptuscuspidatus are new records for Finland.  相似文献

The interaction of an amphiphilic, 40-amino acid β-amyloid (Aβ) peptide with liposomal membranes as a function of sterol mole fraction (Xsterol) was studied based on the fluorescence anisotropy of a site-specific membrane sterol probe, dehydroergosterol (DHE), and fluorescence resonance energy transfer (FRET) from the native Tyr-10 residue of Aβ to DHE. Without Aβ, peaks or kinks in the DHE anisotropy versus Xsterol plot were detected at Xsterol ≈ 0.25, 0.33, and 0.53. Monomeric Aβ preserved these peaks/kinks, but oligomeric Aβ suppressed them and created a new DHE anisotropy peak at Xsterol ≈ 0.38. The above critical Xsterol values coincide favorably with the superlattice compositions predicted by the cholesterol superlattice model, suggesting that membrane cholesterol tends to adopt a regular lateral arrangement, or domain formation, in the lipid bilayers. For FRET, a peak was also detected at Xsterol ≈ 0.38 for both monomeric and oligomeric Aβ, implying increased penetration of Aβ into the lipid bilayer at this sterol mole fraction. We conclude that the interaction of Aβ with membranes is affected by the lateral organization of cholesterol, and hypothesize that the formation of an oligomeric Aβ/cholesterol domain complex may be linked to the toxicity of Aβ in neuronal membranes.  相似文献

The presence and distribution of desmin, vimentin, cytokeratin, and laminin in the gonads of developing male rat embryos (11-17 days) were studied by immunocytochemistry. The findings were correlated with morphological changes of the cells and with the formation of basement membranes, as determined by electron microscopy. The surface epithelial and subepithelial cells of the meesonephros in the prospective gonadal region contained desmin. At the onset of gonadal development, vimentin appeared in the somatic cells of the thickening surface epithelium, which formed the gonadal ridge. Desmin disappeared and cytokeratins appeared in the Sertoli precursor cells at the inception of their epithelial differentiation. Simultaneously, the prospective Sertoli cells became polarized during their assembly into epithelial cell aggregates; the aggregates then fused and formed elongated testicular cords. The epithelial cell differentiation was accompanied by a deposition of basement membrane material around the cords and by an increase of desmin in the cells immediately around the cords. With further differentiation of the testicular cords, some cytokeratins from the Sertoli cells, but not from the cells of the rete cords, disappeared. On the other hand, other cytokeratin polypeptides and vimentin remained in the fetal Sertoli cells. The surface cell layer slowly differentiated towards a proper epithelium after the basic formation of the testicular cords and interstitium. Desmin and vimentin persisted in the interstitial cells throughout the entire study period. The early differentiation of the gonad is apparently under a general sex-independent initiation program. The developmental changes in intermediate filaments offer an opportunity for the further analysis of their general role in early organogenesis. In light of the genetic theory of testicular differentiation, the functions of the regulatory factor(s) include specific organization of cord cells, histological organization into looping cords rather than separated follicles, and male development of the interstitium, surface epithelium and tunica albuginea.  相似文献

Seven human lymph nodes showing different types of reactive change, were examined for the presence of acid cysteine-proteinase inhibitor (ACPI) by the peroxidase-antiperoxidase method. A clear positive reaction was found in the germinal centres. The staining pattern indicated immunoreactivity of the dendritic reticulum cells, but the possibility that other cells, particularly histiocytic reticulum cells, may also react with antiserum raised against ACPI cannot be excluded.  相似文献

The lymphoproliferative response of human peripheral blood mononuclear cells to different measles virus antigen preparations was studied with lymphocytes from 38 measlesseropositive healthy donors and 4 subacute sclerosing panencephalitis patients. The response was very weak or absent in all of the controls and in three of the subacute sclerosing panencephalitis patients. The fourth subacute sclerosing panencephalitis patient had fluctuating levels of lymphocyte stimulation by measles antigens. The response was very strong for several months and during this time the parameters of the test system were characterized. It was discovered that a membrane preparation of measles-infected cells caused stimulation equal to that of highly purified virions. Purified measles ribonucleoprotein also induced specific stimulation, although lower than that seen with other types of measles antigens. Results of experiments on stimulation kinetics and antigen dose responses were compatible with antigen-specific stimulation. Enriched T cells were more vigorously stimulated than unfractionated peripheral blood mononuclear cells suggesting that this transformation test is specific for T cells.  相似文献
