Summary The compound eye of Psychoda cinerea comprises two types of ommatidia, arranged so as to divide the retina into distinct dorsal and ventral regions. The P-type ommatidium, in the ventral part of the eye, differs fundamentally from the other dipteran ommatidia so far described, and is regarded as a primitive ommatidium. The acone dioptric apparatus is the same in both types, with a spherical lens and four Semper cells, the processes of which expand below the rhabdom to form a ring of pigment sacs. Only the distal region of the rhabdom is surrounded by a continuous ring of screening pigment, formed by 2 primary and 12–16 secondary pigment cells. The highly pigmented retinula cells penetrate the basement membrane proximally at about the level of their nuclei; in this region they are separated from the hemolymph by glial elements. The rhabdomeres R1–6 are fused to form a tube. The two types of ommatidia are defined by the arrangement of the retinula cells R7/8: in the T type the central rhabdomeres are one below the other, in the usual tandem position, whereas in the P type only R8 is central, with R7 in the peripheral ring. In the proximal region of the retina, retinula cells with parallel microvilli in neighboring ommatidia are joined in rows by lateral processes from the R8 cells. All the rhabdomeres are short and not twisted, which suggests that the retinula cells are highly sensitive to direction of polarization. The eye can adapt by a number of retinomotor processes. These findings, together with observations of behavior, imply that the psychodids have well-developed visual abilities. 相似文献

Summary Patients with recessive X-linked ichthyosis
Patients with recessive X-linked ichthyosis (RXLI), one hereditary form of scaly skin, lack activity of the enzyme steroid
sulfatase in all tissues studied. To investigate the molecular defect underlying the lack of enzyme activity, we prepared
antisera against normal enzyme by injecting normal placental microsomal suspensions or partially purified steroid sulfatase
into rabbits. Antibody activity was assessed by immunoprecipitation of detergent solubilized steroid sulfatase. In addition,
we prepared rabbit antisera against RXLI placental microsomal suspensions. To detect immunologically cross-reactive material
in patients' placentas, extracts were studied by immunoblot techniques and by competition with normal enzyme for antibody
binding. Patients' extracts did not contain immunoreactive material co-migrating on electrophoresis with purified enzyme nor
did they inhibit immunoprecipitation of normal enzyme. Sera from rabbits immunized with RXLI placental microsomes contain
no antibodies to normal steroid sulfatase, as judged by their failure to immunoprecipitate normal enzyme or to react with
normal steroid sulfatase on immunoblot. Thus the mutation in RXLI appears to reduce steroid sulfatase enzyme protein as well
as enzyme activity.

Extraction with dimethyl sulfoxide of wood-meal of the stem of bracatinga (Mimosa scabrella), a south Brazilian hardwood, that was defatted and delignified by treatment with aqueous chlorine at 0–5° followed by extraction with cold ethanol, gave a soluble O-acetylated 4-O-methyl-d-glucurono-d-xylan having (1→4)-linked β-d-xylopyranosyl residues that were unsubstituted (65%) and 2-O-(14%), 3-O- (16%), and 2,3-di-O-acetylated (5%), as determined by methylation analysis. Another preparation obtained by use of refluxing ethanol in the delignification process showed neither removal nor migration of acetyl groups. By comparison with synthetic, partly O-acetylated d-xylans of known composition, 13C-n.m.r. spectroscopy indicated that O-acetyl group migration does not occur during treatment with cold aqueous chlorine, refluxing ethanol, or water at 70°. Methyl 2-O-acetyl-4-O-methyl-β-d-xylopyranoside (6) was also unaffected by aqueous chlorine. O-Acetyl group migration took place more readily in aqueous and dimethyl sulfoxide solutions of 6 than of O-acetyl-d-xylans. The lowest temperatures at which migration was observed in monosaccharides was at 50 and 70° for solutions in D2O and (CD3)2SO, respectively. 相似文献

Summary Using cultured trophoblast cells obtained by chorionic villus biopsy, we diagnosed Fanconi anemia (FA) in two pregnancies and excluded it in eight pregnancies at risk for the syndrome. Baseline chromosomal breakage and breakage induced by diepoxybutane (DEB) were analyzed. Increased breakage was used as a marker for the syndrome. Our results were unambiguous and provide a reliable method for prenatal detection of FA in the first trimester of pregnancy. 相似文献

Abstract The genetic determinant coding for the P-specific F8 fimbriae was cloned from the chromosome of the Escherichia coli wild-type strain 2980 (O18:K5:H5:F1C, F8). The F8 determinant was further subcloned into the Pst I site of pBR322 and a restriction map was established. In a Southern hybridization experiment identity between the chromosomally encoded F8 determinant of 2980 and its cloned counterpart was demonstrated. The cloned F8 fimbriae and those of the wild type strain consist of a protein subunit of nearly 20 kDa. F8 fimbriated strains were agglutinated by an F8 polyclonal antiserum, caused mannose-resistant hemagglutination and attached to human uroepithelial cells. Summary The intracerebral distribution patterns of 14C-morphine, 3H-dihydromorphine, and 3H-fentanyl after intraventricular injection were studied autoradiographically in rats and rabbits. The extent of permeation into the ventricular wall was measured at different times after injection. The hydrophilic morphine and dihydromorphine could be demonstrated within the tissue up to 4 hours. They seemed to be retained within the gray matter and hindered in crossing fiber bundles. On the other hand, the lipophilic fentanyl was quickly removed from the brain but remained relatively longer demonstrable within the white matter. Also, after intrathecal injection of 14C-morphine a time dependent spread from the injection site was observed. The use of autoradiography in pharmacological experiments as described was found advantageous. Thus, it is possible to correlate directly, the time course of the pharmacological effect and the respective distribution pattern of the drug applied.
