There is now mounting evidence that the aryl hydrocarbon receptor (AhR) plays an important role in physiologic responses such as development, cell cycle regulation, immune function and also malignant transformation in various tissues. The strong nuclear AhR expression is observed in the invasive phenotype, and an elevated nuclear AhR expression is associated with a poor prognosis of human prostate cancer. On the other hand, there are conflicting results that the AhR deficiency results in increased susceptibility to prostate tumors in mouse model. In the present study, we investigated AhR expression and its role in the growth and invasiveness of human prostate cancer cells. The AhR protein expression was detected in prostate cancer cell lines and human prostate cancer tissues. A small interfering RNA targeting AhR, constitutive active AhR expression vector, and AhR agonist and antagonist were used to moderate its expression and signaling. The induction of AhR signaling attenuated invasiveness of prostate cancer cells without affecting the cellular growth rate. These results suggest that AhR signaling in prostate cancer cells facilitates invasion of these cells, and modulation with this signaling can be a potential therapeutic target of invasive tumors.  相似文献

VX-680, also known as MK-0457, is an ATP-competitive small molecule inhibitor of the Aurora kinases that has entered phase II clinical trials for the treatment of cancer. We have solved the cocrystal structure of AurA/TPX2/VX-680 at 2.3 A resolution. In the crystal structure, VX-680 binds to the active conformation of AurA. The glycine-rich loop in AurA adopts a unique bent conformation, forming a pi-pi interaction with the phenyl group of VX-680. In contrast, in the published AurA/VX-680 structure, VX-680 binds to AurA in the inactive conformation, interacting with a hydrophobic pocket only present in the inactive conformation. These data suggest that TPX2, a protein cofactor, can alter the binding mode of VX-680 with AurA. More generally, the presence of physiologically relevant cofactor proteins can alter the kinetics, binding interactions, and inhibition of enzymes, and studies with these multiprotein complexes may be beneficial to the discovery and optimization of enzyme inhibitors as therapeutic agents.  相似文献

An enzyme-coupled colorimetric assay for S-adenosylmethionine-dependent methyltransferase (MT) was established to characterize the enzymatic identity of YabC protein from Escherichia coli. Results showed that the MT activity of YabC is able to be effectively detected with this coupling assay system when filamentous cell lysates from an S-adenosylmethionine synthetase mutant, MEW402metK84, were employed as the source of methylation target, whereas no activity exhibited under same conditions from lysates from E. coli normal shape cells. Moreover, YabB, whose gene always clustered together with yabC, was demonstrated to be an inhibitor, but not a substrate of YabC protein. Analysis of each component's effect on methylation process further confirms that increase of MT's common product - S-adenosylhomocysteine - shows a methylation pattern depending on the protein concentration of cell lysates, which indicates that methylation target of YabC is the rate-limiting factor under current coupling assay conditions. Characterization of YabC's MT properties with the coupling assay system is consistent with the similar results identified with LC/MS and knockout strains by Kimura and Suzuki [Nucleic Acids Res 2010;38:1341-1352].  相似文献

乳酸氧化酶能够氧化乳酸生成丙酮酸,由于反应过程中不需要外源辅酶作为电子受体,而具有较好的应用前景。乳酸氧化酶和许多黄素蛋白酶相比较,具有明显的共性,因此可视为黄素蛋白家族中的一员。乳酸单加氧酶的催化机理和乳酸氧化酶相似,但产物不同,这主要是由于中间产物复合体稳定性的差别。乳酸单加氧酶催化形成的中间复合体EFMNH2pyruvate很稳定,在氧的作用下,生成EFMNpyruvateH2O2中间体,继续反应形成乙酸,CO2和H2O;乳酸氧化酶生成的EFMNH2pyruvate复合体不稳定,丙酮酸很快从复合体上分离下来,还原型中间体EFMNH2被氧氧化,同时形成过氧化氢。  相似文献

一株粘质沙雷氏菌烈性噬菌体污水分离及特性   总被引:1,自引:0,他引:1  
[目的]以粘质沙雷氏菌(8039)为宿主菌从医院污水中分离噬菌体并对其基本生物学特点进行研究.[方法]四步法污水分离噬菌体;单、双层平板噬菌斑实验筛选烈性噬菌体并观察噬菌斑形态;纯化后2%磷钨酸染色电镜观察;手工法提取噬菌体核酸酶切后琼脂糖凝胶电泳分析;利用双层平板噬菌斑实验测定最佳感染复数和完成一步生长实验.[结果]从医院污水中成功分离出粘质沙雷氏菌烈性噬菌体一株(SM701),该噬菌体有一个正多面体立体对称的头部,头径约64nm,无囊膜,有一长尾,无收缩尾鞘,尾长约143nm;基因组核酸能被双链DNA内切酶BamH Ⅰ及Hind Ⅲ切开,大小约57kb;噬菌斑圆形透明,直径1mm左右(培养12h,),边界清楚;当感染复数(multiplicity of infection,MOI)为10时,子代噬菌体滴度较高;按照一步生长实验结果绘制出一步生长曲线,可知感染宿主菌的潜伏期是约为30min,爆发期约100min,平均爆发量约为630[结论]按照国际病毒分类委员会分类标准,该噬菌体属于长尾噬菌体科(siphoviridae)烈性噬菌体,按照Bradley和Ackermann形态分类法属于B1亚群;噬菌斑与周围红色细菌生长区,颜色差异明显,非常便于观察和计数;噬菌体头部大小和形态与呼吸道病毒中的呼肠病毒和腺病毒最为接近;国内尚未见粘质沙雷氏菌噬菌体相关报道.  相似文献

采用碱提酸沉法从野皂荚提胶后的副产物中制备分离蛋白,研究了制备工艺条件对分离蛋白得率的影响。结果表明:当料液比1∶10~1∶12(m/V)、提取液pH8.0~9.0、浸提温度45~50℃、浸提两次、每次浸提45 m in时,分离蛋白得率可达78.45%。研究结果可为野皂荚提胶副产物中蛋白的综合利用奠定了基础。  相似文献

Despite the recent unprecedented development of efficient dopant‐free hole transporting materials (HTMs) for high‐performance perovskite solar cells (PSCs) on small‐area devices (≤0.1 cm2), low‐cost dopant‐free HTMs for large‐area PSCs (≥1 cm2) with high power conversion efficiencies (PCEs) have rarely been reported. Herein, two novel HTMs, 3,3′,6,6′ (or 2,2′,7,7′)‐tetrakis(N,N′‐di‐p‐methoxyphenylamine)‐N,N′‐bicarbazole (3,6 BCz‐OMeTAD or 2,7 BCz‐OMeTAD), are synthesized via an extremely simple route from very cheap raw materials. Owing to their excellent film‐forming abilities and matching energy levels, 3,6 BCz‐OMeTAD and 2,7 BCz‐OMeTAD can be successfully employed as a perfect ultrathin (≈30 nm) hole transporting layer in large‐area PSCs up to 1 cm2. The 3,6 BCz‐OMeTAD and 2,7 BCz‐OMeTAD based large‐area PSCs show highest PCEs up to 17.0% and 17.6%, respectively. More importantly, high performance large‐area PSCs based on 2,7 BCz‐OMeTAD retain 90% of the initial efficiency after 2000 h storage in an ambient environment without encapsulation.  相似文献

Contractile activation in striated muscles requires a Ca2+ reservoir of large capacity inside the sarcoplasmic reticulum (SR), presumably the protein calsequestrin. The buffering power of calsequestrin in vitro has a paradoxical dependence on [Ca2+] that should be valuable for function. Here, we demonstrate that this dependence is present in living cells. Ca2+ signals elicited by membrane depolarization under voltage clamp were compared in single skeletal fibers of wild-type (WT) and double (d) Casq-null mice, which lack both calsequestrin isoforms. In nulls, Ca2+ release started normally, but the store depleted much more rapidly than in the WT. This deficit was reflected in the evolution of SR evacuability, E, which is directly proportional to SR Ca2+ permeability and inversely to its Ca2+ buffering power, B. In WT mice E starts low and increases progressively as the SR is depleted. In dCasq-nulls, E started high and decreased upon Ca2+ depletion. An elevated E in nulls is consistent with the decrease in B expected upon deletion of calsequestrin. The different value and time course of E in cells without calsequestrin indicate that the normal evolution of E reflects loss of B upon SR Ca2+ depletion. Decrement of B upon SR depletion was supported further. When SR calcium was reduced by exposure to low extracellular [Ca2+], release kinetics in the WT became similar to that in the dCasq-null. E became much higher, similar to that of null cells. These results indicate that calsequestrin not only stores Ca2+, but also varies its affinity in ways that progressively increase the ability of the store to deliver Ca2+ as it becomes depleted, a novel feedback mechanism of potentially valuable functional implications. The study revealed a surprisingly modest loss of Ca2+ storage capacity in null cells, which may reflect concurrent changes, rather than detract from the physiological importance of calsequestrin.  相似文献
