Children with idiopathic short stature (ISS) may have normal or increased growth hormone (GH) responses to provocation tests and achieve a final height (FH) below -2.0 standard deviation score (SDS) if untreated. FH of subjects with high stimulated GH levels has not been studied in detail. AIM: It was the aim of this study to analyse FH in ISS patients with high GH peak responses to the provocation test. PATIENTS AND METHODS: We studied 16 patients (9 pre-pubertal) with ISS and a GH peak >or=40 mU/l to insulin-induced hypoglycaemia. The patients were recalled at age 19.7 +/- 2.5 years for measurement of FH when blood samples were obtained for serum insulin-like growth factor (IGF)-I, IGF binding protein 3, acid-labile subunit and GH binding protein measurements. GH bioactivity was determined using the Nb2 bioassay. RESULTS: FH was -3.1 +/- 1.0 SDS, being significantly lower than target height (TH). At FH, IGF-I levels were within -1.5 and +1.5 SDS for age and sex in 10 patients and higher than +1.5 SDS in 6 patients. IGF binding protein 3, acid-labile subunit, GH binding protein levels and GH bioactivity values were normal. SUMMARY: These data suggest that patients with ISS and high GH levels during a GH stimulation test may have a more compromised FH. The association of severe ISS with a peak GH >40 mU/l might suggest a degree of insensitivity for the GH-IGF-I axis. 相似文献

Essential oil yield and composition in seven natural populations of Lavandula latifolia from the eastern Iberian Peninsula were determined by GC/MS. Twenty-eight constituents were identified, accounting for 92.0–95.4% of the total oils. These oils were dominated by the monoterpene fraction and three of them (linalool, cineole and camphor) constituted 79.5–86.9% of the oil from flowers. Essential oil yield in leaves and flowers varied among and within populations, but hierarchic analyses of variance showed that the proportion of variation attributable to individuals was significantly higher than that attributable to population differences. Principal component and cluster analyses allowed three groups of flower essential oils to be distinguished according to their high, intermediate and low proportion of linalool. These essential oil types are respectively correlated to the Supra-, Meso- and Thermo-Mediterranean bioclimatic belts where the populations are located. A genetic analysis based on those terpenes that showed a trimodal distribution roughly corroborated the relationships between the seven populations obtained from the ordination analyses and emphasizes the distinctiveness of some of the populations. 相似文献

Disruption of the seqA gene of Salmonella enterica serovar Typhimurium causes defects similar to those described in E. coli: filament formation, aberrant nucleoid segregation, induction of the SOS response, envelope instability, and increased sensitivity to membrane-damaging agents. Differences between SeqA− mutants of E. coli and S. enterica, however, are found. SeqA− mutants of S. enterica form normal colonies and do not exhibit alterations in phage plaquing morphology. Lack of SeqA causes attenuation of S. enterica virulence by the oral route but not by the intraperitoneal route, suggesting a virulence defect in the intestinal stage of infection. However, SeqA− mutants are fully proficient in the invasion of epithelial cells. Organophosphates induce bronchoobstruction in guinea pigs, and salbutamol only transiently reverses this effect, suggesting that it triggers additional obstructive mechanisms. To further explore this phenomenon, in vivo (barometric plethysmography) and in vitro (organ baths, including ACh and substance P concentration measurement by HPLC and immunoassay, respectively; intracellular Ca2+) measurement in single myocytes) experiments were performed. In in vivo experiments, parathion caused a progressive bronchoobstruction until a plateau was reached. Administration of salbutamol during this plateau decreased bronchoobstruction up to 22% in the first 5 min, but thereafter airway obstruction rose again as to reach the same intensity as before salbutamol. Aminophylline caused a sustained decrement (71%) of the parathion-induced bronchoobstruction. In in vitro studies, paraoxon produced a sustained contraction of tracheal rings, which was fully blocked by atropine but not by TTX, omega-conotoxin (CTX), or epithelium removal. During the paraoxon-induced contraction, salbutamol caused a temporary relaxation of approximately 50%, followed by a partial recontraction. This paradoxical recontraction was avoided by the M2- or neurokinin-1 (NK1)-receptor antagonists (methoctramine or AF-DX 116, and L-732138, respectively), accompanied by a long-lasting relaxation. Forskolin caused full relaxation of the paraoxon response. Substance P and, to a lesser extent, ACh released from tracheal rings during 60-min incubation with paraoxon or physostigmine, respectively, were significantly increased when salbutamol was administered in the second half of this period. In myocytes, paraoxon did not produce any change in the intracellular Ca2+ basal levels. Our results suggested that: 1) organophosphates caused smooth muscle contraction by accumulation of ACh released through a TTX- and CTX-resistant mechanism; 2) during such contraction, salbutamol relaxation is functionally antagonized by the stimulation of M2 receptors; and 3) after this transient salbutamol-induced relaxation, a paradoxical contraction ensues due to the subsequent release of substance P. 相似文献

Dendritic cells (DCs) act not only as sentinels for detection of, but also as target cells for viruses, and this can be important for viral transport and spread. All subsets of DCs are equipped with a battery of receptors recognizing virus-associated molecular signatures, and recognition of those launches a maturation programme that results in substantial alterations of morphology, motility and the DCs' interactive properties with the extracellular matrix and scanning T cells. In addition to being sensed, viruses are internalized into DCs and, for the major proportion, processed into peptides that are subsequently presented by major histocompatibility complex (MHC) molecules. Transmission of virus to T cells can occur after completion of their replication cycle if the intracellular milieu of the DC permits that. Alternatively, viruses can remain protected from degradation following entrapment by pattern recognition receptors in intracellular compartments, also referred to as virosomes, which translocate towards the DC/T cell interface. Most likely, transfer of virus to T cells occurs in these junctions, referred to as infectious synapses. Resource use of Cape mountain zebra was studied in the Baviaanskloof Nature Reserve (BNR) over concerns of the poor performance in population growth. We assessed the seasonal diet, habitat suitability and forage quality of the Bergplaas area in BNR for mountain zebra. Grasses contributed 95.2% to the annual diet of mountain zebra, Tristachya leucothrix contributing the most (39.4%), followed by Themeda triandra (27.6%). Seasonally, T. triandra contributed most to the diet in winter, while T. leucothrix became more important in the summer and also was the only species preferred in all seasons. Mountain zebra concentrated their feeding in Kouga Grassy Fynbos and from our assessment this was the only habitat suitable for mountain zebra. Our analysis of mountain zebra dung indicated that the seasonal nitrogen and phosphorus content was below the threshold values prescribed for grazers, and our study suggests that mountain zebra at Bergplaas are severely resource limited. We emphasize the importance of fire and access to nutrient‐rich lowlands in influencing the nutritional ecology of mountain zebra and provide conservation management recommendations. 相似文献
