One to two percent of all children are born with a developmental disorder requiring pediatric hospital admissions. For many such syndromes, the molecular pathogenesis remains poorly characterized. Parallel developmental disorders in other species could provide complementary models for human rare diseases by uncovering new candidate genes, improving the understanding of the molecular mechanisms and opening possibilities for therapeutic trials. We performed various experiments, e.g. combined genome-wide association and next generation sequencing, to investigate the clinico-pathological features and genetic causes of three developmental syndromes in dogs, including craniomandibular osteopathy (CMO), a previously undescribed skeletal syndrome, and dental hypomineralization, for which we identified pathogenic variants in the canine SLC37A2 (truncating splicing enhancer variant), SCARF2 (truncating 2-bp deletion) and FAM20C (missense variant) genes, respectively. CMO is a clinical equivalent to an infantile cortical hyperostosis (Caffey disease), for which SLC37A2 is a new candidate gene. SLC37A2 is a poorly characterized member of a glucose-phosphate transporter family without previous disease associations. It is expressed in many tissues, including cells of the macrophage lineage, e.g. osteoclasts, and suggests a disease mechanism, in which an impaired glucose homeostasis in osteoclasts compromises their function in the developing bone, leading to hyperostosis. Mutations in SCARF2 and FAM20C have been associated with the human van den Ende-Gupta and Raine syndromes that include numerous features similar to the affected dogs. Given the growing interest in the molecular characterization and treatment of human rare diseases, our study presents three novel physiologically relevant models for further research and therapy approaches, while providing the molecular identity for the canine conditions.  相似文献

The number of African buffaloes (Syncerus caffer Sparrman, 1779) inhabiting Ruaha National Park, Tanzania, is thought to be declining, but little data exist to determine whether the population is actually in decline. As an initial phase of collecting population data, we conducted demographic surveys, faecal egg counts and gastrointestinal parasite identification in Ruaha's buffalo herds in September 2011 and 2013. Most herds encountered in the two surveys appeared to be in good health, but with fewer calves in 2013 compared with 2011. The herd‐level body condition score was positively associated with the number of calves per 100 cows after adjusting for year, and the lower number of offspring in 2013 could possibly be associated with a below average rainfall in the 2012–2013 rainy season. Mean herd‐level egg counts ranged from 83 to 140 and from 28 to 113 eggs per g faeces in 2011 and 2013, respectively. Haemonchus, Nematodirus, Cooperia and Oesophagostomum spp., as well as coccidian oocysts, were detected in the population. Monitoring herd demographics and baseline health parameters over time will provide insight into population performance, increase the understanding of population stressors and contribute to buffalo conservation within Ruaha National Park and other protected areas of Africa.  相似文献

The population dynamics of the enchytraeid Cognettia sphagnetorum originating from an unmanaged forest (FP), a clear-cut area (CCP) or a plot treated with birch ash (APP) and the effects of population origin on labile C and N dynamics were investigated. Twenty individuals of C. sphagnetorum were introduced in microcosms containing humus from the unmanaged forest devoid of enchytraeids and amended with sucrose, and incubated for 14 weeks. Triplicate microcosms from FP, CCP and APP treatments were destructively sampled every second week and enchytraeid population density, individual length, nematode abundance and trophic structure, humus properties and dissolved organic C (DOC) and N (DON), and NH4–N in soil were determined. The enchytraeid body size was initially smaller in CCP and APP than in FP. The enchytraeid propagation rate was lower and individual size less variable in APP than in FP or CCP, and although enchytraeid size increased in all treatments, exponential population models indicated that APP was less stable. Nematode community was dominated by bacterial-feeders especially in the microcosms with APP. N mineralization rate was lower and DOC decomposition rate greater in APP systems. The results show that C. sphagnetorum is more sensitive to wood ash than clear-cutting, and its altered body size distribution has the potential to affect the dynamics of soluble nutrients.  相似文献

The development of high-risk human papillomavirus (hrHPV) infection to cervical cancer is a complicated process. We considered solely hrHPV infections, thus avoiding the confounding effects of disease progression, screening, and treatments. To analyse hrHPV epidemiology and to estimate the overall impact of vaccination against infections with hrHPVs, we developed a dynamic compartmental transmission model for single and multiple infections with 14 hrHPV types. The infection-related parameters were estimated using population-based sexual behaviour and hrHPV prevalence data from Finland. The analysis disclosed the important role of persistent infections in hrHPV epidemiology, provided further evidence for a significant natural immunity, and demonstrated the dependence of transmission probability estimates on the model structure. The model predicted that vaccinating girls at 80% coverage will result in a 55% reduction in the overall hrHPV prevalence and a higher 65% reduction in the prevalence of persistent hrHPV infections in females. In males, the reduction will be 42% in the hrHPV prevalence solely by the herd effect from the 80% coverage in girls. If such high coverage among girls is not reached, it is still possible to reduce the female hrHPV prevalence indirectly by the herd effect if also boys are included in the vaccination program. On the other hand, any herd effects in older unvaccinated cohorts were minor. Limiting the epidemiological model to infection yielded improved understanding of the hrHPV epidemiology and of mechanisms with which vaccination impacts on hrHPV infections.  相似文献

Mechano-acoustic and elastographic techniques may provide quantitative means for the in vivo diagnostics of articular cartilage. These techniques assume that sound speed does not change during tissue loading. As articular cartilage shows volumetric changes during compression, acoustic properties of cartilage may change affecting the validity of mechano-acoustic measurements. In this study, we examined the ultrasound propagation through human, bovine and porcine articular cartilage during stress-relaxation in unconfined compression. The time of flight (TOF) technique with known cartilage thickness (true sound speed) as well as in situ calibration method [Suh, Youn, Fu, J. Biomech. 34 (2001), 1347-1353] were used for the determination of sound speed. Ultrasound speed and attenuation decreased in articular cartilage during ramp compression, but returned towards the level of original values during relaxation. Variations in ultrasound speed induced an error in strain and compressive moduli provided that constant ultrasound speed and time-of-flight data was used to determine the tissue thickness. Highest errors in strain (-11.8 +/- 12.0%) and dynamic modulus (15.4 +/- 17.9%) were recorded in bovine cartilage. TOF and in situ calibration methods yielded different results for changes in sound speed during compression. We speculate that the variations in acoustic properties in loaded cartilage are related to rearrangement of the interstitial matrix, especially to that of collagen fibers. In human cartilage the changes, are, however relatively small and, according to the numerical simulations, mechano-acoustic techniques that assume constant acoustic properties for the cartilage will not be significantly impaired by this phenomenon.  相似文献

We studied the effect of cyanobacteria on foraging and refuge use in small fish. We measured pike larval feeding in the presence of cyanobacteria by counting leftover prey. Our results showed that feeding by pike larvae on zooplankton prey decreased significantly in the presence of non-toxic cyanobacteria. The behaviour can be due to lowered vision caused by turbidity or clogging of the gills. Further, we tested whether the three-spined stickleback use toxic cyanobacteria as a refuge against predators in a choice experiment. The choice experiment was performed in a Y-maze fluviarum, where the fish could select between two different environments. Our results support the refuge use hypothesis because the three-spined stickleback clearly preferred toxic cyanobacteria to the chemical predator signal. To conclude, cyanobacteria decrease feeding rates in fish larvae, but may function as important refuge for e.g. sticklebacks, during predation pressure in pelagic algal blooms.  相似文献

Prey may select suboptimal habitat to alleviate predation risk. Algal blooms and turbidity are potentially harmful to prey in aquatic environments, but can provide refugia against predation, given that predators avoid such conditions. Using a flow-through aquarium, we experimentally studied the habitat choice of the three-spined stickleback (Gasterosteus aculeatus L.) provided with toxic and non-toxic bloom-forming cyanobacteria and green flagellate-induced turbidity in the presence and absence of a chemical predator signal from a perch (Perca fluviatilis L.). We investigated whether sticklebacks separate between different algal strains and between turbid and clear water, and whether they are able to use algal toxicity and turbidity as shelter against predators. Sticklebacks preferred the toxic over the non-toxic Nodularia spumigena (Mertens) habitat in the presence of a predator signal, whereas no differences in times spent in the two habitats were detected when the predator signal was absent. There was a tendency for sticklebacks to prefer clear over turbid water in the absence of a predator signal, but no differences were found when the predator signal was present. Our results suggest that the three-spined stickleback is not fully adapted to the cyanobacterial blooms and turbidity caused by the recent eutrophication of the Baltic Sea. However, the predator-induced shifts in habitat choice are also consistent with the hypothesis that sticklebacks use algal toxicity and turbidity as shelters against predation, since these factors are likely to have only minor fitness consequences for sticklebacks.  相似文献
