Simplified method for the collection, storage, and comet assay analysis of DNA damage in whole blood

Single-cell gel electrophoresis (comet assay) is one of the most common methods used to measure oxidatively damaged DNA in peripheral blood mononuclear cells (PBMC), as a biomarker of oxidative stress in vivo. However, storage, extraction, and assay workup of blood samples are associated with a risk of artifactual formation of damage. Previous reports using this approach to study DNA damage in PBMC have, for the most part, required the isolation of PBMC before immediate analysis or freezing in cryopreservative. This is very time-consuming and a significant drain on human resources. Here, we report the successful storage of whole blood in ~ 250 μl volumes, at − 80 °C, without cryopreservative, for up to 1 month without artifactual formation of DNA damage. Furthermore, this blood is amenable for direct use in both the alkaline and the enzyme-modified comet assay, without the need for prior isolation of PBMC. In contrast, storage of larger volumes (e.g., 5 ml) of whole blood leads to an increase in damage with longer term storage even at − 80 °C, unless a cryopreservative is present. Our “small volume” approach may be suitable for archived blood samples, facilitating analysis of biobanks when prior isolation of PBMC has not been performed. 相似文献

The goal of this review was to provide a historical overview of how molecular techniques have increased the understanding of the ecology and evolution of the family Syngnathidae (pipefishes, seahorses and seadragons). Molecular studies based primarily on mitochondrial DNA markers have proved their worth by elucidating complex phylogenetic relationships within the family. Phylogeographic studies, which have revealed how life-history traits and past climatic events shape geographic distributions and patterns of genetic variation within syngnathid species, also provide interesting case studies for the conservation and management of threatened species. The application of microsatellite DNA markers has opened a floodgate of studies concerned with the breeding biology of these fishes, which are interesting due to their unique reproductive mode of male pregnancy. Research in this area has contributed significantly to the understanding of mating patterns and sexual selection. Molecular markers may also be employed in studies of demography, migration and local breeding population sizes. Genomic studies have identified genes that are probably involved in male pregnancy and promise additional insights into various aspects of syngnathid biology at the level of the gene. Despite these advances, much more remains to be explored. Goals for future research should include: (1) a more inclusive phylogeny to resolve outstanding issues concerning the relationships within the family and higher order taxa, (2) a broader use of molecular studies to aid management and conservation efforts, (3) the inclusion of more genera in comparative behavioural studies and (4) the continued development of genomic resources for syngnathids to facilitate comparative genomic work. 相似文献

1. A number of planktonic cyanobacteria species form resting stages that survive in the sediments of lakes. The significance of this life history strategy to the ecology of new planktonic populations was investigated in Esthwaite Water, a mesotrophic lake in the English Lake District.
2. A simple trapping technique was used to quantify vertical movements of five species of buoyant gas-vacuolate cyanobacteria from close to the sediments, along a depth transect.
3. 'Recruitment' from the sediments was found to be widespread amongst the cyanobacteria species associated with the summer phytoplankton community.
4. Estimates of the vertical upward fluxes of cyanobacteria based upon trap catches could not account for observed increases in the planktonic populations suggesting that 'recruitment' was not a significant source of biomass.
5. Vertical upward movements of Anabaena solitaria were recorded prior to this species becoming established in the plankton suggesting that benthic populations might be a source of cells for initial pelagic growth of populations of this species.
6. Low numbers of vegetative filaments of Anabaena flos-aquae , Aphanizomenon flos-aquae and Oscillatoria agardhii were observed in the plankton through the winter. This paper describes a new approach to the calibration of thermal infrared measurements of leaf temperature for the estimation of stomatal conductance and illustrates its application to thermal imaging of plant leaves. The approach is based on a simple reformulation of the leaf energy balance equation that makes use of temperature measurements on reference surfaces of known conductance to water vapour. The use of reference surfaces is an alternative to the accurate measurement of all components of the leaf energy balance and is of potentially wide application in studies of stomatal behaviour. The resolution of the technique when applied to thermal images is evaluated and some results of using the approach in the laboratory for the study of stomatal behaviour in leaves of Phaseolus vulgaris L. are presented. We examined whether the effects of elevated CO2 on growth of 1-year old Populus deltoides saplings was a function of the assimilation responses of particular leaf developmental stages. Saplings were grown for 100 days
at ambient (approximately 350 ppm) and elevated (ambient + 200 ppm) CO2 in forced-air greenhouses. Biomass, biomass distribution, growth rates, and leaf initiation and expansion rates were unaffected
by elevated CO2. Leaf nitrogen (N), the leaf C:N ratio, and leaf lignin concentrations were also unaffected. Carbon gain was significantly
greater in expanding leaves of saplings grown at elevated compared to ambient CO2. The Rubisco content in expanding leaves was not affected by CO2 concentration. Carbon gain and Rubisco content were significantly lower in fully expanded leaves of saplings grown at elevated
compared to ambient CO2, indicating CO2-induced down-regulation in fully expanded leaves. Elevated CO2 likely had no overall effect on biomass accumulation due to the more rapid decline in carbon gain as leaves matured in saplings
grown at elevated compared to ambient CO2. This decline in carbon gain has been documented in other species and shown to be related to a balance between sink/source
balance and acclimation. Our data suggest that variation in growth responses to elevated CO2 can result from differences in leaf assimilation responses in expanding versus expanded leaves as they develop under elevated
CO2.

A gas chromatographic method for the determination of levorphanol in human plasma is described. The method utilizes extractive alkylation with tetrabutylammonium cation as the phase-transfer catalyst and pentafluorobenzyl bromide as the alkylating agent, and employs a structural analog, d-3-hydroxy-N-ethylmorphinan, as the internal standard. The pentafluorobenzyl ethers formed are separated by capillary gas chromatography and detected by electron capture. The method has good precision and accuracy for concentrations ranging from 0.25 ng/ml to 100 ng/ml and has been used to measure plasma concentrations as part of a study to evaluate the management of chronic neuropathic pain with levorphanol. 相似文献
