For free‐swimming marine species like sharks, only population genetics and demographic history analyses can be used to assess population health/status as baseline population numbers are usually unknown. We investigated the population genetics of blacktip reef sharks, Carcharhinus melanopterus; one of the most abundant reef‐associated sharks and the apex predator of many shallow water reefs of the Indian and Pacific Oceans. Our sampling includes 4 widely separated locations in the Indo‐Pacific and 11 islands in French Polynesia with different levels of coastal development. Four‐teen microsatellite loci were analysed for samples from all locations and two mitochondrial DNA fragments, the control region and cytochrome b, were examined for 10 locations. For microsatellites, genetic diversity is higher for the locations in the large open systems of the Red Sea and Australia than for the fragmented habitat of the smaller islands of French Polynesia. Strong significant structure was found for distant locations with FST values as high as ~0.3, and a smaller but still significant structure is found within French Polynesia. Both mitochondrial genes show only a few mutations across the sequences with a dominant shared haplotype in French Polynesia and New Caledonia suggesting a common lineage different to that of East Australia. Demographic history analyses indicate population expansions in the Red Sea and Australia that may coincide with sea level changes after climatic events. Expansions and flat signals are indicated for French Polynesia as well as a significant recent bottleneck for Moorea, the most human‐impacted lagoon of the locations in French Polynesia. 相似文献
Abstract: A cDNA for Drosophila choline acetyltransferase (EC 2.3.1.6; ChAT) was fused with a polyhistidine sequence and expressed in Escherichia coli. The recombinant enzyme was purified to a specific activity of 500 μmol/min/mg of protein using metal affinity chromatography and ion exchange chromatography. Kinetic properties of the recombinant enzyme did not differ significantly from those previously determined. Circular dichroism (CD) spectra revealed that the secondary structure of the enzyme is largely μ-helical. Intrinsic fluorescence spectra of the enzyme indicate that its tryptophan residues are buried. Neither CD nor fluorescence spectra changed significantly in the presence of substrates. The cysteine content of the recombinant Drosophila ChAT was determined to be 16 in the absence and 22 in the presence of 6 M guanidine hydrochloride. Finally, crystallization of recombinant Drosophila ChAT was achieved. 相似文献

Light-dependent conductance changes of voltage-gated Cav1.4 channels regulate neurotransmitter release at photoreceptor ribbon synapses. Mutations in the human CACNA1F gene encoding the α1F subunit of Cav1.4 channels cause an incomplete form of X-linked congenital stationary night blindness (CSNB2). Many CACNA1F mutations are loss-of-function mutations resulting in non-functional Cav1.4 channels, but some mutations alter the channels’ gating properties and, presumably, disturb Ca2+ influx at photoreceptor ribbon synapses. Notably, a CACNA1F mutation (I745T) was identified in a family with an uncommonly severe CSNB2-like phenotype, and, when expressed in a heterologous system, the mutation was shown to shift the voltage-dependence of channel activation, representing a gain-of-function. To gain insight into the pathomechanism that could explain the severity of this disorder, we generated a mouse model with the corresponding mutation in the murine Cacna1f gene (I756T) and compared it with a mouse model carrying a loss-of-function mutation (ΔEx14–17) in a longitudinal study up to eight months of age. In ΔEx14–17 mutants, the b-wave in the electroretinogram was absent, photoreceptor ribbon synapses were abnormal, and Ca2+ responses to depolarization of photoreceptor terminals were undetectable. In contrast, I756T mutants had a reduced scotopic b-wave, some intact rod ribbon synapses, and a strong, though abnormal, Ca2+ response to depolarization. Both mutants showed a progressive photoreceptor loss, but degeneration was more severe and significantly enhanced in the I756T mutants compared to the ΔEx14–17 mutants. 相似文献
Diatoms differ from higher plants by their antenna system, in terms of both polypeptide and pigment contents. A rapid isolation procedure was designed for the membrane-intrinsic light harvesting complexes (LHC) of the diatom Phaeodactylum tricornutum to establish whether different LHC subcomplexes exist, as well to determine an uneven distribution between them of pigments and polypeptides. Two distinct fractions were separated that contain functional oligomeric complexes. The major and more stable complex ( approximately 75% of total polypeptides) carries most of the chlorophyll a, and almost only one type of carotenoid, fucoxanthin. The minor complex, carrying approximately 10-15% of the total antenna chlorophyll and only a little chlorophyll c, is highly enriched in diadinoxanthin, the main xanthophyll cycle carotenoid. The two complexes also differ in their polypeptide composition, suggesting specialized functions within the antenna. The diadinoxanthin-enriched complex could be where the de-epoxidation of diadinoxanthin into diatoxanthin mostly occurs. 相似文献

Meikle are two little known sibling species of theScilla siberica alliance with previously difficult taxonomy, especially with regard to specific delimitation and geographic distribution. Quantitative C-band karyotyping and DNA content determinations were performed in four provenances, including the type localities. Both species (2n = 12) differ strikingly in their banding patterns despite of similar DNA content. Karyotype diversification involved changes in eu- and heterochromatin quantities and resulted in some changes of chromosome form. Despite this, actual phylogenetic relatedness is indicated by a chromosomal marker (NOR-position) of rare occurrence in the alliance. Karyological results and circumstantial evidence from the literature suggest thatS. morrisii is an endemic, exclusive to Cyprus as a member of theS. siberica alliance.S. cilicica seems to be restricted to continental S. The hematophagous mite Dermanyssus gallinae poses a serious sanitary problem in the Brazilian laying poultry industry. Its control is typically performed with acaricides, either in powder or liquid form. However, the intensive use of these products has caused populations of this species to develop tolerance and even resistance. The aim of the present study is to evaluate the response of eggs and adults of D. gallinae to products in aqueous suspension according to commercial indication and as per the recommendations of the Brazilian Ministry of Agriculture, Livestock and Supply. The study used four acaricide products (product 1: cypermethrin, chlorpyrifos, and piperonyl butoxide; product 2: alkyl-benzyl-dimethyl ammonium chloride, glutaraldehyde, deltamethrin; product 3: dichlorvos; product 4: fluralaner) tested in vitro using the contact method. Distilled water was used in the control group. The effectiveness of each of the products differed significantly between eggs and adults. Products 2, 3, and 4 caused 100% of adult mortality up to day 5 after start of treatment, product 1 97.5%. The corrected mortality (non-viability) of eggs was 21.4% (product 1) 39.4% (product 2), 47.8% (product 3), and 14.4% (product 4). Although the products evaluated were effective against adults of D. gallinae, their effectiveness against eggs was lower under the same conditions. This finding might be directly related to frequent D. gallinae reinfestations in poultry houses.
