Background aimsThe aim of this study was to investigate the correlation between infused lymphocyte populations and lymphocyte subsets at engraftment, and the early clinical implications of lymphocyte subset recovery after autologous stem cell transplantation (ASCT) in multiple myeloma (MM).MethodsWe examined the lymphocyte populations of infused autografts and the lymphocyte subsets of peripheral blood at engraftment from 50 patients using flow cytometry. Each subset was grouped as low (below median) and high (above median) to examine the correlation with mucositis of grade 3 or more and the occurrence of infections and cytomegalovirus (CMV) reactivation.ResultsUsing Spearman correlation coefficients, we found that cell doses of infused CD8+ (P = 0.042) and CD19+ cells (P = 0.044) were significantly associated with the absolute lymphocyte count (ALC) at engraftment. The dose of infused CD34+ cells was not associated with the change of lymphocyte subsets except for an inverse correlation with CD4+ cells (P = 0.006). After adjusting for potential variables in univariate analysis, multivariate analyzes revealed that the lower ratio of infused CD4+ to CD8+ cells (P = 0.030) was an independent factor for severe mucositis. Of lymphocyte subsets at engraftment, a higher frequency of CD3+ (P = 0.024) and a lower frequency of CD56+ (P = 0.020) were independent predictors for infections after engraftment. A higher frequency of CD8+ cells (P = 0.041) and a lower ratio of CD4+ to CD8+ (P = 0.021) were independent predictors for CMV reactivation.ConclusionsOur data suggest that lymphocyte subset analysis of infused autograft and peripheral blood at engraftment may provide new predictors for early complications after ASCT in patient with MM.  相似文献

Rottlerin, a compound reported to be a PKC δ-selective inhibitor, has been shown to induce growth arrest or apoptosis of human cancer cell lines. In our study, rottlerin dose-dependently induced apoptotic cell death in colon carcinoma cells. Treatment of HT29 human colon carcinoma cells with rottlerin was found to induce a number of signature ER stress markers; phosphorylation of eukaryotic initiation factor-2α (eIF-2α), ER stress-specific XBP1 splicing, and up-regulation of glucose-regulated protein (GRP)-78 and CCAAT/enhancer-binding protein-homologous protein (CHOP). However, suppression of PKC δ expression by siRNA or overexpression of WT-PKC δ and DN-PKC δ did not abrogate the rottlerin-mediated induction of CHOP. These results suggest that rottlerin induces up-regulation of CHOP via PKC δ-independent pathway. Furthermore, down-regulation of CHOP expression using CHOP siRNA attenuated rottlerin-induced apoptosis. Taken together, the present study thus provides strong evidence to support an important role of ER stress response in mediating the rottlerin-induced apoptosis.  相似文献

The retinoblastoma protein (RB) plays an important role in growth suppression through the formation of multiple protein complexes with its target proteins using A/B and C pockets. Even though the A/B and C pockets co-operate for growth suppression, the function of RB in growth arrest is inhibited by the coexpression of RB C fragments with full length RB in the absence of p53, which implies that C pocket fragments are likely to act as a dominant-negative inhibitor of RB function. In contrast, the loss of the RB functions in the presence of p53 triggers a cell cycle arrest or apoptosis by p53-dependent pathways. Thus, it still remains to be elucidated whether the expression of RB C pocket fragments in the presence of p53 induces delayed cell cycle progression and sensitizes cells to apoptosis through p53-dependent pathways. Our results show that the expression of RB C pocket fragments not only induces delayed cell cycle progression, which is mediated by the down-regulation of cyclin A, cyclin E, and E2F-1, but also sensitizes cells to apoptosis through p53-dependent pathways.  相似文献

The human neuronal apoptosis inhibitory protein (NAIP) gene was originally discovered because of its deletion in infantile spinal muscular atrophy (SMA), a childhood genetic disorder characterized by motor neuron loss and progressive paralysis with muscular atrophy. Although SMA is now known to be caused by deletions of survival motor neuron (SMN), the fact that NAIP is an anti-apoptotic protein is consistent with the NAIP gene modifying SMA severity. Here we report the cloning of a 1.5 kb rat NAIP cDNA fragment which contains BIR-3 (third baculovirus inhibitory repeat) domain. This fragment shows 78% homology to the human NAIP and 86% homology to the murine counterpart. We have investigated the distribution of NAIP mRNA expressing neurons by in situ RT-PCR technique in the rat central nervous system (CNS). Although all of the neurons appeared to express NAIP mRNA ubiquitously, pronounced elevation of NAIP mRNA expression was observed in the areas innervated by glutamatergic neurons after kainic acid (KA) injection. We have raised an anti-rat NAIP antiserum in rabbits using NAIP cDNA and recombinant rat NAIP, and carried out an immunohistological investigation. We observed highly immunoreactive neuronal subpopulations in the retinal ganglion, cerebral cortex, hippocampus, basal forebrain, thalamus, areas of midbrain, Purkinje cells of the cerebellum, and motor neurons in the spinal cord. Increased immunoreactivity of glutamatergic neurons was also observed broadly in the CNS after KA treatment. This study provides additional evidence that expression of mRNA and gene products of NAIP seem to be regulated in response to excessive stimuli or injuries in rat CNS, and these results are compatible with an anti-apoptotic role of NAIP in acute SMA as well as in brain injuries.  相似文献

The South Korean species of the genus Apethymus Benson are studied. Here we report two species, A. kunugi Togashi, 2005, and A. sidorenkoi Sundukov, 2010, new to South Korea. The male of A. kolthoffi (Forsius, 1927) is described in detail. To identify and assess the distribution patterns of coastal dune vegetation along the eastern, southern, and western coasts of South Korea, we investigated the plant communities and soil factors at 30 sites. In all, 12 communities on CCA (canonical correspondence analysis) Axis 1 and 2 could be arranged into 3 groups: 1 ) 2 communities ofElymus mollis andIschaemum anthephoroides, with medium sand contents; 2) 6 communities ofCarex pumila, Carex kobomugi, Ixeris repens, Zoysia mac-rostachya, Calystegia soldanella, andVitex rotundifolia, with coarse sand contents; and 3) 4 communities ofLathyrus japonicus,Glehnia littoralis, Messerschmidia sibirica, andRosa rugosa, with very coarse sand contents. As identified via CCA ordination, the distribution of these groups was positively correlated with soil particle sizes.  相似文献

We studied how plant species distribution was regulated by the relationships between vegetation and soil factors on the southwestern coast of South Korea. Vegetation was classified using two-way indicator species analysis (TWINSPAN), thereby producing four vegetation groups that were linked to three habitat types. Two ordination techniques —; detrended correspondence analysis (DCA) and canonical correspondence analysis (CCA) —; were applied to examine the relationships between vegetation and 12 edaphic factors, including soil pH, water and osmotic potentials, moisture content, electrical conductivity, Cl and Na+ contents, total Kjeldahl nitrogen, and contents of organic matter, sand, silt, and clay. Results were similar for both types of evaluations. According to DCA and CCA, the 23 communities tended to cluster into three types: salt swamp, salt marsh, and sand dune. The first two canonical axes accounted for 14.9% of the community-soil factor relationship among communities. As identified via CCA, the main gradients were soil-water relations and soil texture.  相似文献

Hepatitis C virus (HCV) is highly dependent on cellular factors for its own propagation. By employing tandem affinity purification method, we identified pyruvate carboxylase (PC) as a cellular partner for NS5A protein. NS5A interacted with PC through the N-terminal region of NS5A and the biotin carboxylase domain of PC. PC expression was decreased in cells expressing NS5A and HCV-infected cells. Promoter activity of PC was also decreased by NS5A protein. However, FAS expression was increased in cells expressing NS5A and cell culture grown HCV (HCVcc)-infected cells. Silencing of PC promoted fatty acid synthase (FAS) expression level. These data suggest HCV may modulate PC via NS5A protein for its own propagation.  相似文献
