油茶子叶体细胞胚形成的细胞学观察   总被引:3,自引:1,他引:2  
以油茶子叶块为外植体,在附加2.0mg·L-1 2,4-D和1.0mg·L-1KT的MS培养基上进行培养,观察诱导产生的体细胞胚性愈伤组织细胞学结构.结果表明:油茶体细胞胚可直接起源于表皮或近表皮的单细胞原胚或者多细胞团.其中,单细胞原胚先分裂形成二细胞原胚,二细胞原胚再进一步分裂后聚集形成多细胞团,最终经过球形胚、梨形胚、心形胚发育形成一个完整的体细胞胚.  相似文献

社会性动物的个性特征是对环境和社群的适应性结果,对个性的研究是探究群居动物复杂社会关系及其差异的重要内容。相对于个性特征评级法和行为编码法,自然观察法是分析判断野生动物个性特征较为理想的方法之一,最大限度地突出了野生动物的自然属性及个性特征类型。本研究以栖息于安徽黄山的野生短尾猴投食群体鱼鳞坑A1群为研究对象,采用目标动物取样法和全事件记录法,于2015年7月至2016年6月期间采集成年个体自然发生的行为数据,并采用主成分分析方法判断成年个体的个性特征及类型,并分析性别差异。结果表明:成年雄性个体具有5种个性特征,即领导型、内向型、社交型、孤僻型以及受欺凌型,成年雌性个体具有6种个性特征,即领导型、孤僻型、内向型、不安型、神经质型以及友好型。研究结果与已有结果一致,说明自然观察法可能是分析野生动物个性类型的较好 选择,同时,不同性别之间个性类型的差异性进一步显示了野生动物自然属性。本研究为深入理解动物个体水平上的差异性及适应性提供了科学依据。  相似文献

在等级森严的非人灵长类群体中,睾酮对雄性顺位维持有着重要作用,且影响着体内寄生虫种类和荷虫量。本研究以栖息于安徽黄山的雄性短尾猴为研究对象,通过计算成年雄性的等级顺位,测定其粪便中睾酮含量以及寄生虫的荷虫量,探讨雄性等级顺位、睾酮水平与健康间的平衡关系。结果表明:共检测出10种肠道寄生虫,包括4种蠕虫、5种原虫和1种螨虫;其中雄性短尾猴个体间睾酮水平(1.00±0.42) ng/g和肠道寄生虫荷虫量(112.44±83.62)EPG均存在显著差异(Kruskal-Wallis H,P<0.05);等级顺位与睾酮水平呈正相关(Spearman, N=8,ρ= 0.326, P<0.05),与肠道原虫类的荷虫量呈负相关(Spearman, N=8,ρ= - 0.345, P<0.05),与肠道蠕虫类的荷虫量无相关性(Spearman, N=8,ρ= 0.065, P>0.05);睾酮水平与肠道原虫类寄生虫的荷虫量呈负相关(Spearman, N=8,ρ= -0.546, P<0.05),与肠道蠕虫类的荷虫量无相关性(Spearman, N=8,ρ= -0.013, P>0.05)。本研究表明高顺位雄性拥有更高的睾酮水平,这不仅对其顺位的维持具有重要作用,对原虫类寄生虫也具有明显的抑制作用。  相似文献

Background

Metabolic reprogramming and hypoxia contribute to the resistance of conventional chemotherapeutic drugs in kinds of cancers. In this study, we investigated the effect of dihydrotanshinone I (DHTS) on reversing dysregulated metabolism of glucose and fatty acid in colon cancer and elucidated its mechanism of action.

Methods

Cell viability was determined by MTT assay. Oxidative phosphorylation, glycolysis, and mitochondrial fuel oxidation were assessed by Mito stress test, glycolysis stress test, and mito fuel flex test, respectively. Anti-cancer activity of DHTS in vivo was evaluated in Colon cancer xenograft. Hexokinase activity and free fatty acid (FFA) content were assessed using respective Commercial kits. Gene expression patterns were determined by performing DNA microarray analysis and real-time PCR. Protein expression was assessed using immunoblotting and immunohistochemistry.

Results

DHTS showed similar cytotoxicity against colon cancer cells under hypoxia and normoxia. DHTS decreased the efficiency of glucose and FA as mitochondrial fuels in HCT116 cells, which efficiently reversed by VO-OHpic trihydrate. DHTS reduced hexokinase activity and free fatty acid (FFA) content in tumor tissue of xenograft model of colon cancer. Gene expression patterns in metabolic pathways were dramatically differential between model and treatment group. Increases in PTEN and a substantial decrease in the expression of SIRT3, HIF1α, p-AKT, HKII, p-MTOR, RHEB, and p-ACC were detected.

Conclusions

DHTS reversed metabolic reprogramming in colon cancer through PTEN/AKT/HIF1α-mediated signal pathway.

General significance

The study is the first to report the reverse of metabolic reprogramming by DHTS in colon cancer. Meantime, SIRT3/PTEN/AKT/HIF1α mediated signal pathway plays a critical role during this process.  相似文献

Supported metal nanoparticles are the most widely investigated heterogeneous catalysts in catalysis community. The size of metal nanostructures is an important parameter in influencing the activity of constructed catalysts. Especially, as coordination unsaturated metal atoms always work as the catalytically active centers, decreasing the particle size of the catalyst can greatly boost the specific activity per metal atom. Single‐atom catalysts (SACs), containing single metal atoms anchored on supports, represent the utmost utilization of metallic catalysts and thus maximize the usage efficiency of metal atom. However, with the decreasing of particle size, the surface free energy increases obviously, and tends to aggregate into clusters or particles. Selection of an appropriate support is necessary to interact with isolated atoms strongly, and thus prevents the movement and aggregation of isolated atoms, creating stable, finely dispersed active sites. Furthermore, with uniform single‐atom dispersion and well‐defined configuration, SACs afford great space for optimizing high selectivity and activity. In this review, a detailed discussion of preparing, characterizing, and catalytically testing within this family is provided, including the theoretical understanding of key aspects of SACs materials. The main advantages of SACs as catalysts and the challenges faced for further improving catalytic performance are also highlighted.  相似文献

Splenic erythroblasts were obtained from mice during the acute disease caused by anemia-inducing virus (FVA). They were cultured in a medium containing EPO, BSA and so on. We studied their biomechanical and hemorheological behavior after 12 h, 24 h and 48 h. The results showed obvious changes in their electrophoretic mobility, osmotic fragility, membrane fluidity and membrane viscoelastic properties with the development of erythroblast. The changes were associated with the interactions of the protein and lipid and the elements of the membrane.  相似文献

Nucleic acids can quench resonance light scattering (RLS) intensity of the Y(III)-1,6-bi(1'-phenyl-3'-methyl-5'-pyrazolone-4'-)hexane-dione(BPMPHD) complex in the pH range 5.0-5.8. Under optimal conditions, there are linear relationships between the quenching of RLS and the concentration of nucleic acids in the range 6.3 x 10(-8)-2.1 x 10(-5) g/mL for fish sperm DNA (fsDNA), 1.2 x 10(-8)-5.0 x 10(-5) g/mL for calf thymus DNA (ctDNA) and 6.0 x 10(-8)-2.0 x 10(-5) g/mL for yeast RNA (yRNA). The detection limits (3 s) of fsDNA, ctDNA and yRNA are 0.7 ng/mL, 3.8 ng/mL and 4.2 ng/mL, respectively.  相似文献

The β-agarase C gene (agaC) of a marine bacterium, Vibrio sp. strain PO-303, consisted of 1,437 bp encoding 478 amino acid residues. β-Agarase C was identified as the first β-agarase that cannot hydrolyze neoagarooctaose and smaller neoagarooligosaccharides and was assigned to a novel glycoside hydrolase family.  相似文献
