In this study we present a detailed, mechanism-based mathematical framework of Drosophila circadian rhythms. This framework facilitates a more systematic approach to understanding circadian rhythms using a comprehensive representation of the network underlying this phenomenon. The possible mechanisms underlying the cytoplasmic “interval timer” created by PERIOD–TIMELESS association are investigated, suggesting a novel positive feedback regulatory structure. Incorporation of this additional feedback into a full circadian model produced results that are consistent with previous experimental observations of wild-type protein profiles and numerous mutant phenotypes.  相似文献

Caloric restriction (CR) studies have shown that females rats conserve energy more efficiently, showing a higher resistance to weight loss and higher protection of vital organs mass than male rats. Gender-dependent inactivation of thermogenesis in brown adipose tissue (BAT) has been proposed as one of these possible energy conserving mechanisms. To study the changes underlying this inactivation in rats, a three month study with 40% CR was undertaken to unravel the effects on BAT recruitment. Under ad libitum conditions female rats had greater BAT recruitment and greater oxygen consumption than their male counterparts. Total and mitochondrial protein, as well as triglyceride and DNA content were more reduced in restricted female rats than in restricted males. Similarly, the levels of key BAT functional proteins (UCP1, LPL, HSL, TFAM) were more reduced in restricted females, whereas no changes were found in mitochondrial DNA levels (mtDNA) and OXPHOS activities in males and females. Furthermore, alpha (2A)/beta (3) adrenergic receptor ratio remained constant in male rats whereas in female rats CR increased 60%. In conclusion, our results suggest that female rats, whose BAT thermogenic activity is higher in ad libitum conditions, is depressed during CR. This inactivation involves the mitochondrial differentiation process and lipolytic system and could be due, at least in part, to the unfavourable adrenergic receptor balance for thermogenic activation.  相似文献

In alstroemeria ( Alstroemeria hybrida ), leaf senescence is effectively retarded by the application of gibberellins and by low fluences of red light. In this study we examined the possible interaction of gibberellins and red light in the regulation of senescence. Determination of endogenous gibberellins revealed that leaf senescence is accompanied by significant changes in the concentrations of non‐ 13‐hydroxylated gibberellins, the onset of senescence coinciding with a dramatic drop in GA4, whereas concentrations of 13‐hydroxylated gibberellins are far less influenced. However, no direct effect of red light on a specific GA‐metabolic step could be determined. When exogenously applied, non‐13‐hydroxylated GAs were more active than the 13‐hydroxylated GAs. It appeared that the effect of red light is additive to that of active GAs. We hypothesise that GA4 and phytochrome control senescence in alstroemeria mainly through separate mechanisms and have independent effects and that the observed differences in gibberellin concentrations are a consequence of delayed leaf senescence rather than a cause for it.  相似文献

The photosynthetic response of the purple sulfur bacterium Chromatium vinosum DSM 185 to different degrees of illumination was analyzed. The microorganism was grown in continuous culture, and samples were taken from the effluent of the culture and incubated at different irradiances to determine the specific rate of sulfur oxidation as a measure of the photosynthetic activity of the organism. The activities obtained were plotted as a function of the specific rate of light uptake, and for each set of data a photosynthesis equation was fitted, which allowed the estimation of Pmax (photosynthetic capacity), qk (the threshold irradiance for light limitation), and m (maintenance coefficient). The results indicated that cells grown under light limitation are able to achieve higher photosynthetic activities than cells grown under light saturation. The photosynthetic capacity (Pmax) remained constant under all the conditions of illumination tested, while the maintenance expenses (m) were higher under light limitation. Mice vaccinated with a single injection of formalin-inactivated suspensions of mouse brain tissue infected with arboviruses were markedly protected against a challenge injection administered hours later. The protection observed during the first 2 days after vaccination seemed to be nonspecific, in that it appeared not only with the homologous system but also between arboviruses of different antigenic groups; this phase may be associated with an interferon-like activity of the serum. Overlapping the nonspecific phase was one of specific protection, which seemed to be well-established in its own right by day 3 or 4 after vaccination. Serum neutralizing antibodies against the homologous viruses were detected as early as 24 h after vaccination and in almost all instances by day 3.  相似文献

Pathogen-induced host phenotypic changes are widespread phenomena that can dramatically influence host–vector interactions. Enhanced vector attraction to infected hosts has been reported in a variety of host–pathogen systems, and has given rise to the parasite manipulation hypothesis whereby pathogens may adaptively modify host phenotypes to increase transmission from host to host. However, host phenotypic changes do not always favour the transmission of pathogens, as random host choice, reduced host attractiveness and even host avoidance after infection have also been reported. Thus, the effects of hosts’ parasitic infections on vector feeding behaviour and on the likelihood of parasite transmission remain unclear. Here, we experimentally tested how host infection status and infection intensity with avian Plasmodium affect mosquito feeding patterns in house sparrows (Passer domesticus). In separate experiments, mosquitoes were allowed to bite pairs containing (i) one infected and one uninfected bird and (ii) two infected birds, one of which treated with the antimalarial drug, primaquine. We found that mosquitoes fed randomly when exposed to both infected and uninfected birds. However, when mosquitoes were exposed only to infected individuals, they preferred to bite the non-treated birds. These results suggest that the malarial parasite load rather than the infection itself plays a key role in mosquito attraction. Our findings partially support the parasite manipulation hypothesis, which probably operates via a reduction in defensive behaviour, and highlights the importance of considering parasite load in studies on host–vector–pathogen interactions.  相似文献
