Ultraturrax and Nagarse released populations of mitochondria isolated from control and day 21 denervated rat soleus muscle were characterized with respect to their oxidative phosphorylation, ADP translocase and ATPase activities. Both Ultraturrax and Nagarse released mitochondrial populations displayed lower capacities for oxidative phosphorylation; lower ADP translocase activities and higher Mg2+ stimulated ATPase activities than their corresponding controls. For both the denervated and control states, the Nagarse-released mitochondrial populations displayed significantly higher respiratory activities than the Ultraturrax released fractions. The significance of these findings is discussed with regard to the process of mitochondrial respiratory control. In addition the role of mitochondrial dysfunction in denervation muscular atrophy is assessed. 相似文献

Abstract: Liver failure and coma are serious complications of Jejunoileal bypass (JIB) in man. Rats underwent either a 90–95% JIB or a sham operation. Six weeks later all animals were sacrificed, and plasma and brain amino acids were determined. In the plasma of rats with JIB compared with sham operation, the concentrations of valine, leucine, isoleucine, lysine, tryptophan, and tyrosine were significantly lower, while in the brain, phenylalanine, tyrosine, histidine, and glutamine were significantly higher. These changes in the brain are similar to those resulting from portalsystemic shunting in the rat. 相似文献

Ultraturrax- and Nagarse-released populations of mitochondria were characterized with respect to their Ca2+-uptake activities (i) by means of the indirect polarographic technique and (ii) directly by the 45Ca Ruthenium Red-quench method of Reed & Bygrave [(1974) Biochem. J. 140, 143-155]. The denervated-muscle subsarcolemmal and intermyofibrillar mitochondrial fractions displayed markedly decreased rates and capacities for Ca2+ uptake compared with their respective controls. Studies on locust femur solid cuticle indicate that there is a continuum of biomechanically important properties from one instar to the next, and that there is a constant ratio of stiffness to mass which is only interrupted at ecdysis. The biochemical nature of both larval and adult solid cuticle is interpreted in terms of both the tangent modulus and the breaking strength of the femur cuticle. A modern system of Fusarium taxonomy is proposed, based on the study over 30 years, of the morphology and growth characteristics of fungi of this genus, and their variability under a wide range of conditions. The system proposed here comprises 13 sections, 33 species and 14 varieties. It differs from systems published by other authors mainly in the sections Eupionnotes, Sporotrichiella, Arthrosporiella, Gibbosum, Discolor, Macroconia and Martiella. 相似文献

Eight isolates of Fusarium of the Sporotrichiella section were critically studied as to their taxonomic position. Most of these isolates had been used for comprehensive toxicological work on mycotoxin effects on animals, chiefly in the United States and Japan. Isolates NRRL 3299 and NRRL 3287, supplied from the United States, were determined to be Fusarium poae. Isolates NRRL 3249, NRRL 5908, 2061-C, and YN-13 from the United States and isolate T-2 from South Africa belonged to Fusarium sporotrichioides. Isolate NRRL 3509 belonged to F. sporotrichioides var. tricinctum. The relevance of recent toxicological work with these isolates on animals to earlier work on alimentary toxic aleukia is discussed in light of the above identifications. 相似文献

A total of 131 isolates of Fusarium poae and F. sporotrichioides from overwintered cereals, which were associated with the alimentary toxic aleukia toxicoses in the Soviet Union, were tested for their ability to produce T-2 toxin [4 beta, 15 diacetoxy-8alpha-(3-methylbutyryloxy)-12,13-epoxytrichothec-9-en 3alpha-ol]. The presence of T-2 toxin was determined by thin-layer chromatography, gas-liquid chromatography, spectroscopic analyses, and the rabbit skin test. A good correlation was demonstrated between T-2 toxin dectetion by thin-layer chromatography and inflammatory skin reactions of rabbits. 相似文献

Although the avian furcula is generally regarded as a dermal bone, this view was challenged by Lansdown (1968 a ), who postulated that it is a cartilage bone. His reinterpretation was predicated upon histochemical data, derived from embryological observations made on the Japanese quail, and is deserving of further investigation as it bears importantly upon the interpretation of the homology of the furcula, and upon aspects of avian phylogeny and the origin of the flight apparatus.
Employing the same investigative methods as Lansdown (1968 a ), together with additional techniques, his interpretations have been reconsidered and no support for his suggestions has been found. In living tissues under inflammatory conditions, superoxide radicals (O(2)*)) are generated and are known to cause oxidative DNA damage. However, the mechanisms of action are poorly understood. It is shown here that the combination of O(2)* with guanine neutral radicals, G(-H)* in single- or double-stranded oligodeoxyribonucleotides (rate constant of 4.7 +/- 1.0 x 10(8) m(-1) s(-1) in both cases), culminates in the formation of oxidatively modified guanine bases (major product, imidazolone; minor product, 8-oxo-7,8-dihydroguanine). The G(-H)* and O(2)* radicals were generated by intense 308 nm excimer laser pulses resulting in the one-electron oxidation and deprotonation of guanine in the 5'-d(CC[2AP]-TCGCTACC) strands and the trapping of the ejected electrons by molecular oxygen (Shafirovich, V., Dourandin, A., Huang, W., Luneva, N. P., and Geacintov, N. E. (2000) Phys. Chem. Chem. Phys. 2, 4399-4408). The addition of Cu,Zn-superoxide dismutase, known to react rapidly with superoxide, dramatically enhances the life-times of guanine radicals from 4 to 7 ms to 0.2-0.6 s in the presence of 5 microm superoxide dismutase. Oxygen-18 isotope labeling experiments reveal two pathways of 8-oxo-7,8-dihydroguanine formation including either addition of O(2)* to the C-8 position of G(-H)* (in the presence of oxygen), or the hydration of G(-H)* (in the absence of oxygen). The formation of the guanine lesions via combination of guanine and superoxide radicals is greatly reduced in the presence of typical antioxidants such as trolox and catechol that rapidly regenerate guanine by the reductive "repair" of G(-H)* radicals. The mechanistic aspects of the radical reactions that either regenerate undamaged guanine in DNA or lead to oxidatively modified guanine bases are discussed. 相似文献

Social group size has been shown to correlate with neocortex size in primates. Here we use comparative analyses to show that social group size is independently correlated with the size of non-V1 neocortical areas, but not with other more proximate components of the visual system or with brain systems associated with emotional cueing (e.g. the amygdala). We argue that visual brain components serve as a social information ''input device'' for socio-visual stimuli such as facial expressions, bodily gestures and visual status markers, while the non-visual neocortex serves as a ''processing device'' whereby these social cues are encoded, interpreted and associated with stored information. However, the second appears to have greater overall importance because the size of the V1 visual area appears to reach an asymptotic size beyond which visual acuity and pattern recognition may not improve significantly. This is especially true of the great ape clade (including humans), that is known to use more sophisticated social cognitive strategies. 相似文献
