Background

Development of the neuromuscular junction (NMJ) is initiated by the formation of postsynaptic specializations in the central zones of muscles, followed by the arrival of motor nerve terminals opposite the postsynaptic regions. The post- and presynaptic components are then stabilized and modified to form mature synapses. Roles of ADAM (A Disintegrin And Metalloprotease) family proteins in the formation of the NMJ have not been reported previously.

Principal Findings

We report here that Meltrin β, ADAM19, participates in the formation of the NMJ. The zone of acetylcholine receptor α mRNA distribution was broader and excess sprouting of motor nerve terminals was more prominent in meltrin β–deficient than in wild-type embryonic diaphragms. A microarray analysis revealed that the preferential distribution of ephrin-A5 mRNA in the synaptic region of muscles was aberrant in the meltrin β–deficient muscles. Excess sprouting of motor nerve terminals was also found in ephrin-A5 knockout mice, which lead us to investigate a possible link between Meltrin β and ephrin-A5-Eph signaling in the development of the NMJ. Meltrin β and EphA4 interacted with each other in developing motor neurons, and both of these proteins localized in the NMJ. Coexpression of Meltrin β and EphA4 strongly blocked vesicular internalization of ephrin-A5–EphA4 complexes without requiring the protease activity of Meltrin β, suggesting a regulatory role of Meltrin β in ephrin-A5-Eph signaling.

Conclusion

Meltrin β plays a regulatory role in formation of the NMJ. The endocytosis of ephrin-Eph complexes is required for efficient contact-dependent repulsion between ephrin and Eph. We propose that Meltrin β stabilizes the interaction between ephrin-A5 and EphA4 by regulating endocytosis of the ephrinA5-EphA complex negatively, which would contribute to the fine-tuning of the NMJ during development.  相似文献

Spinal cord injury typically results in permanent disability. Many studies have indicated that transplantation of several different types of stem cells promotes functional recovery in animal models of spinal cord injury. A conceptually different approach to utilize stem cells for regenerative therapies may be recruitment of endogenous neural stem cells resident in the adult spinal cord. We discuss the possibilities, risks, and mechanisms for stem cells in spinal cord repair.  相似文献

The toxicity of many heavy metals in plants is closely associated with its subcellular distribution and chemical forms. The subcellular distribution and chemical forms of cobalt (Co2+) were investigated using 3 barley genotypes differing in Co2+ toxicity resistance, namely Yan66 (resistant), Ea 52 (sensitive), and Humai 4 (moderate), under two Co2+ levels (25 and 100 µM). Higher Co2+ level in cultural solution significantly increased Co2+ accumulation in all subcellular fractions, with vacuole and cell wall having higher concentration. In comparison with 25 µM Co2+, 100 µM Co2+ treatment caused significant increase of Co2+ concentration in the forms of F-NaCl (extracted with 1 M NaCl), F-Ac (extracted with 2% HAc), F-HCl (extracted by 0.6 M HCl), and F-residue (residue forms) in both shoots and roots. There was a significant difference among genotypes in Co2+ subcellular distribution and chemical forms, with Ea52 accumulating more Co2+ in organelles and Yan66 accumulating more Co2+ in vacuole and cell wall. Moreover, the inorganic form of Co2+ extracted with 80% ethanol (F-ethanol) and water-soluble form (F-H2O) were significantly increased in Ea52, while Yan66 accumulated more Co2+ in the forms of low-bioavailable molecules (F-NaCl, F-HAc, and F-HCl). The results suggest that the vacuolar sequestration and cell wall deposition of Co2+ is a key resistant mechanism for genotype Yan66.  相似文献

High-throughput screening detected transaminases (TAs) and monoamine oxidases (MAOs) in fungi by applying a fluorogenic probe. Strains F026, F037, F041, F053, and F057 showed the highest enzymatic conversions (31, 60, 30, 40, and 32%, respectively) and where evaluated for their ability to transform piperidines. Strain F053 (Neopestalotiopsis sp. CBMAI 2030) revealed unusual enzymatic activity to deracemize 2-methyl-6-alkylpiperidines. Neopestalotiopsis sp. CBMAI 2030 was capable to convert 2-methyl-6-propylpiperidine, 2-methyl-6-butylpiperidine, and 2-methyl-6-pentylpiperidine in piperideine with 11, 14, and 24% conversion, respectively. The activity was enhanced by cultivating the fungus with 2-methyl-6-pentylpiperidine (38% conversion and 73% ee).  相似文献

Nonsystematically collected, a.k.a. opportunistic, species observations are accumulating at a high rate in biodiversity databases. Occupancy models have arisen as the main tool to reduce effects of limited knowledge about effort in analyses of opportunistic data. These models are generally using long closure periods (e.g., breeding season) for the estimation of probability of detection and occurrence. Here, we use the fact that multiple opportunistic observations in biodiversity databases may be available even within days (e.g., at popular birding localities) to reduce the closure period to 1 day in order to estimate daily occupancies within the breeding season. We use a hierarchical dynamic occupancy model for daily visits to analyze opportunistic observations of 71 species from nine wetlands during 10 years. Our model derives measures of seasonal site use within seasons from estimates of daily occupancy. Comparing results from our “seasonal site use model” to results from a traditional annual occupancy model (using a closure criterion of 2 months or more) showed that our model provides more detailed biologically relevant information. For example, when the aim is to analyze occurrences of breeding species, an annual occupancy model will over‐estimate site use of species with temporary occurrences (e.g., migrants passing by, single itinerary prospecting individuals) as even a single observation during the closure period will be viewed as an occupancy. Alternatively, our model produces estimates of the extent to which sites are actually used. Model validation based on simulated data confirmed that our model is robust to changes and variability in sampling effort and species detectability. We conclude that more information can be gained from opportunistic data with multiple replicates (e.g., several reports per day almost every day) by reducing the time window of the closure criterion to acquire estimates of occupancies within seasons.  相似文献

Pandoraea sp. MA03 wild type strain was subjected to UV mutation to obtain mutants unable to grow on propionic acid (PA) but still able to produce poly(3‐hydroxybutyrate‐co‐3‐hydroxyvalerate) [P(3HB‐co‐3HV)] from glycerol and PA at high 3HV yields. In shake flask experiments, mutant prp25 was selected from 52 mutants affected in the propionate metabolism exhibiting a conversion rate of PA into 3HV units of 0.78 g g?1. The use of crude glycerol (CG) plus PA or valeric acid resulted in a copolymer with 3HV contents varying from 21.9 to 30 mol% and 22.2 to 36.7 mol%, respectively. Fed‐batch fermentations were performed using CG and PA and reached a 3HV yield of 1.16 g g?1, which is 86% of the maximum theoretical yield. Nitrogen limitation was a key parameter for polymer accumulation reaching up to 63.7% content and 18.1 mol% of 3HV. Henceforth, mutant prp25 is revealed as an additional alternative to minimize costs and support the P(3HB‐co‐3HV) production from biodiesel by‐products. © 2017 American Institute of Chemical Engineers Biotechnol. The effects of tagging on small cetaceans are difficult to assess due to logistical difficulties in recapturing the whales. In this study two narwhals, Monodon monoceros, and five harbor porpoises, Phocoena phocoena, were recaptured between 297 and 767 days after instrumentation with satellite transmitters. The transmitters were mounted by pins that were pushed through the fins of the porpoises or the backs of the narwhals. Overall body condition seemed unaffected by the instrumentations. Macroscopical examination revealed that umbilicalization of the tissue surrounding the pins was almost complete. On one of the narwhals the reepithelialization created a closed tunnel where the pins were isolated from the subdermal tissue, however the reepithelialization was incomplete around the middle of the pin and a low‐grade inflammation increased with decreasing thickness of epidermis. The inflammation consisted of mononuclear cells, mainly lymphocytes. With increasing inflammation the number of neutrophils and macrophages increased. In the lymphoid follicular hyperplasia macrophages and a few neutrophils were found, in one case accompanied by Splendore‐Hoeppli material with radiating eosinophilic clubs and Gram‐positive cocci. Immunohistochemical staining of the cocci for Staphylococcus aureus was positive. The observations from the recaptured cetaceans suggest that the instrumentations caused only temporary and low‐grade inflammatory responses.  相似文献

Bird migration phenology shows strong responses to climate change. Studies of trends and patterns in phenology are typically based on annual summarizing metrics, such as means and quantiles calculated from raw daily count data. However, with irregularly sampled data and large day‐to‐day variation, such metrics can be biased and noisy, and may be analysed using phenological functions fitted to the data. Here we use count data of migration passage from a Finnish bird observatory to compare different models for the phenological distributions of spring migration (27 species) and autumn migration (57 species). We assess parsimony and goodness‐of‐fit in a set of models, with phenological functions of different complexity, optionally with covariates accounting for day‐to‐day variability. The covariates describe migration intensities of related species or relative migration intensities the previous day (autocovariates). We found that parametric models are often preferred over the more flexible generalized additive models with constrained degrees of freedom. Models corresponding to a mixture of two distinct passing populations were frequently preferred over simpler ones, but usually no more complex models are needed. Slightly more complex models were favoured in spring compared to autumn. Related species’ migration activity effectively improves the model by accounting for the large day‐to‐day variation. Autocovariates were usually not that relevant, implying that autocorrelation is generally not a major concern if phenology is modelled properly. We suggest that parametric models are relatively good for studying single‐population migration phenology, or a mix of two groups with distinct phenologies, especially if daily variation in migration intensity can be controlled for. Generalized additive models may be useful when the migrating population composition is unknown. Despite these guidelines, choosing an appropriate model involves case‐by‐case assessment or the biological relevance and rationale for modelling phenology.  相似文献
