The compatible solute mannosylglucosylglycerate (MGG), recently identified in Petrotoga miotherma, also accumulates in Petrotoga mobilis in response to hyperosmotic conditions and supraoptimal growth temperatures. Two functionally connected genes encoding a glucosyl-3-phosphoglycerate synthase (GpgS) and an unknown glycosyltransferase (gene Pmob_1143), which we functionally characterized as a mannosylglucosyl-3-phosphoglycerate synthase and designated MggA, were identified in the genome of Ptg. mobilis. This enzyme used the product of GpgS, glucosyl-3-phosphoglycerate (GPG), as well as GDP-mannose to produce mannosylglucosyl-3-phosphoglycerate (MGPG), the phosphorylated precursor of MGG. The MGPG dephosphorylation was determined in cell extracts, and the native enzyme was partially purified and characterized. Surprisingly, a gene encoding a putative glucosylglycerate synthase (Ggs) was also identified in the genome of Ptg. mobilis, and an active Ggs capable of producing glucosylglycerate (GG) from ADP-glucose and d-glycerate was detected in cell extracts and the recombinant enzyme was characterized, as well. Since GG has never been identified in this organism nor was it a substrate for the MggA, we anticipated the existence of a nonphosphorylating pathway for MGG synthesis. We putatively identified the corresponding gene, whose product had some sequence homology with MggA, but it was not possible to recombinantly express a functional enzyme from Ptg. mobilis, which we named mannosylglucosylglycerate synthase (MggS). In turn, a homologous gene from Thermotoga maritima was successfully expressed, and the synthesis of MGG was confirmed from GDP-mannose and GG. Based on the measurements of the relevant enzyme activities in cell extracts and on the functional characterization of the key enzymes, we propose two alternative pathways for the synthesis of the rare compatible solute MGG in Ptg. mobilis.Thermophilic and hyperthermophilic organisms, like the vast majority of other microorganisms, accumulate compatible solutes in response to water stress imposed by salt. In fact, many of the (hyper)thermophiles known were isolated from geothermal areas venting seawater (36). However, the compatible solutes of thermophilic and hyperthermophilic prokaryotes are generally different from those of their mesophilic counterparts and some, namely, di-myo-inositol-phosphate (DIP), mannosyl-di-myo-inositol-phosphate (MDIP), diglycerol phosphate, and mannosylglyceramide, are confined to organisms that grow at extremely high temperatures (19, 22, 34, 38). Mannosylglycerate (2-α-d-mannosylglycerate; MG), for example, is a common compatible solute of thermophiles and hyperhermophiles (23, 27, 38) but has also been found in mesophilic organisms, such as red algae, where it was first identified (6). It should also be noted that there is a growing awareness that compatible solutes are involved in other types of stress; trehalose, for example, plays a role in osmotic stress, heat stress, desiccation, and freezing (9). Some compatible solutes of thermophilic organisms are extremely rare and have been encountered in only one or two, generally closely related, species. Among them are mannosylglyceramide in Rhodothermus marinus, diglycerol phosphate in Archaeoglobus fulgidus, and, more recently, mannosylglucosylglycerate (α-d-1→2-mannopyranosyl-α-d-1→2-glucopyranosylglycerate; MGG) identified in Petrotoga miotherma (16, 19, 38).The species of the genus Petrotoga represent slightly thermophilic members of the generally hyperthermophilic and deep-branching bacteria of the order Thermotogales (2, 3, 31). Organisms of this genus have all been isolated from hot oilfield water (21, 25), and have an optimum temperature for growth of 55 to 60°C in medium containing NaCl in the range of 0.5 to 10% (16). In Ptg. miotherma, the levels of MGG increased during low-level osmotic adaptation, whereas glutamate and proline were used for protection against hyperosmotic stress (16). The hyperthermophilic Thermotoga spp. accumulate primarily di-myo-inositol-phosphate and mannosyl-di-myo-inositol-phosphate during osmotic adjustment or during growth at temperatures above the optimum for growth (37).The novel compatible solute MGG is a derivative of glucosylglycerate (2-α-d-glucosylglycerate; GG) identified in the free form in Erwinia chrysanthemi, in the marine cyanobacteria Prochlorococcus marinus and Synechococcus sp. PCC7002, and in the thermophilic bacterium Persephonella marina, the latter of which possesses two alternative pathways for its synthesis (8, 13, 14, 18, 37). Glucosylglycerate has also been detected in trace amounts in Mycobacterium smegmatis, where it probably is the precursor of a polysaccharide involved in the regulation of fatty acid synthesis, as well as in the polar head group of a glycolipid from Nocardia otitidiscaviarum (17, 30).Two alternative pathways for the synthesis of GG have been identified and characterized. In the two-step reaction scheme, the synthesis of GG involves the condensation of nucleoside diphosphate (NDP)-glucose and d-3-phosphoglycerate (3-PGA) into glucosyl-3-phosphoglycerate (GPG), which in turn is dephosphorylated to yield GG. Yet, in a single-step pathway, the synthesis of GG occurs via the condensation of ADP-glucose with d-glycerate (13). Similar routes to those described above also lead to the synthesis of mannosylglycerate in Rhodothermus marinus (4).Two functionally connected genes encoding an “actinobacterial”-type glucosyl-3-phosphoglycerate synthase (GpgS) and an unknown glycosyltransferase were detected in the genome of Petrotoga mobilis (12). In this study, we examine the synthesis of MGG through a phosphorylating pathway (with a phosphorylated intermediate) from 3-phosphoglycerate and UDP-glucose to the final compatible solute, in cell extracts and by functional characterization of recombinant enzymes. We also examine a second nonphosphorylating pathway (no phosphorylated intermediates) that could represent an alternative route for the synthesis of MGG in Ptg. mobilis that could lead to the direct conversion of GG and GDP-mannose to MGG. Pathway multiplicity likely reflects a crucial role for MGG in the physiology of Ptg. mobilis during stress adaptation.  相似文献

Novel palladium(II) complexes with salen-type ligands based on 3-methylsalicyladehyde and a set of aliphatic diamines (C1 to C4) have been synthesised and characterized by spectroscopic techniques (UV-Vis and FTIR), Density Functional Theory (DFT) calculations and single-crystal X-ray diffraction for C1 and C4. X-ray diffraction analysis of these complexes was focused on coordination sphere and supramolecular arrangements. In the two compounds, the molecules form dimers, being the most relevant intermolecular interactions the hydrogen bonds of the type C-H?O, C-H?π and π?π stacking interactions between the six-membered metallocycles.Electronic spectra of all Pd(II) complexes are dominated by charge transfer and intraligand bands at λ < 400 nm. DFT calculations showed that the HOMO is ligand-dominated, with the metal contribution being ∼18% for all complexes. This suggests that the structural/electronic differences between the ligands do not influence significantly the participation of metal orbitals in HOMO. All the complexes exhibit dipole moments with the same direction, from the aldehyde moiety towards the imine bridge with C2 and C3 showing quite similar values, μC2 = 5.49 and μC3 = 5.54 D, whereas complexes C1 and C4 show slightly higher values: μC1 = 5.79 and μC4 = 6.17 D. The magnitude of bond lengths and angles predicted by DFT calculations are comparable to those determined by X-ray crystallography.The experimental vibrational frequencies of the Pd(II) complexes were correlated with the values estimated by DFT calculations. The good agreement between the experimental and theoretical vibrational data allowed the assignment of relevant IR bands to molecular vibration modes.  相似文献

A major challenge for contemporary conservation policies and practices is formulating workable compromises between wildlife conservation and the people who live with wildlife. We strongly support the view that anthropology has a critical role to play in contributing to our understanding of human-environment interactions. The study of complex biophysical and human systems can be greatly assisted by appropriate simulation models that integrate what is known about ecological and human decision-making processes. We have developed an integrated modeling system for assessing scenarios in the Ngorongoro Conservation Area in northern Tanzania to modify the situation there to improve human welfare without compromising conservation value. We present the results of some scenarios that indicate that the current situation there is not sustainable, and that tough policy decisions need to be taken if household well-being of the pastoralists who live there is to be improved or even sustained.  相似文献

BackgroundTriple negative breast cancer (TNBC) has the worst prognosis amongst all subtypes. Studies have shown that the achievement of pathologic complete response in the breast and axilla correlates with improved survival. The aim of this study was to identify clinical or pathological features of real-life TNBC patients with a higher risk of early relapse.Materials and methodsSingle-centre retrospective analysis of 127 women with TNBC, stage II–III, submitted to neoadjuvant treatment and surgery between January 2016 and 2020. Multivariate Cox regression analysis for disease free survival (DFS) at 2 years was performed and statistically significant variables were computed into a prognostic model for early relapse.ResultsAfter 29 months of median follow-up, 105 patients (82.7%) were alive and, in total, 38 patients (29.9%) experienced recurrence. The 2-year DFS was 73% (95% CI: 21.3–22.7). In multivariate analysis, being submitted to neoadjuvant radiotherapy [HR 2.8 (95% CI: 1.2–6.4), p = 0.017] and not achieving pathologic complete response [HR 0.3 (95% CI: 0.1–1.7), p = 0.011] were associated with higher risk of recurrence. In our prognostic model, the presence of at least one of these variables defined a subgroup of patients with a worse 2-year DFS than those without these features (59% vs. 90%, p < 0.001, respectively).ConclusionsIn this real-life non-metastatic TNBC cohort, neoadjuvant radiotherapy (performed due to insufficient clinical response to neoadjuvant chemotherapy or significant toxicity) impacted as an independent prognostic factor for relapse along with the absence of pathologic complete response identifying a subgroup of higher risk patients for early relapse that might merit a closer follow-up.  相似文献

Biotrophic phytopathogens are typically limited to their adapted host range. In recent decades, investigations have teased apart the general molecular basis of intraspecific variation for innate immunity of plants, typically involving receptor proteins that enable perception of pathogen-associated molecular patterns or avirulence elicitors from the pathogen as triggers for defense induction. However, general consensus concerning evolutionary and molecular factors that alter host range across closely related phytopathogen isolates has been more elusive. Here, through genome comparisons and genetic manipulations, we investigate the underlying mechanisms that structure host range across closely related strains of Pseudomonas syringae isolated from different legume hosts. Although type III secretion-independent virulence factors are conserved across these three strains, we find that the presence of two genes encoding type III effectors (hopC1 and hopM1) and the absence of another (avrB2) potentially contribute to host range differences between pathovars glycinea and phaseolicola. These findings reinforce the idea that a complex genetic basis underlies host range evolution in plant pathogens. This complexity is present even in host-microbe interactions featuring relatively little divergence among both hosts and their adapted pathogens.  相似文献

Protein-protein interactions play a pivotal role in a large number of biological processes exemplified by the assembly of the cellulosome. Integration of cellulosomal components occurs through the binding of type I cohesin modules located in a non-catalytic molecular scaffold to type I dockerin modules located at the C terminus of cellulosomal enzymes. The majority of type I dockerins display internal symmetry reflected by the presence of two essentially identical cohesin-binding surfaces. Here we report the crystal structures of two novel Clostridium thermocellum type I cohesin-dockerin complexes (CohOlpC-Doc124A and CohOlpA-Doc918). The data revealed that the two dockerins, Doc918 and Doc124A, are unusual because they lack the structural symmetry required to support a dual binding mode. Thus, in both cases, cohesin recognition is dominated by residues located at positions 11, 12, and 19 of one of the dockerin binding surfaces. The alternative binding mode is not possible (Doc918) or highly limited (Doc124A) because residues that assume the critical interacting positions, when dockerins are reoriented by 180°, make steric clashes with the cohesin. In common with a third dockerin (Doc258) that also presents a single binding mode, Doc124A directs the appended cellulase, Cel124A, to the surface of C. thermocellum and not to cellulosomes because it binds preferentially to type I cohesins located at the cell envelope. Although there are a few exceptions, such as Doc918 described here, these data suggest that there is considerable selective pressure for the evolution of a dual binding mode in type I dockerins that direct enzymes into cellulosomes.  相似文献

In order to obtain an improved understanding of the assembly of the bacterial photosynthetic apparatus, we have conducted a proteomic analysis of pigment-protein complexes isolated from the purple bacterium Rhodobacter sphaeroides undergoing acclimation to reduced incident light intensity. Photoheterotrophically growing cells were shifted from 1,100 to 100?W/m(2) and intracytoplasmic membrane (ICM) vesicles isolated over 24-h were subjected to clear native polyacrylamide gel electrophoresis. Bands containing the LH2 and reaction center (RC)-LH1 complexes were excised and subjected to in-gel trypsin digestion followed by liquid chromatography (LC)-mass spectroscopy (MS)/MS. The results revealed that the LH2 band contained distinct levels of the LH2-α and -β polypeptides encoded by the two puc operons. Polypeptide subunits encoded by the puc2AB operon predominated under high light and in the early stages of acclimation to low light, while after 24?h, the puc1BAC components were most abundant. Surprisingly, the Puc2A polypeptide containing a 251 residue C-terminal extension not present in Puc1A, was a protein of major abundance. A predominance of Puc2A components in the LH2 complex formed at high light intensity is followed by a >2.5-fold enrichment in Puc1B levels between 3 and 24?h of acclimation, accompanied by a nearly twofold decrease in Puc2A levels. This indicates that the puc1BAC operon is under more stringent light control, thought to reflect differences in the puc1 upstream regulatory region. In contrast, elevated levels of Puc2 polypeptides were seen 48?h after the gratuitous induction of ICM formation at low aeration in the dark, while after 24?h of acclimation to low light, an absence of alterations in Puc polypeptide distributions was observed in the upper LH2-enriched gel band, despite an approximate twofold increase in overall LH2 levels. This is consistent with the origin of this band from a pool of LH2 laid down early in development that is distinct from subsequently assembled LH2-only domains, forming the LH2 gel band.  相似文献

In this study, we describe the association of three Campylobacter jejuni metabolism-related traits, γ-glutamyl-transpeptidase (GGT), fucose permease (fucP), and secreted L-asparaginase [ansB(s)], with multilocus sequence types (STs). A total of 710 C. jejuni isolates with known STs were selected and originated from humans, poultry, bovines, and the environment. Among these isolates, we found 31.1% to produce GGT and 49.3% and 30.3% to be positive for ansB(s) and fucP, respectively. The combination of GGT production, the presence of ansB(s), and the absence of fucP was associated with ST-22, ST-586, and the ST-45 and ST-283 clonal complexes (CCs), which were the main STs and CCs found among the human and chicken isolates. The ST-21 CC was associated with the presence of fucP and was the major CC among the bovine isolates. Although the ST-61 CC was the second major CC among the bovine isolates, these isolates did not have any of the markers studied, making the role of fucP in bovine gut colonization questionable. The ST-45 CC was subdivided into three groups that were attributed solely to ST-45. One group showed a marker combination described previously, another group was found to be positive for ansB(s) only, and the third group did not have any of the markers studied. These results suggest that the host association of these markers seems to be indirect and may arise as a consequence of host-ST and -CC associations. Thus, a representative collection of STs should be tested to draw sensible conclusions in similar studies.  相似文献

Cationic cross-linked starch (CCS)-iodine complexes containing different amounts of quaternary ammonium groups (different degrees of substitution (DS)) and iodine have been obtained by iodine adsorption on CCS from aqueous iodine potassium iodide solution. Equilibrium adsorption studies showed that with an increase of DS the amount of iodine adsorbed on CCS and the affinity of iodine to CCS increased linearly. The influences of the DS of CCS and the amount of adsorbed iodine on the stability of CCS-iodine complexes in a solution of 0.02M sodium acetate and reactivity toward l-tyrosine have been investigated. At the same DS, the stability of CCS-iodine complexes decreased with an increase of the amount of adsorbed iodine. With increasing the DS, the stability of CCS-iodine complexes increased. The iodine consumption in the reaction with l-tyrosine increased significantly with an increase of the amount of adsorbed iodine. The influence of DS on iodine consumption was lower and depended on the amount of adsorbed iodine. The antibacterial activity of CCS-iodine complexes against Bacillus cereus, Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli was determined by the broth-dilution and spread-plate methods. The obtained results have demonstrated that an appropriate selection of the CCS-iodine complex composition (the DS of CCS and the amount of adsorbed iodine) could ensure good antimicrobial properties by keeping a low concentration of free iodine in the system. The main advantage of using CCS-iodine complexes as antimicrobial agents is the biodegradability of the polymeric matrix.  相似文献
