Three strains ofClostridium butyricum exhibited elevated minimal inhibitory concentrations (MICs) to penicillin (64–1,024 g/ml), ampicillin (32–256 g/ml), carbenicillin (128–1,024 g/ml), and oxacillin (32–64 g/ml). Cephalosporin/cephamycin agents were more active than penicillin drugs. All isolates were found to possess a -lactamase. The -lactamases were primarily cell associated during the logarithmic phase of growth. Stationary-phase cells released most of the enzyme into the culture medium. Cephalothin supplementation of broth cultures with concentrations equivalent to one-eighth of the MIC significantly increased the quantity of -lactamase synthesized. The -lactamases produced by these three isolates exhibited greatest activity with penicillin followed by ampicillin>cephaloridine>carbenicillin and oxacillin. No enzymatic activity was observed using cephalothin, cephalexin, cefazolin, cefoxitin, or cephamandole substrates. The -lactamases were inhibited by clavulanic acid and para-chloromurcuribenzoate and not inhibited by cloxacillin. Each enzyme exhibited an isoelectric point of 4.2. 相似文献

Summary Treatment of human lymphocytes in vitro with trimethylpsoralen or 8-methoxypsoralen and UVA irradiation (PUVA) induced chromosome damage, mainly constrictions and gaps, but also breaks and exchanges, and increased the frequency of sister chromatid exchange (SCE). The localization of the chromosome aberrations was nonrandom. The coincidence of many PUVA hits with mercaptoenthanol hits suggests that PUVA may have other targets in the cell than the DNA, perhaps the folding proteins of the chromosomes and the nuclear membrane/chromatin attachment organelles.Caffeine increased in a synergistic way the chromosome aberration yield if added after PUVA treatment, but there was no effect when caffeine was present before and during PUVA treatment. Abstract— Cortical monoamine changes during ischemic episodes of varied duration and their sequence of changes following cerebral reperfusion were studied in the gerbil. Forty-one percent of 280 animals exhibited signs of cerebral hemispheric ischemia (stroke) after unilateral common carotid artery occlusion. Norepinephrine (NE) levels decreased after 60 min in the occluded hemisphere of stroked animals but dopamine (DA) levels were unaltered. S-Hydroxytryptamine (5-HT) levels became bilaterally reduced in both stroked and non-stroked animals as soon as S min after occlusion. Upon reperfusion after periods of 30 or 60 min of occlusion there was a bilateral rebound increase of cortical NE and DA levels to well above control values in stroked and non-stroked animals. 5-HT levels remained reduced in both groups. Results suggest disorder of monoamine metabolism in ischemic brain which persists during the early reperfusion period, perhaps contributing to deficits in neurological function. Monoamine changes in contralateral non-ischemic hemispheres both during the occlusion and reperfusion periods are thought further evidence of diaschisis. 相似文献

Corn yields were measured after application of nematicides in 16 experiments, mostly in medium-to-heavily textured soil, at 12 locations in Iowa during 1973-1976. The average maximum yield increase in plots treated with nematicides was 21% over yields in untreated plots. Yields were correlated negatively with nematode numbers or nematode biomass in nearly all comparisons. Correlations of nematode numbers in the soil with yield averaged -0.56 for Helicotylenchus pseudorobustus, -0.45 for Hoplolaimus galeatus, -0.51 for Pratylenchus spp., and -0.64 for Xiphinema americanum. Correlation coefficients for numbers of nematodes in the roots and yield averaged -0.63 for Pratylenchus spp. and -0.56 H. galeatus. Correlation coefficients for yield and total number of nematodes averaged -0.65 in roots and -0.55 in soils. Negative correlations also were greater for comparisons of yield with total parasitic-nematode biomass than with numbers of individual nematodes of a species or total numbers of parasitic nematodes. 相似文献

Summary Cells of embryos carrying a lethal nucleolar mutation have been maintained in vitro for extended periods of time. Normally
these mutants live only 9 to 12 days after fertilization but their cells in culture will survive for more than 3 months. The
extent of ribosomal RNA (rRNA) synthesis was determined in primary cultures prepared from normal embryos and nucleolar mutants
having different numbers of ribosomal RNA genes. We found that the accumulation of radioactivity into rRNA for normal and
mutant embryos was similar in vivo and in vitro. In primary cultures of normal embryos which have two nucleoli per cell and
mutant embryos which have only one nucleolus per cell, the incorporation of radio-activity into rRNA was similar even though
the normal cells have twice as many rRNA genes. Thus the mechanism which regulates dosage compensation of the rRNA genes operates
both in vivo and in vitro.

Sixteen growing pigs were fed a vitamin E and selenium deficient diet; half of the animals (Group 2) were given a daily supply of vitamin E and selenium. After having been fed these diets for 53 days, the pigs were infected orally with minced colonic material from cases with typical swine dysentery. This exposure resulted in outbreaks of swine dysentery in both groups. The incubation times were, however, distinctly shorter and the clinical symptoms much more pronounced in Group 1 than in Group 2. The patho^morphological lesions in the colon also differed between the 2 groups. In the pigs of Group 1 evident pseudomembraneous lesions were observed in the spiral colon. In Group 2, the colonic alterations consisted predominantly of a catarrhal enteritis; pseudomembranes occurred in a minor part of colon in only 4 pigs. Both the clinical and the chemical observations and the pathological findings indicated a much better vitamin E and selenium balance in the pigs of Group 2. It is concluded that the treatment with vitamin E and selenium in Group 2 greatly increased resistance to swine dysentery. 相似文献

Ipomoea pandurata rarely produces seed. Sporophytic self-incompatibility is a secondary factor contributing to the reduced seed production because the principal pollinators, Bombus pennsylvanicus (Apidae) and Melitoma taurea (Anthophoridae), effect cross-pollination in most populations. Sterility, expressed as pollen tube failure in the style and as a sterility mechanism in the ovary, is the primary factor accounting for reduced seed production. In mammalian cells, two of the so-called heat shock (hsp) or stress proteins are components of the mitochondria. One of these, hsp 58, is a member of the bacterial GroEL family, whereas the other, glucose-regulated protein (grp) 75, represents a member of the hsp 70 family of stress proteins. Owing to previous studies implicating a role for both the hsp 70 and GroEL families in facilitating protein maturation events, we used the method of native immunoprecipitation to examine whether hsp 58 and grp 75 might interact with other proteins of the mitochondria. In cells pulse-labeled with [35S]-methionine, a significant number of newly synthesized mitochondrial proteins co-precipitated with either hsp 58 or grp 75. Such interactions appeared transient. For example, providing the pulse-labeled cells a subsequent chase period in the absence of radiolabel resulted in a reduction of co-precipitating proteins. If the pulse-chase labeling experiments were performed in the presence of an amino acid analogue, somewhat different results were obtained. Specifically, although many of the newly synthesized and analogue-containing proteins again were observed to co-precipitate with grp 75, the interactions did not appear transient, but instead were stable. Under steady-state labeling conditions, we also observed a portion of hsp 58 and grp 75 in an apparent complex with one another. On addition of ATP, the complex was dissociated. Accompanying this dissociation was the concomitant autophosphorylation of grp 75. On the basis of these observations, as well as previous studies examining the structure/function of the hsp 70 and GroEL proteins, we suspect that both hsp 58 and grp 75 interact with and facilitate the folding and assembly of proteins as they enter into the mitochondria. 相似文献

Regional variation in fish predation intensity: a historical perspective in the Gulf of Maine 总被引:3,自引:0,他引:3
Summary Regional variation in the intensity of fish predation on tethered brittle stars and crabs was measured at 30–33 m depths in the rocky subtidal zone at seven sites representing coastal and offshore regions of the Gulf of Maine, USA. Analysis of covariance comparing the slopes of brittle star survivorship curves followed by multiple comparisons tests revealed five groupings of sites, with significantly greater predation rates in the two offshore than in the three coastal groups. Brittle stars tethered at the three offshore sites were consumed primarily by cod, Gadus morhua, with 60–100% prey mortality occuring in 2.5 h. In striking contrast, only 6–28% of brittle star prey was consumed in the same amount of time at the four coastal sites, which were dominated by cunner, Tautogolabrus adspersus. In several coastal trials, a majority of brittle star prey remained after 24 h. The pattern of higher predation offshore held for rock crabs as well with only 2.7% of tethered crabs consumed (n=36) at coastal sites versus 57.8% of crabs (n=64) consumed at offshore sites. Another important predatory fish, the wolffish, Anarhichas lupus, consumed more tethered crabs than brittle stars. Videos and time-lapse movies indicated that cod and wolffish were significantly more abundant at offshore than at coastal sites. Three hundred years of fishing pressure in New England has severely depleted stocks of at least one important benthic predator, the cod, in coastal waters. We speculate that this human-induced predator removal has lowered predation pressure on crabs and other large mobile epibenthos in deep coastal communities.
