A procedure was developed for plant regeneration of Hybanthus enneaspermus, a rare ethnobotanical herb from the Deccan peninsula in India, through seed-derived callus. Seeds demonstrated a high induction
frequency (69.4±2.8%) and a high yield (364.4±2.5 mg) of light-yellow friable callus on Murashige and Skoog's (MS) medium
containing 2.6 μm NAA and 2.2 μm BA within 4 weeks of incubation. After 1 year of subculture, yellow friable and light-green compact calli types were established
from initial light-yellow friable callus. Shoot differentiation was achieved from light-green compact callus, but not from
yellow friable callus. Shoot differentiation resulted when light-green compact callus was transferred to MS medium supplemented
with 8.8 μm BA and 2.6 μm NAA; the highest percentage of calli forming shoots (66.6±4.8%) and the highest number of shoots (8.9±0.3) were achieved
in this medium. Differentiated shoot buds elongated to 4–5 cm within 4 weeks. The addition of casein hydrolysate (500 mg/l)
and more potassium phosphate (1.86 mm) to the culture medium enhanced shoot differentiation. Rooting was achieved on the shoots using half-strength MS medium containing
4.8 μm IBA. About 70% of the plants were established in pots containing pure garden soil after 2 weeks of hardening. The regenerated
plants were morphologically uniform and exhibited normal seed set.

金属蛋白酶组织抑制因子-4(TIMP-4)是TIMP家族的最新成员。已有研究表明,TIMP-4mRNA大量表达于成年小鼠的睾丸中。为了证实TIMP-4基因在出生后小鼠睾丸中的表达是否具有发育依赖性,本实验利用RT-PCR、Western blotting和间接免疫荧光染色三种方法,分别检测了TIMP-4mRNA和蛋白在出生后小鼠睾丸不同发育期中的时空表达方式。RT-PCR和Western blotting结果分别显示,TIMP-4mRNA和蛋白均只在成年小鼠睾丸中表达,而在出生后的其它各阶段都不表达;间接免疫荧光染色进一步证实TIMP-4蛋白只定位在成年小鼠睾丸的Leydig细胞中。结果提示,TIMP-4在出生后小鼠睾丸中的表达具有显著的发育依赖性. 相似文献

MicroRNAs (miRNAs) may regulate diverse biological processes and play an important role in cancer. And MiRNAs have been proposed as a useful tool for lung cancer diagnosis and therapeutics in cancer. The purpose of the present study was to investigate the association among the expression level of mature miR-200b-5p in peripheral blood and the risk of lung cancer and clinic pathological characteristics. This case-control study included 24 patients with lung cancer and 12 healthy controls. MiR-200b expression was deleted using real-time PCR. and the miR-200b expression of normal controls was significantly higher than that in lung cancer patients (1732.13?pg/mL vs 881.67?pg/mL, P?<?0.05), no difference with age, sex, tissue type and clinical stage of lung cancer patients (P?>?0.05). Furthermore, miR-200b expression level fluctuated with tumor progression in lung cancer, and there was highly significant for clinical stage II compared with the clinical stage III (P?<?0.05). In addition, the down-regulation of miR-200b showed a highly discriminative receiver operating characteristic (ROC) curve profile, clearly distinguishing cancer patients from cancer-free subjects with an area under the ROC curve (AUROC) of 0.87. The detection of miR-200b expression yielded 83.30% sensitivity and 100.00% specificity in the diagnosis of lung cancer. Therefore, these findings suggested that miR-200b may be used as a marker for the detection and diagnosis of lung cancer in peripheral blood. 相似文献

To investigate the roles of the active site residues in the catalysis of Bacillus thuringiensis WB7 chitinase, twelve mutants, F201L, F201Y, G203A, G203D, D205E, D205N, D207E, D207N, W208C, W208R, E209D and E209Q were constructed by site-directed mutagenesis. The results showed that the mutants F201L, G203D, D205N, D207E, D207N, W208C and E209D were devoid of activity, and the loss of the enzymatic activities for F201Y, G203A, D205E, W208R and E209Q were 72, 70, 48, 31 and 29%, respectively. The pH-activity profiles indicated that the optimum pH for the mutants as well as for the wildtype enzyme was 8.0. E209Q exhibited a broader active pH range while D205E, G203A and F201Y resulted in a narrower active pH range. The pH range of activity reduced 1 unit for D205E, and 2 units for G203A and F201Y. The temperature-activity profiles showed that the optimum temperature for other mutants as well as wildtype enzyme was 60°C, but 50°C for G203A, which suggested that G203A resulted in a reduction of thermostability. The study indicated that the six active site residues involving in mutagenesis played an important part in WB7 chitinase. In addition, the catalytic mechanisms of the six active site residues in WB7 chitinase were discussed. 相似文献

为了解黄土高原不同植被土壤微量元素的根际效应,分析了该地区柠条、沙棘、沙打旺、柳枝稷、阿尔泰狗娃花和茵陈蒿6种植被根际与非根际土壤中有机碳、全氮、Mn、Cu、Fe、Zn含量.结果表明: 6种植被中,柠条、阿尔泰狗娃花和茵陈蒿根际土壤的有机碳、全氮含量高于非根际土壤;除柠条和沙棘外,其余4种植被非根际土壤pH值均显著高于根际土壤.6种植被根际土壤有效Mn含量均低于非根际;柠条、沙打旺和柳枝稷根际有效Cu含量显著高于非根际,表现出强烈根际富集现象.除沙打旺外,其他5种植被根际有效Fe含量均略高于非根际.沙打旺、柳枝稷、茵陈蒿和阿尔泰狗娃花表现出强烈的有效Zn根际富集现象.根际与非根际土壤有机碳、全氮与有效Mn、有效Zn,以及有效Mn与有效Zn呈极显著正相关.根际土壤pH值与有效Mn和有效Zn呈显著负相关.由于不同植物根系的生长特征、根际pH值及微生物种类等的差异,不同植被根际的微量元素含量不同,茵陈蒿根际4种微量元素含量高于其他植被. 相似文献

目的:利用植物悬浮细胞体系对莪二酮进行结构改造研究.方法:采用生物转化技术和天然药物化学手段,分离转化产物单体,并利用波谱学手段对转化产物进行结构鉴定,并利用MTT法对转化产物的抗肿瘤活性进行了评价.结果:分离并鉴定了5个转化产物,分别为1β,10α-环氧基莪二酮(2),3α-羟基-莪二酮(3),3β-羟基-莪二酮(4),1α,10β-环氧基-11-羟基莪二酮(5)和2β-羟基-莪二酮(6).结论:桔梗悬浮细胞对于莪二酮具有良好的转化能力,可以利用其作为植物反应器对莪二酮进行结构改造,以获得水溶性更好或活性更佳的衍生物. 相似文献

Background
Intestinal bacteria are known to regulate bile acid (BA) homeostasis via intestinal biotransformation of BAs and stimulation of the expression of fibroblast growth factor 19 through intestinal nuclear farnesoid X receptor (FXR). On the other hand, BAs directly regulate the gut microbiota with their strong antimicrobial activities. It remains unclear, however, how mammalian BAs cross-talk with gut microbiome and shape microbial composition in a dynamic and interactive way.Results
We quantitatively profiled small molecule metabolites derived from host-microbial co-metabolism in mice, demonstrating that BAs were the most significant factor correlated with microbial alterations among all types of endogenous metabolites. A high-fat diet (HFD) intervention resulted in a rapid and significant increase in the intestinal BA pool within 12 h, followed by an alteration in microbial composition at 24 h, providing supporting evidence that BAs are major dietary factors regulating gut microbiota. Feeding mice with BAs along with a normal diet induced an obese phenotype and obesity-associated gut microbial composition, similar to HFD-fed mice. Inhibition of hepatic BA biosynthesis under HFD conditions attenuated the HFD-induced gut microbiome alterations. Both inhibition of BAs and direct suppression of microbiota improved obese phenotypes.Conclusions
Our study highlights a liver–BA–gut microbiome metabolic axis that drives significant modifications of BA and microbiota compositions capable of triggering metabolic disorders, suggesting new therapeutic strategies targeting BA metabolism for metabolic diseases.999.

To examine the bacterial community structure in the Fildes Peninsula, King George Island, Antarctica, we examined the bacterial diversity and community composition of samples collected from lacustrine sediment, marine sediment, penguin ornithogenic sediments, and soils using culture-dependent and culture-independent methods. The 70 strains fell into five groups: Actinobacteria, Bacteroidetes, Firmicutes, Gammaproteobacteria, and Betaproteobacteria. Bacterial diversity at the phylum level detected in Denaturing Gradient Gel Electrophoresis (DGGE) profiles comprised Proteobacteria (including the subphyla Alpha-, Beta-, Gamma-, Deltaproteobacteria), Bacteroidetes, Firmicutes, Chlorobi, and Deinococcus-Thermus. Gammaproteobacteria was identified to be the dominant bacterial subphylum by cultivation and DGGE method. By cluster analysis, the overall structure and composition of bacterial communities in the soil and lacustrine sediment were similar to one another but significantly different from bacterial communities in penguin ornithogenic sediment and marine sediment, which were similar to one another. The majority of 16S rDNA sequences from cultured bacteria were closely related to sequences found in cold environments.
