To elucidate the bacterial diversity in biofilms formed on a condenser tube from a nuclear power plant, 16S rRNA gene sequences were examined using a PCR-cloning-sequencing approach. Twelve operational taxonomic units were retrieved in the clone library, and the estimated species richness was low (13.2). Most of the clones (94.7%) were affiliated with α-Proteobacteria; Planctomycetes and γ-Proteobacteria were much rarer. Interestingly, except for one clone belonging to Pseudoalteromonas, most of the sequences displayed sequence similarities <97% of those of the closest type strains. Based on 16S rRNA phylogenetic analysis, most bacteria were assigned to novel taxa above the species level. The low species richness and unusual bacterial composition may be attributable to selective pressure from chlorine in the cooling water. To prevent or control bacterial biofilms in cooling circuits, additional studies of the physiology and ecology of these species will be essential.  相似文献

The purpose of this microcosm experiment was to determine whether the freshwater snail Bellamya aeruginosa affected phytoplankton community and water quality. Three treatments of different snail densities (low, medium, and high) and a control (no snails) were set up in twelve enclosures. Chlorophyll a (chl a), transparency (SD), water temperature (WT), dissolved oxygen (DO), pH, nutrients, and abundance of phytoplankton were determined on days 0, 3, 7, 11, 15, 23, 31, 46, and 61. The total chl a concentration decreased and SD increased in the treatments, whereas the proportion of cyanobacteria chl a increased. Bellamya seemed to result indirectly in a decrease of DO, by reducing the algal biomass rather than by respiration. A significant decrease in nitrogen–phosphorus ratios was observed in the treatments. In the enclosed system the abundance of colonial green algae decreased over time whereas that of cyanobacteria and flagellates increased. Principal response curves showed that both phytoplankton community and water quality in the medium and high-density treatments were consistently different from in the control. These results indicate that the presence of snails resulted in a significant change of water physicochemical properties and phytoplankton community.  相似文献

Thaumatin‐like proteins (TLPs) share structural similarity with the sweet‐tasting thaumatin protein but exhibit antifungal activity by inhibiting growth of fungal pathogens. In a Tenebrio model, two TLP genes were identified by RNA‐sequencing analysis and genome sequencing. Both TmTLP1 and TmTLP2 genes contain 729 nucleotide sequences encoding 242 amino acid residues, including an initiation codon (ATG) and a termination codon (TAA). Interestingly, TmTLPs are proteins with 14 central cysteine residues that may have an important role in structure formation. These data will be used to characterize the innate immune functions of TmTLPs in Tenebrio molitor.  相似文献

To explore an effective and reliable karyotyping method in Brassica crop plants, Cot-1 DNA was isolated from Brassica oleracea genome, labeled as probe with Biotin-Nick Translation Mix kit, in situ hybridized to mitotic spreads, and where specific fluorescent bands showed on each chromosome pair. 25S and 5S rDNA were labeled as probes with DIG-Nick Translation Mix kit and Biotin-Nick Translation Mix kit, respectively, in situ hybridized to mitotic preparations, where 25S rDNA could be detected on two chromosome pairs and 5S rDNA on only one. Cot-1 DNA contains rDNA and chromosome sites identity between Cot-1 DNA and 25S rDNA was determined by dual-colour fluorescence in situ hybridization. All these showed that the karyotyping technique based on a combination of rDNA and Cot-1 DNA chromosome landmarks is superior to all but one. A more exact karyotype of B. oleracea has been analyzed based on a combination of rDNA sites, Cot-1 DNA fluorescent bands, chromosome lengths and arm ratios. __________ Translated from Journal of Wuhan University (Nat.
