Mutational analysis of a ras catalytic domain.   总被引:48,自引:23,他引:25       下载免费PDF全文
We used linker insertion-deletion mutagenesis to study the catalytic domain of the Harvey murine sarcoma virus v-rasH transforming protein, which is closely related to the cellular rasH protein. The mutants displayed a wide range of in vitro biological activity, from those that induced focal transformation of NIH 3T3 cells with approximately the same efficiency as the wild-type v-rasH gene to those that failed to induce any detectable morphologic changes. Correlation of transforming activity with the location of the mutations enabled us to identify three nonoverlapping segments within the catalytic domain that were dispensable for transformation and six other segments that were required for transformation. Segments that were necessary for guanosine nucleotide (GDP) binding corresponded to three of the segments that were essential for transformation; two of the three segments share strong sequence homology with other purine nucleotide-binding proteins. Loss of GDP binding was associated with apparent instability of the protein. Lesions in two of the three other required regions significantly reduced GDP binding, while small lesions in the last required region did not impair GDP binding or membrane localization. We speculate that this latter region interacts with the putative cellular target of ras. The results suggest that transforming ras proteins require membrane localization, guanosine nucleotide binding, and an additional undefined function that may represent interaction with their target.  相似文献

ABSTRACT: BACKGROUND: Hip fractures are common among frail elderly persons and often have serious consequences on function, mobility and mortality. Traditional treatment of these patients is performed in orthopedic departments without additional geriatric assessment. However, studies have shown that interdisciplinary geriatric treatment may be beneficial compared to traditional treatment. The aim of the present study is to investigate whether treatment of these patients in a Department of Geriatrics (DG) during the entire hospital stay gives additional benefits as compared to conventional treatment in a Department of Orthopaedic Surgery (DOS). FINDINGS: A new clinical pathway for in-hospital treatment of hip fracture patients was developed. In this pathway patients were treated pre-and postoperatively in DG. Comprehensive geriatric assessment was performed as an interdisciplinary, multidimensional, systematic assessment of all patients focusing on each patient's capabilities and limitations as recommended in guidelines and systematic reviews. Identification and treatment of co-morbidities, pain relief, hydration, oxygenation, nutrition, elimination, prevention and management of delirium, assessment of falls and osteoporosis were emphasized. Discharge planning started as early as possible. Initiation of rehabilitation with focus on early mobilisation and development of individual plans was initiated in hospital and continued after discharge from hospital. Fracture specific treatment was based upon standard treatment for the hospital, expert opinions and a review of the literature. CONCLUSION: A new treatment program for old hip fracture patients was developed, introduced and run in the DG, the potential benefits of which being compared with traditional care of hip fracture patients in the DOS in a randomised clinical trial.  相似文献

Two populations of Bosmina longispina existing in two similarand closely situated Norwegian mountain lakes were investigatedwith respect to morphological variations. The two lakes areinhabited by different predators, Lake Gopollen is dominatedby whitefish and Lake Djupen by a sparse population of browntrout and invertebrate predators. The B. longispina in LakeDjupen were both larger and also longer at maturation than theB. longispina of Lake Gopollen. In Lake Djupen the mucronesof B. longispina were twice as long and their relative antennulalength was also larger than in Lake Gopollen. However, the relativeeye size did not differ between the two populations. The observationsfit the present hypothesis on morphological predator avoidanceadaptations in B. longispina, fairly well. Invertebrate predatorsfavour large B. longispina with long protuberances, body sizeand length of mucro being the most important features.  相似文献

Reflection spectra obtained from hyperspectral imaging can be used as a bio-optical taxonomic identification tool if the pigment composition and the corresponding optical absorption signatures of an organism are known. In this study we elucidate species-specific absorption and corresponding reflection signatures of marine organisms and discuss optical fingerprints from underwater hyperspectral imaging (UHI) for future automated identification of organisms on the seafloor. When mounted on underwater robots, UHI has the potential to be a time- and cost-efficient identification and mapping method covering large areas over a short time. Hyperspectral imaging in vivo and in situ were used to obtain species-specific reflection signatures (optical fingerprints). High performance liquid chromatography, liquid chromatography–mass spectroscopy and nuclear magnetic resonance were used for pigment identification and to obtain species-specific absorption signatures of four marine benthic species; the spoonworm Bonellia viridis, and the sponges Isodictya palmata, Hymedesmia paupertas and Hymedesima sp. Species-specific optical fingerprints based on a UHI-based reflectance signature were verified successfully in the organisms investigated.  相似文献

Bathycoccus prasinos Eikrem et Throndsen exhibited a complex carotenoid distribution pattern including the carotenes β,β-carotene (0.8% of total carotenoids) and β, ° Carotene (0.4%) and several xanthophylls. These were prasinoxanthin (49% of total carotenoids), micromonal (16%), neoxanthin (14%), uriolide (7%), violaxanthin (0.8%), 31-dehydrouriolide (0.8%), dihydrolutein (0.1%), two partly characterized esterified carotenols (together 10%), and five minor unidentified carotenols (together 2%). The identifications were based on high-performance liquid chromatography (HPLC), thin-layer chromatography (TLC), visible spectroscopy (VIS), and mass spectra (MS) and in part on 1H nuclear magnetic resonance (NMR), circular dichroism (CD), and chemical derivatization. The carotenoid composition of B. prasinos was related to that of other prasinoxanthin / uriolide / micromonal-producing prasinophytes (Mantoniella squamata, Micromonas pusilla, and Pseudoscourfieldia marina). The relative distribution of chlorophylls (w/w) were chlorophyll a (chl a; 63%), chl b (31%), and an unknown chl c-like chlorophyll (7%) with spectral characteristics similar to magnesium 2,4-divinylphaeoporphyrin a, monomethyl ester, compatible with other prasinophytes. The chemosystematic data and ultrastructural characteristics for the order Mamiellales are discussed. We conclude that HPLC studies alone are insufficient for the identification and characterization of the carotenoids, including the minor carotenoids essential for biosynthetic/chemosystematic considerations.  相似文献

The epifauna associated with two of the most common species of kelp in the Arctic, Laminaria digitata (Hudson) Lamouroux, 1813 and Saccharina latissima (Linnaeus) Lane, , Mayes, Druehl and Saunders 2006 [synonym: L. saccharina (Linnaeus) Lamouroux, 1813] were examined in Kongsfjorden, Svalbard. The aim of this study was to test whether species richness of epifauna was influenced by seasonality of recruitment supply (spring and summer) or by age of the substrate (lamina). From 20 algae specimens (ten—L. digitata, ten—S. latissima), a total of 27 epifaunal taxa were identified with bryozoans as the most species-rich group. Total species richness ranged from one to nine species per lamina. No significant difference in species richness was observed between the two kelp species. In both macroalgae, more epifauna taxa were registered on lamina in May (mean 6) than in August (mean 3). This indicates that the time chosen for sampling had a significant impact on the species richness pattern. The number of epifaunal species was significantly negatively correlated with increasing age of the lamina. There are indications that diversity of the epifauna on lamina is influenced to a large extent by substrate (lamina) surface area, environmental stress and the presence of meroplankton in the water column.  相似文献

Subtypes of human immunodeficiency virus type 1 circulating in 21 north Indian patients were characterized based on the partial sequence of the gp120 envelope protein. A majority of viruses (85.7%, 18/21) were subtype C, while 14.3% (3/21) were subtype A. Sequence analysis revealed that the V3 region was highly conserved compared with V4 and V5. The predicted use of co-receptors indicated exclusive usage of R5, except for two subtype A viruses (AIIMS279 and AIIMS281). Our results demonstrate conservation within the V3 loop of subtype C viruses, and suggest the emergence of non-clade C viruses in the north Indian population.  相似文献
