The interaction of one anticancer drug (caffeic acid phenethyl ester; CAPE) with three proteases (trypsin, pepsin and α-chymotrypsin) has been investigated with multispectral methods and molecular docking. As an active components in propolis, the findings are of great benefit to metabolism, design, and structural modification of drugs. The results show that CAPE has an obvious ability to quench the trypsin, pepsin, or α-chymotrypsin fluorescence mainly through a static quenching procedure. Trypsin has the largest binding affinity to CAPE, and α-chymotrypsin has the smallest binding affinity to CAPE. The data obtained from thermodynamic parameters and molecular docking prove that the spontaneously interaction between CAPE and each protease is mainly due to a combination of van der Waals (vdW) force and hydrogen bond (H-bond), controlled by an enthalpy-driven process. The binding force, strength, position, and the number of H-bond are further obtained from the results of molecular docking. Through ultraviolet spectroscopy, dynamic light scattering and circular dichroism experiments, the change in the protease secondary structure induced by CAPE was observed. Additionally, the addition of protease had a positive effect on the antioxidative activity of CAPE, and α-chymotrypsin has the greatest effect on the removal of 2,2-diphenyl-1-picrylhydrazyl free radicals by CAPE. 相似文献

This study aims to determine the feasibility of using oligodeoxynucleotides with unmethylated cytosine-guanine dinucleotide sequences (CpG ODN) as an immunity protection strategy for a mouse model of acute respiratory distress syndrome (ARDS). This is a prospective laboratory animal investigation. Twenty-week-old BALB/c mice in Animal research laboratory were randomized into groups. An ARDS model was induced in mice using lipopolysaccharides (LPSs). CpG ODN was intranasally and transrectally immunized before or after the 3rd and 7th days of establishing the ARDS model. Mice were euthanized on Day 7 after the second immunization. Then, retroorbital bleeding was carried out and the chest was rapidly opened to collect the trachea and tissues from both lungs for testing. CpG ODN significantly improved the pathologic impairment in mice lung, especially after the intranasal administration of 50 μg. This resulted in the least severe lung tissue injury. Furthermore, interleukin-6 (IL-6) and IL-8 concentrations were lower, which was second to mice treated with the rectal administration of 20 µg CpG ODN. In contrast, the nasal and rectal administration of CpG ODN in BALB/c mice before LPS immunization did not appear to exhibit any significant protective effects. The intranasal administration of CpG ODN may be a potential treatment approach to ARDS. More studies are needed to further determine the protective mechanism of CpG ODN. 相似文献

Protein kinase monopolar spindle 1 plays an important role in spindle assembly checkpoint at the onset of mitosis. Over expression of MPS1 correlated with a wide range of human tumors makes it an attractive target for finding an effective and specific inhibitor. In this work, we performed molecular dynamics simulations of protein MPS1 itself as well as protein bound systems with the inhibitor and natural substrate based on crystal structures. The reported orally bioavailable 1 h-pyrrolo [3,2-c] pyridine inhibitors of MPS1 maintained stable binding in the catalytic site, while natural substrate ATP could not stay. Comparative study of stability and flexibility of three systems reveals position shifting of β-sheet region within the catalytic site, which indicates inhibition mechanism was through stabilizing the β-sheet region. Binding free energies calculated with MM-GB/PBSA method shows different binding affinity for inhibitor and ATP. Finally, interactions between protein and inhibitor during molecular dynamic simulations were measured and counted. Residue Gly605 and Leu654 were suggested as important hot spots for stable binding of inhibitor by molecular dynamic simulation. Our results reveal an important position shifting within catalytic site for non-inhibited proteins. Together with hot spots found by molecular dynamic simulation, the results provide important information of inhibition mechanism and will be referenced for designing novel inhibitors. 相似文献

Seroprevalence of Bartonella henselae, Toxoplasma gondii, feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV) infections was investigated in 1,447 domestic cats derived from the north (Hokkaido) to the south (Okinawa) prefectures in Japan. Of the cats investigated, 8.8% (128/1,447) were seropositive to B. henselae, 5.4% (78/1,447) to T. gondii, 9.8% (107/1,088) to FIV, and 2.9% (32/1,088) to FeLV, respectively. For B. henselae infection, the positive rate varied from 11.5% in cats of 1 to <2 years old to 7.2% in those over 3 years old. Outdoor cats showed higher positive rate (14.5%) than that (7.0%) in indoor ones. The rate (13.5%) in flea-infested cats was significantly higher than that (7.4%) in flea-negative cats. The positive rates in southern and urban sites were more likely to be higher than those in northern and suburban sites, suggesting that warm and humid environments, density of cat population, and raising status, including hygienic condition and flea infestation in cats may correlate to higher seroprevalence of B. henselae infection. For T. gondii, FIV and FeLV infections, the seroprevalence also tended to be higher in outdoor, flea-infested cats and advanced age groups. For FIV infection, the positive rates in male (14.3%) and outdoor cats (15.0%) were significantly higher than those in female (5.0%) and indoor cats (4.6%). On the other hand, no significant difference in seropositivities was observed in FeLV and T. gondii infections concerning to both genders and raising status. 相似文献

科尔沁左翼后旗位于科尔沁沙地东部农牧交错带,生态环境复杂、脆弱,资源、人口和环境的协调发展至关重要,其土地承载力状况是协调三者关系的重要依据。本文应用系统动力学原理,以1995年为起始年份建立了系统动力学模型(SD模型),包括人口、土地资源、种植业、林业、畜牧业、渔业和消费等7个子系统。选择3种人口增长速度和3种营养水平组成9种方案,通过模型的运行预测了科左后旗未来50年人口的动态变化和最大土地资源承载力,并对1995—2000年实际的人口发展状况和生态状况进行了讨论。研究显示,科左后旗土地承载力46.7l—74.5l万人,不同的方案达到最大承载力的时间不同,指出控制人口增长是实现经济环境协调发展的必要条件,科左后旗的实际人口增长控制较好,现有的环境问题是不合理的开发活动造成的。 相似文献

钠-氢交换蛋白(Na^ -H^ exchangers,NHE)至少包含6个不同的亚型,生长因子可激活其表达。目前,对在发育过程中NHE的表达了解甚少。本文利用RT-PCR观察了4种NHE亚型的mRNA在人胎儿的两个不同发育阶段(11周、16周)在不同组织中的表达,以研究它们的发育调控。结果显示,NHE1 mRNA在两种胎龄的多种组织中均有表达,和16周胚胎相比,11周的胚胎的NHE1 mRNA的表达较弱,并且表现出明显的组织差异。据此推测,NHE1的管家(house-keeping)功能可能至少在11周就开始形成,而最迟在16周已基本建立;NHE2和NHE3 mRNA在11周和16周的胚胎组织中的特异性表达呈现相反的变化趋势及组织分布上的重叠,后者与NHE2和NHE3在成人组织中的分布及功能的重叠的特点相吻合;NHE5 mRNA的表达在11周的胚胎组织中比较普遍,而在16周的胚胎组织中则局限在小脑组织中,本研究表明,在人胚胎发育11-16周期间,NHE的组织特异性表达表现出时间依赖性的调控,而在不迟于胚胎发育的第16周,具有“管家功能”的NHE1的基因表达已与成人相似。 相似文献
