DNA methylation plays a crucial role in higher organisms. Coupling bisulfite treatment with next generation sequencing enables the interrogation of 5-methylcytosine sites in the genome. However, bisulfite conversion introduces mismatches between the reads and the reference genome, which makes mapping of Illumina and SOLiD reads slow and inaccurate. BatMeth is an algorithm that integrates novel Mismatch Counting, List Filtering, Mismatch Stage Filtering and Fast Mapping onto Two Indexes components to improve unique mapping rate, speed and precision. Experimental results show that BatMeth is faster and more accurate than existing tools. BatMeth is freely available at http://code.google.com/p/batmeth/. 相似文献

Electron paramagnetic resonance (EPR) spectroscopy was used to detect the light-induced formation of singlet oxygen (1O2*) in the intact and the Rieske-depleted cytochrome b6f complexes (Cyt b6f) from Bryopsis corticulans, as well as in the isolated Rieske Fe–S protein. It is shown that, under white-light illumination and aerobic conditions, chlorophyll a (Chl a) bound in the intact Cyt b6f can be bleached by light-induced 1O2*, and that the 1O2* production can be promoted by D2O or scavenged by extraneous antioxidants such as l-histidine, ascorbate, β-carotene and glutathione. Under similar experimental conditions, 1O2* was also detected in the Rieske-depleted Cyt b6f complex, but not in the isolated Rieske Fe–S protein. The results prove that Chl a cofactor, rather than Rieske Fe–S protein, is the specific site of 1O2* formation, a conclusion which draws further support from the generation of 1O2* with selective excitation of Chl a using monocolor red light. 相似文献

持续常温弱光(25℃/18℃,l00umol m-2 s-1)、低温弱光(12℃/12℃,100 umol m-2 s-1和7℃/7℃,l00μmolm-2s-1)均导致黄瓜生长减慢或停滞、叶绿素含量、气孔导度和净光合速率、光合电子传递速率下降以及胞间CO2浓度上升.常温弱光和12℃弱光处理对光系统II的最大光化学效率Fv/Fm无显著影响,而7℃弱光处理导致Fv/Fm的可逆性下降.常温弱光和7℃、12℃弱光处理均导致了光化学反应速率的降低以及天线热耗散和反应中心过剩能量的增加.在胁迫后,12℃弱光0比7℃弱光更有利于植株光合功能的恢复. 相似文献

气孔是植物与环境发生联系的重要门户,控制着植物与外界的气体和水分交换。本文针对全球大气CO2浓度升高、气候变暖、干旱加剧等环境问题,分析了气孔对全球水循环、碳循环的重要贡献。系统总结了气孔的形态发育和生理功能对大气高CO2浓度、干旱、土壤盐渍化、病虫害等的响应及其调控防御机制。综述了脱落酸(ABA)、Ca2+、H2O2、一氧化氮(NO)和光信号调控气孔运动的分子机制。从理论和实践两方面,提出了通过调控气孔运动协调CO2同化和水分散失的矛盾,在不影响光合效率的前提下提高水分利用率等未来的研究方向。 相似文献

Diversity of Crenarchaeota was investigated in eight terrestrial hot springs (pH 2.8–7.7; temperature 44–96°C) located in
Tengchong, China, using 16S rRNA gene phylogenetic analysis. A total of 826 crenarchaeotal clones were sequenced and a total
of 47 operational taxonomic units (OTUs) were identified. Most (93%) of the identified OTUs were closely related (89–99%)
to those retrieved from hot springs and other thermal environments. Our data showed that temperature may predominate over
pH in affecting crenarchaeotal diversity in Tengchong hot springs. Crenarchaeotal diversity in moderate-temperature (59–77°C)
hot springs was the highest, indicating that the moderately hot-temperature springs may provide optimal conditions for speciation

通过构建16S rRNA基因片段的克隆文库对腾冲热海两温泉中泉古菌的多样性和系统发育关系进行了初步的研究.一共得到18个泉古茵克隆序列,可分为12个OTUs,两温泉的克隆序列与已知GenBank上关系最近序列的平均相似性较低,无名泉为92.56%,热爆区为93%.从基于16S rRNA基因片段序列构建的系统发育树来看,74℃的无名泉样点中既有属于超高温环境类群的泉古菌,同时又有属于和常温环境较接近的泉古菌;45℃的热爆区样点的泉古茵,相对来说则更接近于常温类群.本次研究表明,腾冲热泉与世界其它同类热泉之间的泉古茵类群存在着一定的差异;而且两实验样点代表了超高温和高温环境泉古菌逐渐向常温过度的两个重要环境. 相似文献
