A comparative litter fall study was made in five rain forest stands along a gradient of humus form development and soils in the Amazon lowlands of eastern Colombia. The total fine litter fall was highest in a plot on a well drained soil of the flood plain of the Caquetá River (1.07 kg · m-2 · y-1), lower in three plots on well drained upland soils (0.86, 0.69, and 0.68 kg · m-2 · y-1), and lowest in a plot on a poorly drained, upland podzolised soil (0.62 kg · m-2 · y-1). In the four upland plots, leaf litter fall patterns were highly associated, which points at climatic regulation. Litter resource quality, as represented by nutrient concentrations and area/weight ratio of the leaf litter fall, was comparatively high in the flood plain plot. In the upland plots, concentrations and fluxes of Ca, Mg, K, and P were as low as in oligotrophic central Amazonian upland forests. This questions generalisations that the western peripheral region of the Amazon basin should be less oligotrophic than central Amazonia. The upland plot on the podzolised soil showed the lowest concentrations and fluxes of N. Mean residence times of organic matter and nutrients in the L horizons hardly differed between the five plots, suggesting that edaphic properties and litter resource quality are of little importance in the first step of decomposition. Mean residence time of organic matter in all ectorganic horizons combined (estimated on the basis of litter input and necromass on the forest floor, and uncorrected for dead fine root input) varied from 1.0 y in the flood plain forest, 1.1–3.3 y in the well drained upland forests, and 10.2 y in the forest on the podzolised soil. 相似文献

Exponentially growing cultures of Nannochloropsis sp. were transferredfrom high light (650 µ.mol quanta m2 s1)to low light (30 (µrnol quanta m2 s1). Thekinetics of changes in the various parameters of the photoacclimationprocess were followed until steady state was reached. In thisorganism, the observed 4.25 increase in cellular chlorophylla in the low-light-acclimated cells was linearly correlatedwith an increase in the number of photosynthetic units (PSU),while the PSU size remained constant. The new steady state wasreached after a transition period of 87 h and following an initialovershoot in the chlorophyll a concentration. During the processof photoacclimation, harvested and utilized light was increaseddue to a combination of increased cellular chlorophyll and anincrease of the quantum yield. These improvements in efficiencywere coupled with the reduction of respiration. The overalleffect of photoacclimation, therefore, amounted to reducingthe impact of low light on the growth rate of the algae. Ourstudy demonstrates the importance of the photoacclimation processin the context of the planktonic way of life. An index of the'effectiveness of photoacclimation' is proposed. 相似文献

Greenhouse and growth chamber studies were established to determine if there are pathological and physiological differences among Meloidogyne hapla populations from California (CA), Nevada (NV), Utah (UT), and Wyoming (WY) on alfalfa cultivars classified as resistant or susceptible to root-knot nematodes. In the greenhouse, plant survival was not consistent with resistance classifications. While all highly resistant Nevada Synthetic germplasm (Nev Syn XX) plants survived inoculation with all nematode populations, two cultivars classified as moderately resistant (''Chief'' and ''Kingstar'') survived (P ≤ 0.05) inoculation with M. hapla populations better than did ''Lobo'' cultivar, which is classified as resistant. Plant growth of Nev Syn XX was suppressed by only the CA population, whereas growth of the other alfalfa cultivars classified as M. hapla resistant or moderately resistant was suppressed by all nematode populations. Excluding Nev Syn XX, all alfalfa cultivars were severely galled and susceptible to all nematode populations. Except for Nev Syn XX, reproduction did not differ among the nematode populations on alfalfa cultivars. Nev Syn XX was not as favorable a host to CA as were the other cultivars; but, it was a good host (reproductive factor [Rf] = 37). Temperature affected plant resistance; the UT and WY populations were more pathogenic at 15-25 C, and CA was more pathogenic at 30 C. Nev Syn XX was susceptible to all nematode populations, except for CA, at only 30 C, and all other alfalfa cultivars were susceptible to all nematode populations at all temperatures. 相似文献

The microbial community structure of twenty-one single-phase and one two-phase full-scale anaerobic sewage sludge digesters was evaluated using oligonucleotide probes complementary to conserved tracts of the 16S rRNAs of phylogenetically defined groups of methanogens and sulfate-reducing bacteria. These probe results were interpreted in combination with results from traditional chemical analyses and metabolic activity assays. It was determined that methanogens in healthy mesophilic, single-phase sewage sludge digesters accounted for approximately 8–12% of the total community and thatMethanosarcinales andMethanomicrobiales constituted the majority of the total methanogen population.Methanobacteriales andMethanococcales played a relatively minor role in the digesters. Phylogenetic groups of mesophilic, Gram-negative sulfate-reducing bacteria were consistently present at significant levels:Desulfovibrio andDesulfobulbus spp. were the dominant sulfate-reducing populations,Desulfobacter andDesulfobacterium spp. were present at lower levels, andDesulfosarcina, Desulfococcus, andDesulfobotulus spp. were absent. Sulfate reduction by one or more of these populations played a significant role in all digesters evaluated in this study. In addition, sulfate-reducing bacteria played a role in favoring methanogenesis by providing their substrates. The analysis of the two-phase digester indicated that true phase separation was not accomplished: significant levels of active methanogens were present in the first phase. Summary As part of our programme directed at the development of enzyme inhibitors based on transition-state mimics, we discovered in the early 1980s that P3-P3 fragments of human fibrinogen A, containing the ketomethylene isostere Arg--[COCH2]Gly at P1-P1, were potent inhibitors of thrombin. Such low-molecular-weight inhibitors are expected to be clinically useful as anticoagultant drugs. In our more recent investigations, the P1-P1 moiety has been replaced with various arginine or lysine ketones. The resulting compounds showed the following order of thrombin inhibitory potency: -ketoesters > fluoroketones >alkoxymethylketones > difluoro--ketoamides >-ketoesters >alkyl ketones. In contrast to all other lysine/arginine pairs studied previously, the inhibitor based on a lysine -ketoester proved superior to the corresponding arginine analogue. A possible explanation for this finding is discussed. All the highly electrophilic ketones (e.g., fluoroketones) were found to exhibit slow-binding kinetics with thrombin, which is likely to be a disadvantage in clinical use. Alkoxymethyl ketones were devoid of such behaviour and have been developed further to yield nanomolar inhibitors of low molecular weight and good selectivity for thrombin. One of these ketones was found to compare favourably with known thrombin inhibitors in anticoagulant assays. Abstract A plasmid-based food-grade vector system was developed for Lactococcus lactis by exploiting the genes for lactose metabolism. L. lactis MGS267 is a plasmid-free strain containing the entire lactose operon as a chromosomal insertion. The lacF gene was deleted from this strain by a double cross-over homologous recombination event. The lacF -deficient strain produced a Lac− phenotype on indicator agar. A cloned copy of the lacF gene expressed on a plasmid was capable of complementing the lacF -deficient strain resulting in a Lac+ phenotype. This stably maintained system fits the requirements of a self-selecting vector system and has the potential to be exploited in the food industry. 相似文献

The orientation of signal–anchor proteins in the endoplasmic reticulum membrane is largely determined by the charged residues flanking the apolar, membrane-spanning domain and is influenced by the folding properties of the NH2-terminal sequence. However, these features are not generally sufficient to ensure a unique topology. The topogenic role of the hydrophobic signal domain was studied in vivo by expressing mutants of the asialoglycoprotein receptor subunit H1 in COS-7 cells. By replacing the 19-residue transmembrane segment of wild-type and mutant H1 by stretches of 7–25 leucine residues, we found that the length and hydrophobicity of the apolar sequence significantly affected protein orientation. Translocation of the NH2 terminus was favored by long, hydrophobic sequences and translocation of the COOH terminus by short ones. The topogenic contributions of the transmembrane domain, the flanking charges, and a hydrophilic NH2-terminal portion were additive. In combination these determinants were sufficient to achieve unique membrane insertion in either orientation. 相似文献
