Subcellular fractionation studies were performed on human neutrophils stimulated with ionomycin (a Ca(2+)-specific ionophore). The results of these studies revealed NADPH-oxidase activity, without any additive, both in the plasma membrane and in the specific granule fractions. After comparing these results with the NADPH oxidase activity induced by the ionophore in intact neutrophils, in differentiated HL-60 cells and in neutrophil cytoplasts, we conclude that ionomycin preferentially activates the NADPH oxidase pool located in the membrane of specific granules. Furthermore, we suggest that incorporation of granule membrane into the plasma membrane makes the associated NADPH oxidase less sensitive to activation induced by a rise in [Ca(2+)]i. 相似文献

The effects of size and previous sexual maturity on downstream migration in two-summer-old Baltic salmon were studied during their normal seaward migratory period in spring. We used an automatic PIT (Passive Integrated Transponder) tag monitoring system for individual recognition of tagged fish. Size affected the probability of migration among both previously immature and previously mature males while there was no such relationship among females. Nearly all females and 63.6% of the previously immature males migrated during the smolting period, while only 24.5% of the previously mature males did. In migrating fish the individual activity level increased from almost zero in May to about 450 antenna passages per day in mid June, coinciding with a pronounced increase in ambient water temperature. During the period of increasing activity in early June the fish changed from night activity to day activity and finally activity all day and night. The fish moved downstream in schools but no specific school size dominated. This paper demonstrates the strong effect of body size and previous sexual maturity on the probability of migration. 相似文献

The total aestival ectoparasitic burden of six small mammal species ( Sorex araneus, Clethrionomys glareolus, C. rutilus, C. rufocanus, Microtus agrestis , and M. oeconomus ) was investigated in terms of frequency distribution, frequency of occurrence, species diversity and joint occurrences. The mammals were collected in northern Fennoscandia during peak density years. The frequency distribution of the ectoparasites was best described as negative binomial on C. glareolus, M. agrestis , and M. oeconomus but not so on S. araneus, C. rutilus and C. rufocanus. The distribution did not fit the Poisson distribution in any species. The percentage of S. araneus that had ectoparasites was 49%, and of the microtidae species, 73-96% had ectoparasites. The median number of ectoparasites on the vole species was between 2 and 9 specimens of 1 or 2 species. There was a significant, positive correlation between the number of ectoparasitic species and the total number of individuals on all host species. Pairs of ectoparasitic species occurring together more or less often than expected by chance were found on all host species. However, the pairs rarely repeated themselves on the same host species under different environmental conditions, or on other host species under similar circumstances. In 5 anaesthetized pigs blood was sampled both from the pancreatic venous effluent and from the peripheral venous circulation. Insulin and somatostatin were determined by radioimmunoassay. The somatostatin level in pancreatic venous blood was increased by a factor 2 as compared to the peripheral blood. Concomitant with the increase in insulin after intravenous glucose a significant decrease in somatostatin immunoreactivity was found, supporting the hypothesis that endogenous somatostatin participates in the local regulation of insulin release. 相似文献

Pancreatic polypeptide has been suggested as a marker for endocrine malignancies of the gastrointestinal tract. However, the secretion of PP shows great intra- and inter-individual variation, causing both false negative and positive results. In order to reduce these risks, we have evaluated a new combined stimulatory and inhibitory test of PP secretion. Six healthy subjects, 23 members of three MEA I families, seven patients with malignant pancreatic endocrine tumours and four patients with carcinoid tumours of the gastrointestinal tract were subjected to a standardized test meal, followed by intravenous atropine 60 min after the start of the meal. Serum PP was monitored during 2 h. In healthy subjects the meal caused a rapid increase in serum PP within 20 min and intravenous atropine caused a significant (P less than 0.05) decrease of serum PP within 15 min. Patients with malignant endocrine pancreatic tumours or carcinoids had a delayed response after the test meal, with maximum levels at 45 min, but still with a significant inhibition by atropine. Even tumour patients with initially normal or slightly increased basal PP levels showed this secretion pattern. Healthy members of MEA I families displayed identical PP curves to healthy subjects, whereas members with elevated basal PP levels who had been previously affected by hyperparathyroidism and/or prolactinomas showed similar secretion patterns to pancreatic tumour patients. We think that a meal stimulatory test is of great value in the diagnosis of gastrointestinal endocrine tumours and also in the identification of subjects with the MEA I trait, who are at high risk of having pancreatic endocrine tumours. 相似文献

Size-dependent attack rate and handling capacity: inter-cohort competition in a zooplanktivorous fish 总被引:1,自引:0,他引:1
Two size‐dependent processes, metabolic requirements and foraging capacity, heavily influence the competitive ability of organisms. We studied the size‐dependent competitive ability of roach (Rutilus rutilus) in a laboratory experiment to determine the attack rate of roach as a function of roach and zooplankton sizes. The estimated size‐dependent attack rates, size‐dependent metabolic demands and handling capacities were subsequently used to interpret the outcome of a competition experiment between two size classes of roach. Furthermore, size‐dependent attack rates were implemented in an optimal foraging model to predict consumption rates and zooplankton selection to reveal the mechanisms behind competition. The attack rate first increased with roach size to a maximum around 160 mm to thereafter decrease. Based on this result, we predicted that, small (150 mm) roach had a double advantage in competition for zooplankton in the pond experiment due to their higher attack rate and their lower metabolism compared to large (230 mm) roach. As expected, small roach depressed total zooplankton biomass to a higher extent than large roach, included more zooplankton in their diet and consumed smaller zooplankton. Predicted smallest size class of zooplankton in the diet was close to the observed. Also as expected, large roach fed more on the benthic resource and depressed the benthic resource to a larger extent than small roach. Large roach affected large zooplankton to a higher extent during the first part of the experiment, which could be related to their overall higher handling capacity. The higher impact of large roach on large zooplankton during the first part of the experiment, in turn, resulted in a lower estimated mass intake of zooplankton by small roach in the mixed treatment compared to small roach only treatments. Both small and large roach had a lower growth rate in the mixed treatment compared to single size class treatments. We relate the lower growth rate of small roach in the mixed treatment to large roach's higher efficiency on benthic resources and on large zooplankton during the first part of the experiment. In correspondence with diet data, large roach preferred the shore area of the pond with more benthic invertebrates and were also found more often close to the bottom. Although our results are explainable by exploitative interactions, the fact that the presence of large roach affected the feeding position of small roach points to that social interactions were also involved. Overall, our study implies that a mechanistic understanding is crucial for the interpretation of competition experiments, especially in systems with size‐structured interactions. 相似文献
