The effects on sex pheromone-releasing, or calling behaviour, of diel photoperiods of varying daylength, of light cycle phase shifts, and of continuous illumination were investigated in Trogoderma glabrum females. On light régimes with 8 to 20 hr daylengths, calling maxima tended to centre close to photophase midpoints. Although influencing the time of day at which calling occurred, daylength had little effect on the amount of activity or the length of the calling period. When 16 : 8 LD light cycles were advanced or delayed by 4 hr, the time of day at which calling peaks were observed shifted within 2 to 4 cycles so that a constant phase relationship with photoperiod was maintained. Daily calling peaks were evident in groups of females exposed to between 1 and 5 days of continuous illumination, but mean calling time occurred earlier in the day as light exposures were lengthened. It was concluded that a circadian rhythm of calling behaviour exists in T. glabrum females. and that the rhythm can be entrained to 24 hr periodicity by photoperiod.  相似文献

Before parasitoids and predators are fully endorsed as biological control agents in storage facilities, a reliable technique must be developed to determine how much they contribute to the overall insect contamination of stored commodities. Because determining the origin of insect fragments by visual examination is difficult, labor-intensive, and requires special skills, we propose using immunological techniques to differentiate among insect species biochemically. In the example presented here, we generated monoclonal antibodies (MAbs) against the parasitic wasps Laelius pedatus (Say) and Bracon hebetor Say. The MAbs reacted with all life stages and both sexes of the parasitoids. In Western blots of acrylamide and agarose gels, the MAbs recognized high molecular weight proteins (>500 kDa) composed of multiple subunits, with mildly acidic to neutral isoelectric focusing points. The MAbs did not cross-react with the additional 22 insect species we tested in an indirect enzyme-linked immunosorbent assay. These data suggest that MAb-based immunoassays could be used to differentiate among beneficial and destructive insects found in stored products.  相似文献

This research assessed the efficacy of three harvesting methods on a strain of Dunaliella viridis. While there is strong potential to use lipids from microalgae as a feedstock for biofuels to replace petroleum-based fuel, at present microalgal harvesting for biofuel production is not yet economically feasible or energy efficient. pH-induced flocculation (by adjusting the pH of exponentially growing cells), indirect electrocoagulation (applying aluminum hydroxide coagulant to culture), and hollow fiber filtration (separating biomass from medium using tangential flow) were compared as potential harvesting mechanisms for small-scale (3–10 L) and large-scale (30–150 L) volumes of D. viridis. Both pH-induced flocculation and electrocoagulation yielded significant biomass recovery (>95 %), but both methods required removal of added chemicals and/or coagulant before the medium could be reused. In contrast, hollow fiber filtration did not require added chemicals or coagulant, and as another advantage, the filtrate was successfully reused as culture medium without apparent detrimental effects on cell size, cell shape, or cell production. When high salinity stress was imposed on the concentrate produced from the filtration method, total fatty acids (FAs) did not increase. However, total FAs did significantly increase after hollow fiber filtration (49 %) in comparison to FA content before filtration (36 %). This research indicates that hollow fiber filtration as a commercial harvesting mechanism offers attractive advantages as a harvesting mechanism for microalgae such as Dunaliella, relative to pH-induced flocculation and indirect electrocoagulation.  相似文献

Epidemiologists aim to inform the design of public health interventions with evidence on the evolution, emergence and spread of infectious diseases. Sequencing of pathogen genomes, together with date, location, clinical manifestation and other relevant data about sample origins, can contribute to describing nearly every aspect of transmission dynamics, including local transmission and global spread. The analyses of these data have implications for all levels of clinical and public health practice, from institutional infection control to policies for surveillance, prevention and treatment. This review highlights the range of epidemiological questions that can be addressed from the combination of genome sequence and traditional ‘line lists’ (tables of epidemiological data where each line includes demographic and clinical features of infected individuals). We identify opportunities for these data to inform interventions that reduce disease incidence and prevalence. By considering current limitations of, and challenges to, interpreting these data, we aim to outline a research agenda to accelerate the genomics-driven transformation in public health microbiology.  相似文献

Electron microscopy has been used to study the fine structural organization of R-banded chromosomes prepared by treatment of the chromosomes with a hot NaH2PO4 solution. The results indicate that there is a structural basis for R-banding with this technique. In comparison to untreated control chromosomes, the R-banded chromosomes had a greatly reduced electron density, suggesting that the heat treatment has a general adverse effect on chromosome structure. Chromatin fibers formed a coarse, irregular network throughout the chromosome and were often enlarged, probably as a result of the fusion of two or more native fibers. The chromatin fibers were more aggregated and had an increased electron density in the R-band regions of the chromosome than in the interbands. This indicates that the treatment has a differential effect on the structure of bands and interbands. A comparison of the ultrastructure of R- and G-banded chromosomes demonstrated that the distribution of aggregated chromatin was reversed by these two types of banding techniques; however, the treatments producing R-banding appeared to induce less extreme differences in the degree of chromatin condensation in band and interband regions than those giving rise to G-banding. It is suggested that alterations of DNA-protein interactions may arise from the differential denaturation of proteins and/or DNA in R-band and interband regions during the heat pretreatment. Such differential alterations in DNA-protein interactions may induce localized changes in the organization of chromatin and may account for the subtle morphological differences observed between the band and interband regions.  相似文献

Human metapneumovirus (hMPV) is a recently described member of the Paramyxoviridae family/Pneumovirinae subfamily and shares many common features with respiratory syncytial virus (RSV), another member of the same subfamily. hMPV causes respiratory tract illnesses that, similar to human RSV, occur predominantly during the winter months and have symptoms that range from mild to severe cough, bronchiolitis, and pneumonia. Like RSV, the hMPV virus can be subdivided into two genetic subgroups, A and B. With RSV, a single monoclonal antibody directed at the fusion (F) protein can prevent severe lower respiratory tract RSV infection. Because of the high level of sequence conservation of the F protein across all the hMPV subgroups, this protein is likely to be the preferred antigenic target for the generation of cross-subgroup neutralizing antibodies. Here we describe the generation of a panel of neutralizing monoclonal antibodies that bind to the hMPV F protein. A subset of these antibodies has the ability to neutralize prototypic strains of both the A and B hMPV subgroups in vitro. Two of these antibodies exhibited high-affinity binding to the F protein and were shown to protect hamsters against infection with hMPV. The data suggest that a monoclonal antibody could be used prophylactically to prevent lower respiratory tract disease caused by hMPV.  相似文献
