目的:明确人新基因spindlin1的组织表达谱、相互作用蛋白。方法与结果:应用PCR技术获得spindlin1的开放读框,并将其插入原核表达载体pGEX-2T,转化大肠杆菌BL21(DE3),经IPTG诱导后,融合蛋白GST-Spindlin1获得了可溶性表达。用SDS-PAGE胶上的目的条带做抗原制备了多克隆抗体,进行了抗体纯化。结论:spindlin1在OVCar3细胞中可能以二聚体形式存在,这一现象还有待于进一步研究。  相似文献

Both the ventral and dorsal visual streams in the human brain are known to be involved in reading. However, the interaction of these two pathways and their responses to different cognitive demands remains unclear. In this study, activation of neural pathways during Chinese character reading was acquired by using a functional magnetic resonance imaging (fMRI) technique. Visual-spatial analysis (mediated by the dorsal pathway) was disassociated from lexical recognition (mediated by the ventral pathway) via a spatial-based lexical decision task and effective connectivity analysis. Connectivity results revealed that, during spatial processing, the left superior parietal lobule (SPL) positively modulated the left fusiform gyrus (FG), while during lexical processing, the left SPL received positive modulatory input from the left inferior frontal gyrus (IFG) and sent negative modulatory output to the left FG. These findings suggest that the dorsal stream is highly involved in lexical recognition and acts as a top-down modulator for lexical processing.  相似文献

A europium‐sensitized fluorescence spectrophotometry method using an anionic surfactant, sodium dodecyl benzene sulphonate (SDBS), was developed for the determination of gatifloxacin (GFLX). The GFLX–Eu3+–SDBS system was studied and it was found that SDBS significantly enhanced the fluorescence intensity of the GFLX–Eu3+ complex (about 25‐fold). The optimal experimental conditions were determined as follows: excitation and emission wavelengths of 338 and 617 nm, pH 7.5, 3.0 × 10–6 mol/L europium(III), and 5.0 × 10–5 mol/L SDBS. The enhanced fluorescence intensity of the system (ΔIf) showed a good linear relationship with the concentration of GFLX over the range 1.0 × 10–8–8.0 × 10–7 mol/L with a correlation coefficient of 0.9990. The detection limit (S:N = 3) was determined as 1.0 × 10–9 mol/L. This method has been successfully applied for the determination of GFLX in pharmaceuticals and human urine/serum samples. Compared with most other methods reported, the rapid and simple procedure proposed here offered higher sensitivity, wider linear range and good stability. Human retinal pigment epithelium (HRPE) cells are important in maintaining the normal physiology within the neurosensory retina and photoreceptors. Recently, transplantation of HRPE has become a possible therapeutic approach for retinal degeneration. By negative immunoselection (CD45 and glycophorin A), in this study, we have isolated and cultivated adult human bone marrow stem cells (BMSCs) with multilineage differentiation potential. After a 2- to 4-week culture under chondrogenic, osteogenic, adipogenic, and hepatogenic induction medium, these BMSCs were found to differentiate into cartilage, bone, adipocyte, and hepatocyte-like cells, respectively. We also showed that these BMSCs could differentiate into neural precursor cells (nestin-positive) and mature neurons (MAP-2 and Tuj1-positive) following treatment of neural selection and induction medium for 1 month. Furthermore, the plasticity of BMSCs was confirmed by initiating their differentiation into retinal cells and photoreceptor lineages by co-culturing with HRPE cells. The latter system provides an ex vivo expansion model of culturing photoreceptors for the treatment of retinal degeneration diseases.  相似文献

目的分析大连市2011-2017年7例输入性卵形疟原虫感染病例的病原、抗原及核酸检测结果,以提高卵形疟原虫诊断的准确率和效率。方法收集7例疑似疟疾患者的血样,对血样进行血涂片镜检、快速疟疾诊断试纸条检测(RDT)及荧光定量PCR检测。结果血涂片镜检结果显示7例患者均感染卵形疟原虫。RDT检测结果显示2例阴性,另外5例为泛疟原虫抗原阳性。荧光定量PCR结果显示7例患者均检出疟原虫属和卵形疟原虫核酸。结论综合镜检、RDT及荧光定量PCR检测结果确定该7例患者均为卵形疟原虫感染。  相似文献

Lipopolysaccharide is strongly associated with septic shock, leading to multiple organ failure. It can activate monocytes and macrophages to release proinflammatory mediators such as tumor necrosis factor- (TNF-), interleukin-1 (IL-1), and nitric oxide (NO). The present experiments were designed to induce endotoxin shock by an intravenous injection ofKlebsiella pneumoniae lipopolysaccharide (LPS, 10 mg/kg) in conscious rats. Arterial pressure and heart rate (HR) were continuously monitored for 48 h after LPS administration. N-Acetyl-cysteine was used to study its effects on organ damage. Biochemical substances were measured to reflect organ functions. Biochemical factors included blood urea nitrogen (BUN), creatinine (Cre), lactic dehydrogenase (LDH), creatine phosphokinase (CPK), aspartate transferase (GOT), alanine transferase (GPT), TNF-, IL-1, methyl guanidine (MG), and nitrites/nitrates. LPS caused significant increases in blood BUN, Cre, LDH, CPK, GOT, GPT, TNF-, IL-1, MG levels, and HR, as well as a decrease in mean arterial pressure and an elevation of nitrites/nitrates. N-Acetylcysteine suppressed the release of TNF-, IL-1, and MG, but enhanced NO production. These actions ameliorate LPS-induced organ damage in conscious rats. The beneficial effects may suggest a potential chemopreventive effect of this compound in sepsis prevention and treatment.  相似文献

目的:建立定量检测伤寒沙门菌表面呈现表达的流感病毒抗原的间接ELISA方法。方法:ELISA板以2.5%的戊二醛溶液预处理,将呈现表达M2e等流感病毒抗原表位的伤寒沙门菌的全细胞抗原在ELISA板上进行干燥包被,通过间接ELISA确立全菌抗原的最佳包被浓度;分别采用化学合成多肽M2e和GST-M2e融合蛋白干燥包被ELISA板,绘制标准曲线,对沙门菌表面呈现表达的抗原进行定量分析;对多肽和融合蛋白干燥包被进行比较,同时确立用于定量表面展示量的回归方程。结果:用多肽包被测定的呈现表达的M2e分子数为9.8×104,以GST-M2e包被测定的呈现表达的M2e分子数为1.3×105。结论:利用全菌干燥包被ELISA板可以对伤寒沙门菌表面呈现表达的抗原进行很好的定量分析。  相似文献
