ABSTRACT: BACKGROUND: Hepatitis E virus (HEV) genotype 3 and 4 can cause liver disease in human and has its main reservoir in pigs. HEV investigations in pigs worldwide have been performed but there is still a lack of information on the infection dynamics in pig populations. FINDINGS: The HEV transmission dynamics in commercial pig farms in six different European countries was studied. The data collected show prevalence in weaners ranging from 8% to 30%. The average HEV prevalence in growers was between 20% and 44%. The fatteners prevalence ranged between 8% and 73%. Sows prevalence was similar in all countries. Boar faeces were tested for HEV only in Spain and Czech Republic, and the prevalence was 4.3% and 3.5% respectively. The collected data sets were analyzed using a recently developed model to estimate the transmission dynamics of HEV in the different countries confirming that HEV is endemic in pig farms. CONCLUSIONS: This study has been performed using similar detection methods (real time RT-PCR) for all samples and the same model (SIR model) to analyse the data. Furthermore, it describes HEV prevalence and within-herd transmission dynamics in European Countries (EU): Czech Republic, Italy, Portugal, Spain, The Netherlands and United Kingdom, confirming that HEV is circulating in pig farms from weaners to fatteners and that the reproductive number mathematical defined as R0 is in the same range for all countries studied.  相似文献

Chronic pancreatitis is characterized by inflammation, fibrosis, pain, and loss of exocrine function of the pancreas. We aimed to identify differentially expressed proteins in the ePFT-collected pancreatic fluid from individuals with chronic pancreatitis (CP; n = 9) and controls with chronic abdominal pain not associated with the pancreas (NP; n = 9). Using GeLC-MS/MS techniques, we identified a total of 1391 different proteins in 18 pancreatic fluid samples. Of these proteins, 257 and 413 were identified exclusively in the control and chronic pancreatitis cohorts, respectively, and 721 were identified in both cohorts. Spectral counting and statistical analysis thereof revealed an additional 38 and 77 proteins that were up- or down-regulated, respectively, in the pancreatic fluid from individuals with chronic pancreatitis. As expected, gene ontology analysis illustrated that the largest percentage of differentially regulated proteins was secreted/extracellular in origin. In addition, proteins that were down-regulated with statistical significance in the chronic pancreatitis cohort were determined to have biological function of proteases, corresponding to the canonical pancreatic insufficiency associated with chronic pancreatitis. Proteins enriched in the pancreatic fluid of chronic pancreatitis patients had roles in fibrosis, inflammation, and pain, whereas digestive enzymes were significantly less abundant. Our workflow provided a mass spectrometry-based approach for the further study of the pancreatic fluid proteome, which may lead to the discovery potential biomarkers of chronic pancreatitis.  相似文献

Close to 50 genetic loci have been associated with type 2 diabetes (T2D), but they explain only 15% of the heritability. In an attempt to identify additional T2D genes, we analyzed global gene expression in human islets from 63 donors. Using 48 genes located near T2D risk variants, we identified gene coexpression and protein-protein interaction networks that were strongly associated with islet insulin secretion and HbA(1c). We integrated our data to form a rank list of putative T2D genes, of which CHL1, LRFN2, RASGRP1, and PPM1K were validated in INS-1 cells to influence insulin secretion, whereas GPR120 affected apoptosis in islets. Expression variation of the top 20 genes explained 24% of the variance in HbA(1c) with no claim of the direction. The data present a global map of genes associated with islet dysfunction and demonstrate the value of systems genetics for the identification of genes potentially involved in T2D.  相似文献

Heart rate monitoring in soccer: Interest and limits during competitive match play and training-Practical application. J Strength Cond Res 26(10): 2890-2906, 2012-The identification of physiological loads imposed by soccer training or match play reveals essential information, which may help improve training and recovery strategies. Until today, the use of heart rate (HR) monitoring is not standardized in soccer. Thus, the aim of this review was to analyze, determine and compare the exercise intensity (EI) monitored by HR in professional, youth, and recreational soccer players during matches and training sessions using a meta-analysis. Heart rate is one of the most common physiological variables used to determine exercise internal training load. The mean EI recorded during competitive matches was described as 70-80% of V[Combining Dot Above]O2max or 80-90% of maximal heart rate (HRmax), independent of the playing level. With respect to HR training zones, approximately 65% of the total match duration is spent at intensity of 70-90% HRmax and rarely below 65% HRmax. However, although HRmax is mostly employed in the literature, monitoring EI should be expressed in relation to reserve heart rate, as it was described as a more reliable indicator of HR, allowing interindividual comparisons. The HR response according to the playing position indicates that midfielders are characterized by the highest EI, followed by forwards and fullbacks. Moreover, in the second half of the match, the EI is lower than that observed during the first half; this reduction could be correlated with the level of the player's physical conditioning. Consequently, coaches may favor the use of interval training or small-sided training games because these are shown to improve both aerobic capacity and the ability to repeat high-intensity actions. Small-sided games allow reaching similar HR responses to those found during interval training and match play but with greater heterogeneity values. Future investigations should include a larger sample of players with special reference to playing position and the expression of EI in percentage of the reserve heart rate, analyzing the possible intergender differences in HR response.  相似文献

The effect of barium chloride (Ba2+) on the contractile mechanism of isolated dog uteri was analysed under three different hormonal states: control, estradiol-treated (E) and estradiol + progesterone-treated (E + P). The maximal contractile effect of Ba2+ was significantly depressed in the E + P group. Tachyphylaxis developed rather quickly (t 12 ? 10 min) to successive additions of 1.0 mM Ba2+ following the Ca2+ removal from the nutrient solution. The t 12 was further reduced in the E or the E + P groups. No tachyphylaxis was observed when the much higher concentration of 30.0 mM Ba2+ was employed.In the normal nutrient solution, the calcium-blocker D600 (methoxyverapamil) produced a non-competitive antagonism of Ba2+ which was not reversed by an increase in extracellular Ca2+ concentration. In excess-potassium depolarizing solution, the dose-response curves to both Ca2+ and Ba2+ were displaced to the right by compound D600. The Schild plot yielded straight lines suggestive of competitive antagonism with the similar pA2 values of 8.57 for Ca2+ and 8.35 for Ba2+.These results suggest that Ba2+ has a dual mechanism of action on isolated dog uteri, depending on the concentration employed. At low doses (1.0 mM), the contractile effect of Ba2+ is due to a release of intracellular Ca2+, an action which can be modulated by the ovarian hormones. The effect of high concentrations of Ba2+ (30.0 mM) seems to be the result of a direct interaction with the contractile proteins and is independent of the animal's hormonal state. The results also suggest that the access of Ba2+ to the uterine fibre is possibly through the same Ca2+ channel which is acted upon by compound D600.  相似文献

The generative cell (GC) development during three sequentialstages of Magnolia x soulangeana pollen grain maturation wasinvestigated by light and electron microscopy. Plastids werenot identified in this cell but mitochondria, Golgi bodies andvesicles as well as rough endoplasmic reticulum profiles werealways present. Microtubules were also present, their numberincreasing and their disposition varying during GC maturation.The most conspicuous components of the GC cytoplasm were themicrobodies. The latter were few in number in the newly formedGC, and the appearance of their matrix was different from laterdevelopmental stages. A clear microbodial proliferation occurredin the GC during an intermediate stage of pollen maturation.Then, the microbody matrix was either fibrillar to granularas in the vegetative cell microbodies or very dense and compact.The polymorphism and size range and the frequent aggregationof these organelles in one or more clusters were also noteworthy.Tilting of semithin sections as well as the analysis of serialsections suggested that a number or enlarged and irregularlyshaped microbodies co-exist with smaller and more sphericalones, the latter probably originating by budding. In the GCof the mature pollen the microbody-like organelles were in generalmore uniform both in shape and size. The cytochemical test ofDAB was positive in the microbodies of both the pollen cells,thus demonstrating their peroxisomic nature. The function ofthe microbodies in the GC is not clear. Branching morphogenesis is a developmental process characteristic of many organ systems. Specifically, during renal branching morphogenesis, its been postulated that the final number of nephrons formed is one key clinical factor in the development of hypertension in adulthood. As it has been established that BMPs regulate, in part, renal activity of p38 MAP kinase (p38MAPK) and it has demonstrated that the cytoplasmic protein Neurotrophin Receptor MAGE homologue (NRAGE) augments p38MAPK activation, it was hypothesized that a decrease in the expression of NRAGE during renal branching would result in decreased branching of the UB that correlated with changes in p38MAPK activation. To verify this, the expression of NRAGE was reduced in ex vivo kidney explants cultures using antisense morpholino. Morpholino treated ex vivo kidney explants expression were severely stunted in branching, a trait that was rescued with the addition of exogenous GDNF. Renal explants also demonstrated a precipitous drop in p38MAPK activation that too was reversed in the presence of recombinant GDNF. RNA profiling of NRAGE diminished ex vivo kidney explants resulted in altered expression of GDNF, Ret, BMP7 and BMPRIb mRNAs. Our results suggested that in early kidney development NRAGE might have multiple roles during renal branching morphogenesis through association with both the BMP and GDNF signaling pathways.  相似文献
