The A2A adenosine receptor (A2AR) is a G‐protein–coupled receptor. We previously reported that the C terminus of the A2AR binds to translin‐associated protein X (TRAX) and modulates nerve growth factor (NGF)‐evoked neurite outgrowth in PC12 cells. Herein, we show that neuritogenesis of primary hippocampal neurons requires p53 because blockage of p53 suppressed neurite outgrowth. The impaired neuritogenesis caused by p53 blockage was rescued by activation of the A2AR (designated the A2A rescue effect) in a TRAX‐dependent manner. Importantly, suppression of a TRAX‐interacting protein (kinesin heavy chain member 2A, KIF2A) inhibited the A2A rescue effect, whereas overexpression of KIF2A caused a rescue effect. Expression of a KIF2A fragment (KIF2A514), which disturbed the interaction between KIF2A and TRAX, blocked the rescue effect. Transient colocalization of TRAX and KIF2A was detected in the nucleus of PC12 cells upon NGF treatment. These data suggest that functional interaction between KIF2A and TRAX is critical for the A2A rescue effect. Moreover, p53 blockage during NGF treatment prevented the redistribution of KIF2A from the nucleus to the cytoplasmic region. Expression of a nuclear‐retained KIF2A variant (NLS‐KIF2A) did not rescue the impaired neurite outgrowth as did the wild‐type KIF2A. Therefore, redistribution of KIF2A to the cytoplasmic fraction is a prerequisite for neurite outgrowth. Collectively, we demonstrate that KIF2A functions downstream of p53 to mediate neuritogenesis of primary hippocampal neurons and PC12 cells. A new family of diphenylpyrimidine derivatives bearing an amino acid substituent were identified as potent BTK inhibitors. Among them, compound 7b, which features an l-proline substituent, was identified as the strongest BTK inhibitor, with an IC50 of 8.7?nM. Compound 7b also displayed similar activity against B-cell lymphoma cell lines as ibrutinib. Moreover, 7b exhibited low cytotoxic activity against normal PBMC cells. In addition, the acridine orange/ethidium bromide (AO/EB) staining assay, Western blot analysis and flow cytometry analysis also showed its effectiveness in interfering with B-cell lymphoma cell growth. The molecular simulation performance showed that 7b forms additional strong hydrogen bonds with the BTK protein. All these findings provided new clues about the pyrimidine scaffold as an effective BTK inhibitor for the treatment of B-cell lymphoma.  相似文献

The seemly paradoxical Gq agonist-stimulated phosphoinositide production has long been known, but the underlying mechanism and its physiological significance are not known. In this study, we studied cardiac phosphoinositide levels in both cells and whole animals under the stimulation of norepinephrine (NE), angiotensin II (Ang II), and other physiologically relevant interventions. The results demonstrated that activation of membrane receptors related to NE or Ang II caused an initial increase and a later fall in phosphatidylinositol 4,5-bisphosphate (PIP2) levels in the primary cultured cardiomyocytes from adult rats. The possible mechanism underlying this increase in PIP2 was found to be through an enhanced activity of phosphatidylinositol 4-kinase IIIβ, which was mediated by an up-regulated interaction between phosphatidylinositol 4-kinase IIIβ and PKC; the increased activity of phosphatidylinositol 4-phosphate 5-kinase γ was also involved for NE-induced increase of PIP2. When the systolic functions of the NE/Ang II-treated cells were measured, a maintained or failed contractility was found to be correlated with a rise or fall in corresponding PIP2 levels. In two animal models of cardiac hypertrophy, PIP2 levels were significantly reduced in hypertrophic hearts induced by isoprenaline but not in those induced by swimming exercise. This study describes a novel mechanism for phosphoinositide metabolism and modulation of cardiac function.  相似文献

构建GST/金黄色葡萄球菌分选酶A (SrtA)的原核表达载体,在大肠杆菌中表达、纯化分选酶,并利用展示在酵母表面的底物检测分选酶的活性.以pMD20-SrtA为模板,PCR扩增得到SrlA△N59基因,经BamH I和Xho I双酶切,连接到原核表达栽体pGEX-4T-1中,构建重组表达栽体pGEX-SrtA△N59,转化大肠杆菌BL21( DE3),IPTG诱导表达,GST亲和层析分离纯化得到SrtA△N59,与展示在酵母表面的底物序列QALPETGEE-linker-EGFP作用,产生游离的EGFP,通过酶标仪检测EGFP荧光强度确定分选酶的活性.结果显示,重组表达栽体pGEX-SrtA△N59经IPTG诱导,表达出相对分子质量约为42 kD的融合蛋白,SDS-PAGE分析,该融合蛋白是以可溶形式表达.分离纯化得到的分选酶与底物作用,其荧光强度由568.66±12.14增加至921.43±13.02.以上结果表明,成功构建了重组表达裁体pGEX-SrtA△N59,并在大肠杆菌中获得了可溶表达的有活性的分选酶.  相似文献

用等电聚焦电泳分析的方法,测定了小麦(Triticum aestivum )3 种细胞质雄性不育类型(A 型、E型、T型)及其相应同核保持系萌动胚及幼芽可溶性蛋白. 发现雄性可育系等电点(pI)为4.90 的蛋白质合成数量高于相应的不育系;pI为6.85 的蛋白质可能是T 型细胞质基因表达的结果;pI为7.6 的蛋白质可能为津丰A 不育系特有的区带.表明细胞质来源不同的不育类型,其萌动胚及幼芽可溶性蛋白等电聚焦电泳图谱差异明显,有可能作为鉴别它们的依据  相似文献

供试13种化学物质中,12种物质对Bt制剂的杀虫效果有增效作用,各物质的不同质量浓度对杀虫效果影响差异显,在1g/L的剂量以下,多数添加剂的增效作用,随剂量的增加而增强,添加0.5g/L巯基乙醇增效作用最佳,增效倍数达3.61倍,0.5g/L的甲酸和尿素,及0.1g/L碳酸钾具有较好的增效作用,增效倍数分别为2.52,2.32和2.56和2.56倍,0.5g/L的马来酸,氯化钙,乙二胺四乙酸,氯化镁,硼酸,碳酸钠和1g/L的柠檬酸,磷酸氢二钾8种添加剂的增效倍数为0.53-1.88倍。  相似文献

本文旨在探讨大鼠新鲜离体输精管平滑肌细胞中乙酰胆碱(acetylcholine,ACh)引起超极化反应的机制,采用细胞内微电极记录技术和细胞内荧光标记技术研究ACh对大鼠输精管不同走行方向平滑肌细胞的作用。用尖端含0.1%碘化吡啶(propidium iodide,PI)的记录电极标记电生理记录后的平滑肌细胞,其中37个为外层纵行细胞,17个为内层环行细胞。它们的平均静息膜电位分别为(-53.56±3.88)mV和(-51.62±4.27)mV,膜输入阻抗分别为(2245.60±372.50)MQ和(2101.50±513.50)MQ。ACh引起的膜超极化反应是浓度依赖性的,EC50为36 μmol/L。ACh引起的超极化反应可被非选择性的毒草碱(muscarinic receptor,M)受体阻断剂阿托品(atropine,1 μmol/L)和选择性的M3受体阻断剂diphenylacetoxy-N-methylpiperidine-methiodide(DAMP,100nmol/L)阻断。ACh引起的超极化还能被一氧化氮合酶抑制剂L-硝基-精氨酸甲酯(N-nitro-L-arginine methylester,L.NAME,300μmol/L)阻断,并可被ATP敏感的钾通道阻断剂glipizide(5μmol/L)或内向整流钾通道阻断剂钡离子(50μmol/L)部分阻断。Glipizide和钡离子联合使用可完全阻断ACh引起的超极化反应。上述结果表明:ACh通过作用于大鼠输精管平滑肌细胞膜上的M3受体引起超极化反应,一氧化氮、ATP敏感性钾通道和内向整流钾通道参与了ACh引起的超极化反应。  相似文献

结直肠癌(Colorectal cancer,CRC)是世界第三大癌症,涉及因素众多,而肠道菌群失调、菌群致病性与结直肠癌的发生、发展有着密不可分的关系。近期的研究发现具核梭杆菌(Fusobacterium nucleatum,F.nucleatum)与结直肠癌的发生存在显著的相关性。具核梭杆菌为革兰阴性厌氧菌,广泛存在于人体肠道中。分子机制研究发现,其能影响宿主细胞因子水平,促进血管生成、巨噬细胞M2极化和抑制机体免疫调节能力,进而增强肿瘤细胞增殖、侵袭及转移。本文就具核梭杆菌与结直肠癌相关机制研究进展作一综述。  相似文献

Aphids, the largest group of sap-sucking pests, cause significant yield losses in agricultural crops worldwide every year. The massive use of pesticides to combat this pest causes severe damage to the environment, putting in risk the human health. In this study, transgenic potato plants expressing Galanthus nivalis agglutinin (GNA) gene were developed using CaMV 35S and ST-LS1 promoters generating six transgenic lines (35S1-35S3 and ST1-ST3 corresponding to the first and second promoter, respectively). Quantitative real-time polymerase chain reaction (qRT-PCR) analysis indicated that the GNA gene was expressed in leaves, stems and roots of transgenic plants under the control of the CaMV 35S promoter, while it was only expressed in leaves and stems under the control of the ST-LS1 promoter. The levels of aphid mortality after 5 days of the inoculation in the assessed transgenic lines ranged from 20 to 53.3%. The range of the aphid population in transgenic plants 15 days after inoculation was between 17.0 ± 1.43 (ST2) and 36.6 ± 0.99 (35S3) aphids per plant, which corresponds to 24.9–53.5% of the aphid population in non-transformed plants. The results of our study suggest that GNA expressed in transgenic potato plants confers a potential tolerance to aphid attack, which appears to be an alternative against the use of pesticides in the future.  相似文献
