The distribution pattern of 115 chironomid taxa at 664 sites, distributed over all kinds of water, was analyzed and related to 42 water types, based on the complete macrofauna composition. The distinction of chironomid groups was not useful to recognize water types. The use of individual chironomid taxa was more sensitive, though also too limited to recognize water types clearly. Chironomids are distributed along a continuum which is directed by master factors and detailed by habitat factors.  相似文献

Rapid, transient changes of the membrane potential upon lighttransitions are generally observed in microelectrode studies.In a patch-clamp study similar responses to light transitionswere found in current clamp. Corresponding with the changesof membrane potential, light-induced current changes in voltageclamp were observed. This paper evaluates the involvement ofoutward rectifying conductances and plasma membrane bound H+-ATpases(proton pump) to these light responses in mesophyll protoplastsof Nicotiana tabacum L. The contribution of K+-channels to theseresponses, could be minimized by variation of the holding potentialor addition of the K+-channel blocker verapamil. It was concludedthat light transitions modulate both proton pump and K+-channelactivity. Effects of light on membrane current were not observedin root cells and chlorophyll-deficient cells, suggesting thatthe response requires photosynthetic activity. Monoclonal antibody PNK-E binds to approximately 15% of porcine peripheral blood lymphocytes (PBL) which are PT4 negative and PT8 positive. When cells from tissues of adult pigs are treated with PNK-E, enhancement of natural killer (NK) cell activity is observed from PBL and spleen cells, and a dramatic induction of NK activity is observed from bone marrow cells. With cells derived from tissues of neonatal piglets, PNK-E induces NK activity from PBL and bone marrow cells. To investigate the mechanism of PNK-E-mediated enhancement of NK, proliferation assays, calcium-pulse assays, single-cell assays, and kinetic analyses were performed. PNK-E did not induce proliferation of PBL. PNK-E could be added as late as 30 min prior to termination of Ca(2+)-pulse assays and still enhance NK activity. Using kinetic analysis PNK-E was found to increase the rate of NK lysis (Vmax) and rate of lytic programming per NK cell (k2). In addition, results from single-cell assays indicate that PNK-E activates a population of normally inactive effector cells. These results indicate that PNK-E enhances the lytic capacity of mature NK cells and induces a population of nonlytic cells to become highly cytolytic cells. Furthermore, the enhancing effects are immediate and do not require an induction period. Thus, PNK-E recognizes and activates a unique triggering molecule that is present on NK cells.  相似文献

Chromosome preparations from Chinese hamster lymphocytes were made by culturing for 2 to 6 days 50,000 to 800,000 lymphocytes in flat-bottomed Cooke microtiter plates in 0.1 or 0.2 ml Roswell Park Memorial Institute-1640 medium (RPMI-1640) supplemented with 5--40 microliters PHA/ml, 20% fetal calf serum (FCS), and 40 microns 2-mercaptoethanol. Depending on blood volume and cell concentrations used, it was possible to obtain up to 1400 well-spread metaphases from one venepuncture.  相似文献

Dormant and nondormant isogenic barley grains were obtained by maturing grains under short day (SD) or long day (LD) growth conditions, respectively. Hormonal responses of isolated embryos and aleurone layers from these grains were studied. Addition of abscisic acid (ABA) reduced germination rate and percentage of embryos, and induced Rab (ABA-responsive) mRNA in aleurone layers from both types of grain. Embryos and aleurone layers from dormant grains responded stronger to ABA than those from nondormant grains. Gibberellic acid (GA3) increased the germination rate and percentage of embryos from dormant grains and counteracted the ABA-induced inhibition of embryo germination. GA3 did not affect the amount of Rab mRNA in aleurone layers, suggesting that expression of the Rab gene has no direct correlation with germination. The stronger response of embryos and aleurone layers from dormant grains to ABA may not be explained by higher endogenous ABA levels, but might be due to differences in hormone signal transduction. Aleurone protoplasts from dormant grains had a higher cytosolic pH than those from nondormant grains. To inhibit the ABA-induced Rab mRNA, a much higher concentration of weak acid was required for aleurone layers from dormant grains than for those from nondormant grains. A possible difference in ABA signal transduction between dormant and nondormant grains is discussed.  相似文献

Sodium ion batteries (NIBs) are one of the versatile technologies for low‐cost rechargeable batteries. O3‐type layered sodium transition metal oxides (NaMO2, M = transition metal ions) are one of the most promising positive electrode materials considering their capacity. However, the use of O3 phases is limited due to their low redox voltage and associated multiple phase transitions which are detrimental for long cycling. Herein, a simple strategy is proposed to successfully combat these issues. It consists of the introduction of a larger, nontransition metal ion Sn4+ in NaMO2 to prepare a series of NaNi0.5Mn0.5? y Sn y O2 (y = 0–0.5) compositions with attractive electrochemical performances, namely for y = 0.5, which shows a single‐phase transition from O3 ? P3 at the very end of the oxidation process. Na‐ion NaNi0.5Sn0.5O2/C coin cells are shown to deliver an average cell voltage of 3.1 V with an excellent capacity retention as compared to an average stepwise voltage of ≈2.8 V and limited capacity retention for the pure NaNi0.5Mn0.5O2 phase. This study potentially shows the way to manipulate the O3 NaMO2 for facilitating their practical use in NIBs.  相似文献

This study uses molecular and cytogenetic methods to determine the origin of a B chromosome in some males of the wasp Trichogramma kaykai. This so-called paternal sex ratio (PSR) chromosome transmits only through sperm and shortly after fertilization triggers degeneration of the paternal genome, while keeping itself intact. The resulting embryos develop into haploid B-chromosome-carrying males. Another PSR chromosome with a very similar mode of action is found in the distantly related wasp Nasonia vitripennis and its origin was traced by transposon similarity to the genus Trichomalopsis, which is closely related to Nasonia. To determine whether both PSR chromosomes have a similar origin we aimed to reveal the origin of the Trichogramma PSR chromosome. Using fluorescent in situ hybridization, we discovered a major satellite repeat on the PSR chromosome, the 45S ribosomal DNA. Analysis of the internal transcribed spacer 2 (ITS2) of this repeat showed the presence of multiple ITS2 sequences on the PSR chromosome resembling either the ITS2 of T. oleae or of T. kaykai. We therefore conclude that the Trichogramma PSR chromosome originates from T. oleae or a T. oleae-like species. Our results are consistent with different origins for the PSR chromosomes in Trichogramma and Nasonia.  相似文献
