The integrins are a class of adhesion molecules which have been implicated in the homing of hemopoietic stem cells and in their restriction within the bone marrow. Integrins function as mediators of cell-extracellular matrix (ECM) interactions and also of cell-cell interactions. They are unique membrane receptors which are capable of activation, change in affinity, and change in expression. Because of their broad potential for modulation we examined the effect of a cytokine growth factor which is present constitutively in the marrow, interleukin 3 (IL3), on integrin-mediated adherence of hemopoietic progenitor cells to the matrix component fibronectin (FN). The multipotential murine cell line B6Sut and the committed granulocyte progenitor cell line FDCP-1 were used. Both of these cell lines have been shown to bind to FN-coated dishes and to dishes coated with the 120 kDa and 40 kDa chymotryptic fragments of FN. It was found that after a brief withdrawal of IL3 the cells lost 80% adherence to the 120 kDa FN fragment containing the RGD cell binding site. This loss of binding was not related to a loss of viability, appeared unrelated to the growth/survival activity of IL3, and was quickly reversible by readdition of the growth factor. Adhesion of these cells to the RGD site was likely mediated by α5b?1 integrin which was identified in the cell membrane of both cell lines, but present in low copy number in B6Sut cells. Two antibodies against the external and internal domains of α5 and one antibody against b?1 were used to study expression of the integrin. By flow cytometry the expression of α5 was found to decrease in both cell lines by 4 h in the absence of IL3. The relative mean fluorescence intensity for B6Sut cells decreased from 1.0 (control cells always in the presence of IL3) to 0.6 over 4 h, and for FDCP-1 cells the decrement was from 1.0 to 0.8. The loss of RGD-mediated adhesion in the absence of IL3 appeared to proceed through this decrement in expression of the integrin; a loss of affinity of the receptor for its substrate was not detected. The general modulation of integrin activity by growth factors is of great interest because of its potential negative impact on the endothelium in cytokine-treated patients, and also because of its potential positive impact on engraftment during clinical bone marrow transplantation. © 1995 Wiley-Liss, Inc.  相似文献

Systematic mutation of bacteriophage T4 lysozyme   总被引:22,自引:0,他引:22  
Amber mutations were introduced into every codon (except the initiating AUG) of the bacteriophage T4 lysozyme gene. The amber alleles were introduced into a bacteriophage P22 hybrid, called P22 e416, in which the normal P22 lysozyme gene is replaced by its T4 homologue, and which consequently depends upon T4 lysozyme for its ability to form a plaque. The resulting amber mutants were tested for plaque formation on amber suppressor strains of Salmonella typhimurium. Experiments with other hybrid phages engineered to produce different amounts of wild-type T4 lysozyme have shown that, to score as deleterious, a mutation must reduce lysozyme activity to less than 3% of that produced by wild-type P22 e416. Plating the collection of amber mutants covering 163 of the 164 codons of T4 lysozyme, on 13 suppressor strains that each insert a different amino acid substitutions at every position in the protein (except the first). Of the resulting 2015 single amino acid substitutions in T4 lysozyme, 328 were found to be sufficiently deleterious to inhibit plaque formation. More than half (55%) of the positions in the protein tolerated all substitutions examined. Among (N-terminal) amber fragments, only those of 161 or more residues are active. The effects of many of the deleterious substitutions are interpretable in light of the known structure of T4 lysozyme. Residues in the molecule that are refractory to replacements generally have solvent-inaccessible side-chains; the catalytic Glu11 and Asp20 residues are notable exceptions. Especially sensitive sites include residues involved in buried salt bridges near the catalytic site (Asp10, Arg145 and Arg148) and a few others that may have critical structural roles (Gly30, Trp138 and Tyr161).  相似文献

The hippocampus receives major noradrenergic and serotoninergic (5-HT) innervations which interact with corticosteroid-sensitive cells. However, the subregional localization of these actions and the corticosteroid receptor types involved have not been defined and current ligand binding techniques for estimating corticosteroid receptors are hampered by several methodological limitations. We have developed in situ hybridization histochemical techniques to allow specific and sensitive estimation of glucocorticoid (GR) and mineralocorticoid receptor (MR) mRNA expression in rat hippocampus. Investigation of the effects of 5,7-dihydroxytryptamine lesions of 5-HT neurons showed significantly reduced GR and MR mRNA expression in some hippocampal subregions. Both abnormal 5-HT neurotransmission and excessive corticosteroid secretion are associated with major affective disorders, particularly depression. The crucial interaction between these two systems may occur, at least in part, at the level of regulation of hippocampal corticosteroid receptor expression.  相似文献

Cytokinins from normal and witchesbroom malformed stems of proteas were determined by radioimmunoassay following sample resolution by high-performance liquid chromatography (HPLC). Material from the early stages of shoot malformation had increased cytokinin concentrations which, over time, declined to the concentrations found in normal-growing stems. The cytokinin complement of the malformed structures was different from that of normal stems. The high concentrations of isopentenyladenosine detected appear to be related to the loss of the correlative inhibition of lateral buds and the development of the witchesbroom structures and may result from localized changes in cytokinin biosynthesis and/or metabolism.  相似文献

Abstract The exact algorithms of two commonly used parsimony programs, Hennig86 by J. S. Farris and PAUP by D. Swofford, sometimes produce different solutions, and sometimes produce resolutions that are not supported by the data being analysed. The discrepancies apparently involve the treatment of missing entries, which can currently represent unknown data, inapplicable character and/or polymorphic taxa. Each of those potential sources of ambiguity is logically (if not computationally) different; with regard to binary characters, unknown data could be either 0 or 1, inapplicable characters are neither 0 nor 1 and polymorphisms are both 0 and 1. Resolutions that cannot be supported by any possible combination of known state attributions should either be flagged as such or suppressed entirely.  相似文献

Decapitation or red light irradiation (R) inhibited growth and Golgi-localized glucan synthetase (GS I) activity in the mesocotyl of intact maize (Zea mays L.) seedlings. Applied auxin (indole-3-acetic acid) prevented the effects of R and of decapitation on both growth and GS I. Auxin applied several hours after irradiation prevented any further decline in GS I but did not restore it. Mesocotyl segments incubated in solution elongated in response to auxin but lost GS I with time regardless of the presence of exogenous auxin.
