Uptake of [3H]-glycine by sections of Mimosa pudica L. pulvini is pH dependent (maximum at pH 5.5) and exhibits biphasic saturation kinetics in the range of concentrations tested (1–75 m M ). Effects of compounds which increase [fusicoccin (FC)] or decrease (uncouplers, ATPase inhibitors) the proton-motive force were tested both on the pH variations induced in the incubation medium and on glycine uptake by the pulvinar tissues: there is a close relationship between the time required for the effect of these compounds on the acidification (for FC) and the pH rise (for the inhibitors) of the medium and that needed respectively for promotion and inhibition of glycine uptake. Experiments with sulfhydryl-reacting compounds show that N-ethylmaleimide induces a large rise in pH in the incubation medium and strongly inhibits glycine uptake, whereas p -chloromercuribenzenesulfonic acid has less effect on these processes. These results argue for a proton-glycine symport mechanism in the pulvinar tissue and thus support the previously postulated involvement of a proton pump in the regulation of pulvinar movement.  相似文献

The dielectric constant and conductivity of calf thymus DNA were investigated at frequencies between 0.1 MHz and 70 GHz. This work is to investigate the dielectric properties of DNA in low gigahertz region and also to study whether the dielectric behavior of the water is affected by the presence of highly charged DNA. The results of these measurements indicate the presence of two anomalous dispersions, the one between 1 MHz and 1 GHz and the second one above 1 GHZ. The dispersion at low frequencies is likely to arise from polar groups in the DNA molecule. The relaxation behavior of unbound water in DNA solution is only slightly affected by the presence of DNA at concentrations below 1%.  相似文献

Summary We investigated whether human peripheral blood monocytes isolated by centrifugal elutriation from healthy donors could be acitivated to become tumoricidal and release tumor cytolytic factor (TCF) subsequent to incubation with recombinant human interferon-gamma (r-IFN-) or a derivative of muramyl dipeptide (nor-MDP), or both. Blood monocytes incubated in endotoxin-free medium containing up to 1000 U/ml of r-IFN- or in medium containing less than 1 g/ml of nor MDP were not activated to lyse radiolabeled allogeneic human tumor cells. In contrast, the incubation of monocytes with various dose combinations of r-IFN- and nor-MDP generated significant direct cytotoxic activity as well as production of TCF. Preincubation of the r-IFN- and nor-MDP mixture with polymyxin B did not inhibit the synergism, thus ruling out the possibility that the process was due to endotoxin contamination. TCF harvested from monocyte culture supernatants was cytolytic against five allogeneic tumor targets, but not against a nontumorigenic cell line. Intestinal brush border vesicles of a Mediterranean sea fish (Dicentrarchus labrax) were prepared using the Ca2+-sedimentation method. The transport of glucose, glycine and 2-aminoisobutyric acid is energized by an Na+ gradient (out > in). In addition, amino acid uptake requires Cl? in the extravesicular medium (2-aminoisobutyric acid more than glycine). This Na+- and Cl?-dependent uptake is electrogenic, since it can be stimulated by negative charges inside the vesicles. The specific Cl? requirement of glycine and 2-aminoisobutyric acid transport is markedly influenced by pH, a change from 6.5 to 8.4 reducing the role played by Cl?. In the presence of Cl?, the Km of 2-aminoisobutyric acid uptake is reduced and its Vmax is enhanced. Cl? affects also a non-saturable Na+-dependent component of this amino acid uptake. Amino acid transport is also increased by intravesicular Cl? (2-aminoisobutyric acid less than glycine). This effect is more concerned with glucose uptake, which can be then multiplied by 2.3. A concentration gradient (in > out) as well as the presence of Na+ in the incubation medium seems to enter into this requirement. Dimethylsulfoxide (DMSO) acts on dark- and light-induced movements exhibited by leaflets of isolated leaves of Cassia fasciculate Michx. The closing movement (scotonasty), induced when the leaves are placed in darkness during the normal period of daylight, was inhibited, whereas the opening movement (photonasty), when the leaves arc transferred to light during the normal period of darkness, was promoted. The concentration for significant effects of DMSO was 1% (v/v) when applied over a 3-h period. After five days, a necrosis of the leaflets was observed for DMSO concentrations as small as 0.1%, applied over a 6-h period. Complete abscission took place if 3% DMSO was applied for more than 30 min.  相似文献

Slowly cooled cells of Streptomyces aureofaciens contained mainly tight-couple ribosomes. Maximum rate of polyphenylalanine synthesis on ribosomes of S. aureofaciens was observed at 40°C, while cultures grew optimally at 28°C. Ribosomes of S. aureofaciens differed from those of E. coli in the amount of poly(U) required for maximum synthetic activity. The polyphenylalanine-synthesizing activity of E. coli ribosomes was about 3-times higher than that of S. aureofaciens ribosomes. The addition of protein S1 of E. coli or the homologous protein from S. aureofaciens had no stimulatory effect on the translation of poly(U). In order to localize alteration(s) of S. aureofaciens ribosomes in the elongation step of polypeptide synthesis we developed an in vitro system derived from purified elongation factors and ribosomal subunits. The enzymatic binding of Phe-tRNA to ribosomes of S. aureofaciens was significantly lower than the binding to ribosomes of E. coli. This alteration was mainly connected with the function of S. aureofaciens 50 S subunits. These subunits were not deficient in their ability to associate with 30 S subunits or with protein SL5 which is homologous to L7/L12 of E. coli.  相似文献

We have isolated a cDNA clone carrying a 1560 bp long insert which contains the entire coding and 3' untranslated regions of an H-2K(d) mouse histocompatibility antigen. From the Caspian coastal region of Iran a new taxon,Sparganium erectum L. subsp.mazanderanicum Ponert, is described and illustrated. This new subspecies in some of its characters is near toSparganium erectum L. subsp.microcarpum (Neumann)Domin (with its east distribution trend); in other characters it is near toSparganium erectum L. subsp.neglectum (Beeby)Schinz etThell.
