A Tam3 two-element system has been designed by combining an immobilized Tam3 element with a non-autonomous dTam3 element inserted into the HPT gene. The phenotypic assay employed, restored hygromycin resistance, indicated thattrans-activation of the non-autonomous dTam3 element occurred. Molecular analyses of the excision sites revealed that the ends of the dTam3 element remain in the empty donor sites. The predominant consequence of this type of excision appears to be that excised fragments fail to re-integrate into the tobacco genome. Only one case of dTam3 re-integration could be detected. The ends of this element had been degraded upon integration into the tobacco genome. Either the altered structure of the Tam3 derivatives or tobacco host factors are influencing thetrans-activation of a dTam3 element, resulting in aberrant excision.  相似文献

Four species of riparian vegetation (alder, birch, willow and poplar) were fertilized with nitrogen, phosphorus, nitrogen + phosphorus, or no fertilizer (control). The resulting leaf detritus (leached but not microbially colonized) was offered to a stream shredder, Hydatophylax variabilis (Trichoptera: Limnephilidae). In one experiment, shredder consumption of leaf detritus from different nutrient treatments (within tree species) was compared, and in a second experiment, consumption of different tree species (within nutrient treatments) was compared. Larvae preferred leaf detritus from nitrogen + phosphorus treatments (except in poplar where nitrogen treatment was preferred). Alder was preferred over other tree species for all treatments. Chemical and physical analyses of leaf litter showed differences between tree species and nutrient treatments in nutrient content, tannins and leaf toughness. Leaf consumption by larvae was positively associated with nitrogen content and negatively associated with condensed tannin content. Species composition and nutrient status of riparian vegetation may strongly influence detrital food webs in streams.  相似文献

We have constructed a genealogy of strain S288C, from which many of the mutant and segregant strains currently used in studies on the genetics and molecular biology of Saccharomyces cerevisiae have been derived. We have determined that its six progenitor strains were EM93, EM126, NRRL YB-210 and the three baking strains Yeast Foam, FLD and LK. We have estimated that approximately 88% of the gene pool of S288C is contributed by strain EM93. The principal ancestral genotypes were those of segregant strains EM93-1C and EM93-3B, initially distributed by C. C. Lindegren to several laboratories. We have analyzed an isolate of a lyophilized culture of strain EM93 and determined its genotype as MATa/MATα SUC2/SUC2 GAL2/gal2 MAL/MAL mel/mel CUP1/cup1 FLO1/flo1. Strain EM93 is therefore the probable origin of genes SUC2, gal2, CUP1 and flo1 of S288C. We give details of the current availability of several of the progenitor strains and propose that this genealogy should be of assistance in elucidating the origins of several types of genetic and molecular heterogeneities in Saccharomyces.  相似文献

Summary We have mapped and sequenced the globin gene and seven surrounding Alu repeat sequences in the orangutan globin gene cluster and have compared these and other orangutan sequences to orthologously related human sequences. Noncoding flanking and intron sequences, synonymous sites of , , and globin coding regions, and Alu sequences in human and orangutan diverge by 3.2%, 2.7%, and 3.7%, respectively. These values compare to 3.6% from DNA hybridizations and 3.4% from the globin gene region. If as suggested by fossil evidence and molecular clock calculations, human and orangutan lineages diverged about 10–15 MYA, the rate of noncoding DNA evolution in the two species is 1.0–1.5×10–9 substitutions per site per year. We found no evidence for either the addition or deletion of Alu sequences from the globin gene cluster nor is there any evidence for recent concerted evolution among the Alu sequences examined. Both phylogenetic and phenetic distance analyses suggest that Alu sequences within the and globin gene clusters arose close to the time of simian and prosimian primate divergence (about 50–60 MYA). We conclude that Alu sequences have been evolving at the rate typical of noncoding DNA for the majority of primate history.Presented at the FEBS Symposium on Genome Organization and Evolution, held in Crete, Greece, September 1–5, 1986  相似文献

Specific cell surface insulin binding to embryonic chick neural retina cells has been demonstrated in vivo. Kinetics of insulin binding as well as hormonal specificity were similar to those reported for other vertebrate cells and tissues, both neural and nonneural. When surface insulin binding to retinal cells was studied as a function of embryonic age, a developmental relationship was observed. Scatchard analysis revealed that the number of cell surface insulin receptors decreased approximately 75% between days 10 and 16 of embryonic development. Plasma levels of growth hormone (GH), free fatty acids (FFA) and glucose were measured in vagotomized (VgX) and sham-operated (VgS) control pigeons. In VgX pigeons, GH level was significantly lower whereas FFA and glucose levels were higher than in VgS pigeons. The depression in GH level in VgX pigeons has been attributed to the significantly high levels of norepinephrine (NE) and corticosterone in these Birds. The higher plasma FFA concentration in VgX pigeons was therefore due to adipokinetic hormonal action other than of GH. It has been suggested that the adrenocorticotrophic hormone (ACTH) and/or NE could have produced the increase in plasma FFA in VgX pigeons. The pronounced hyperglycemia seen in VgX pigeons has been attributed to catecholamine action in the absence of the vagal tone.  相似文献

Summary It has been proposed that within rather broad habitat types the distribution and abundance of bird species may be more closely associated with plant taxonomic composition than with the structure and configuration of the vegetation. Birds from a sample of eight representative grassland habitats in middle and western North America are consistent with this hypothesis. Over half (55%) of the variation in bird community composition was associated with floristic variation, but only a third (35%) was associated with physiognomy. Separating the interacting effects of floristics and physiognomy from each other served to accentuate the difference between them with respect to the avifauna. It is postulated that bird species/plant taxa associations, especially within similar habitat types, are mediated by the specific food resources that different plant taxa provide. Summary indices such as diversity measures obscure the taxonomic information content of plant or animal assemblages, and the use of such indices has likely impeded detection of the relationships described here.  相似文献

Summary We developed models of inter-individual interference to predict the fecundity of individuals in populations of the annual plant species, Arabidopsis thaliana. An individual plant is modeled as having a neighborhood which is a circular area of fixed radius with the plant at its center. Other plants which share the circle with the focal plant are termed neighbors of the focal plant. We developed an index of neighborhood interference which is the independent variable in a non-linear regression model that predicts individual plant fecundity. We present methods of exploratory data analysis that are useful in determining a best neighborhood radius, defined as that radius which minimizes residual sum of squares, and in deciding on the functional form of the interference index. In developing the interference index for Arabidopsis, we focus on aspects of the spatial distribution of neighbors: their number, distance and angular dispersion.We found that a best (or optimal) neighborhood radius can be resolved, which provides the best predictor of plant performance. Fecundity predictors based on adult neighbors were noticeably better than those based on neighbors at the seedling stage. Rosettes of Arabidopsis may change location during development (they fall over) and the new fallen positions do provide some improvement in the predictor. Taking into account distance to neighbors within the neighborhood provided only negligible improvement in the model. Finally, the incorporation of angular dispersion in the crowding index produced a considerably better fit. The fecundity predictor that included number of neighbors and angular dispersion in the crowding index explained about 70% of the variation in individual seed set.  相似文献
