Background  

Omptins are a family of outer membrane proteases that have spread by horizontal gene transfer in Gram-negative bacteria that infect vertebrates or plants. Despite structural similarity, the molecular functions of omptins differ in a manner that reflects the life style of their host bacteria. To simulate the molecular adaptation of omptins, we applied site-specific mutagenesis to make Epo of the plant pathogenic Erwinia pyrifoliae exhibit virulence-associated functions of its close homolog, the plasminogen activator Pla of Yersinia pestis. We addressed three virulence-associated functions exhibited by Pla, i.e., proteolytic activation of plasminogen, proteolytic degradation of serine protease inhibitors, and invasion into human cells.  相似文献

Background  

A controversial topic in evolutionary developmental biology is whether morphological diversification in natural populations can be driven by expansions and contractions of amino acid repeats in proteins. To promote adaptation, selection on protein length variation must overcome deleterious effects of multiple correlated traits (pleiotropy). Thus far, systems that demonstrate this capacity include only ancient or artificial morphological diversifications. The Hawaiian Islands, with their linear geological sequence, present a unique environment to study recent, natural radiations. We have focused our research on the Hawaiian endemic mints (Lamiaceae), a large and diverse lineage with paradoxically low genetic variation, in order to test whether a direct relationship between coding-sequence repeat diversity and morphological change can be observed in an actively evolving system.  相似文献

Transmission of parasites and diseases may be one of the mechanisms for the displacement of native and endemic rodents of Madagascar (subfamily Nesomyinae) by the introduced Rattus rattus (subfamily Murinae). We studied the occurrence of trypomastigotes in rodents at several rainforest sites on the island. Examination of blood smears showed Trypanosoma lewisi-like trypomastigotes in 11.5% of the R. rattus (n = 52). Trypomastigotes differing in morphology from those of T. lewisi were detected in 4% of the endemic rodent Nesomys rufus (n = 23). In contrast to the relatively heavy infections found in R. rattus, only a few trypomastigotes were found in the infected N. rufus. Trypomastigotes were not found in other nesomyine rodents including Eliurus minor (n = 18), E. tanala (n = 15), E. grandidieri (n = 12), E. majori (n = 9) or E. webbi (n = 9). Of potential vectors of trypomastigotes, six endemic species of fleas were identified from the rodents.  相似文献

Super-enhancers are clusters of enhancers associated with cell lineage. They can be powerful gene-regulators and may be useful in cell-type specific viral-vector development. Here, we have screened for endothelial super-enhancers and identified an enhancer from within a cluster that conferred 5–70-fold increase in transgene expression. Importantly, CRISPR/Cas9 deletion of enhancers demonstrated regulation of ADAMTS18, corresponding to evidence of chromatin contacts between these genomic regions. Cell division-related pathways were primarily affected by the enhancer deletions, which correlated with significant reduction in cell proliferation. Furthermore, we observed changes in angiogenesis-related genes consistent with the endothelial specificity of this SE. Indeed, deletion of the enhancers affected tube formation, resulting in reduced or shortened sprouts. The super-enhancer angiogenic role is at least partly due to its regulation of ADAMTS18, as siRNA knockdown of ADAMTS18 resulted in significantly shortened endothelial sprouts. Hence, functional characterization of a novel endothelial super-enhancer has revealed substantial downstream effects from single enhancer deletions and led to the discovery of the cis-target gene ADAMTS18 and its role in endothelial function.  相似文献

We investigated the factors mediating selection acting on two MHC class II genes (DQA and DRB) in water vole (Arvicola scherman) natural populations in the French Jura Mountains. Population genetics showed significant homogeneity in allelic frequencies at the DQA1 locus as opposed to neutral markers (nine microsatellites), indicating balancing selection acting on this gene. Moreover, almost exhaustive screening for parasites, including gastrointestinal helminths, brain coccidia and antibodies against viruses responsible for zoonoses, was carried out. We applied a co-inertia approach to the genetic and parasitological data sets to avoid statistical problems related to multiple testing. Two alleles, Arte-DRB-11 and Arte-DRB-15, displayed antagonistic associations with the nematode Trichuris arvicolae, revealing the potential parasite-mediated selection acting on DRB locus. Selection mechanisms acting on the two MHC class II genes thus appeared different. Moreover, overdominance as balancing selection mechanism was showed highly unlikely in this system.  相似文献

We examined the temporal variation in capture rates of shrewsNotiosorex crawfordi (Coues, 1877) and Sorex ornatus (Merriam, 1895) in 20 sites representing fragmented and continuous habitats in southern California, USA. InN. crawfordi, the temporal variation was significantly correlated with the mean capture rates. Of the 6 landscape variables analyzed (size of the landscape, size of the sample area, altitude, edge, longitude and latitude), sample area was positively correlated with variation in capture rates ofN. crawfordi. InS. ornatus, longitude was negatively correlated with variation in capture rates. Analysis of the effect of precipitation on the short- and long-term capture rates at 2 of the sites showed no correlation between rainfall and capture rates of shrews even though peak number of shrews at both sites were reached during the year of highest amount of rainfall. A key problem confounding capture rates of shrews in southern California is the low overall abundance of both shrew species in all habitats and seasons.  相似文献
