An electrogenic proton pump associated with the Golgi apparatus of mouse liver driven by NADH and ATP

Golgi-apparatus membranes, isolated from mouse liver, pump protons inwards, when supplied with NADH or ATP. The acidification of Golgi-apparatus cisternae and vesicles was detected with neutral red, a permeant dye, as a difference in absorbance at 550 nm minus that at 600 nm. The maximum rates detected with NADH and ATP were between 0.0006-0.0009 and 0.0030-0.0050 delta OD units/mg of protein/min, respectively, at pH 7.5. The outside buffer used was a bovine serum albumin suspension. The acidification of Golgi apparatus was inhibited from 45 to 100% by ionophores and from 22 to 100% by uncouplers. The results implicate both ATP and a redox system coupled to NADH oxidation in the acidification of Golgi-apparatus membranes.     

Effect of gossypol on testicular blood flow and testosterone production in rats

Rats were treated with highly purified gossypol acetic acid at doses of 15 or 30 mg/kg day-1 for 6 weeks to produce an effect on spermatogenesis as shown by reduced sperm motility and increased sperm malformation rates. The treated rats did not differ from the controls in the body weight growth curves and reproductive organ weights. When stimulated with hCG, testicular blood flow was increased in the low dose group; the testosterone concentrations in peripheral and testicular venous blood were also increased to a greater extent than those of the control group. No difference was found between the high dose and control groups in testicular blood flow or testosterone concentrations. The morphology of the Leydig cells was apparently normal, although some degenerative changes in the germinal epithelium were observed in the high dose group. Therefore, there is no evidence in our experiment to show any anti-androgenic effect following 6-week treatment of gossypol in rats, even at the dose of 30 mg/kg day-1.     

油松茎次生结构中树脂道的结构分布和发育的研究

油松茎的次生结构中树脂道存在于次生维管组织中。其中,次生木质部内具有水平的和垂直的两类树脂道,而次生韧皮部内则仅有水平的树脂道。两类树脂道都由上皮细胞和鞘细胞包围着胞间道构成,其中木质部内的树脂道具有死鞘细胞,而韧皮部中的则都系生活细胞。在心材中,垂直树脂道形成拟侵填体。在次生木质部内,垂直树脂道常分布于早材的外部区域和最初形成的晚材中,它们与水平树脂道连接,腔道贯通,从而形成二维网状结构。垂直树脂道来源于纺锤状原始细胞的衍生细胞,而水平树脂道来源于射线原始细胞,两者都以裂生方式发生。     

广谱肾综合征出血热病毒单克隆抗体的A35的生物学性状

具有中和及血凝抑制活性的、能和世界各地分离到的肾综合征出血热病毒(HFRSV)发生反应的、广谱的单克隆抗体(McAb),对HFRSV的诊断和分子生物学研究都有重要意义。 本文着重比较了HFRSV McAbA5、A19、A25-1、A25-7和A35的生物学性状,并观察了对感染动物的实验治疗效果。     

三种核型多角体病毒核酸同源性的研究

用分子杂交技术研究了黑点银纹夜蛾(Arqyrogramma agnata Stgr,)单粒包埋型核型多角体病毒(简称A,A,SNPV),斜纹夜蛾(Prodenia litura)多粒包埋型核型多角体病毒(简称PL MNPV)以及用A,A,SNPV感染斜纹夜蛾幼虫所获得的多粒包埋型核型多角体病毒(暂称PoI MNPV)三种病毒核酸的同源性,用SDS-Tris酚法分别提取病毒核酸,用内切酶Eco RⅠ酶解,比较了病毒核酸的酶解图谱及分子量,用〔α-~(32)P〕dATP标记的三种病毒核酸Eco RⅠ酶解片段作探针,分别与各病毒核酸的Eco RI酶解片段杂交,结果表明PoI MNPV与PL MNPV的病毒核酸同源,而A,A,SNPV DNA不与PoI MNPV DNA、PL MNPV DNA杂交,无同源序列。     

广西德峨苗族、彝族体质调查

对广西隆林县德峨乡男性22至60岁与女性20至60岁的576名苗族人(男395、女181)和178名彝族人(男88、女90)进行了活体观察与测量,计算出各项数据,总结德峨苗族、彝族的体质特征,并与国内一些民族相比较。     

cGMP、cAMP及两种cAMP衍生物对大鼠肝细胞核体外转录系统的影响

 本文介绍了以α-鹅膏蕈碱和低浓度KCl为手段建立了RNA聚合酶Ⅰ、RNA聚合酶Ⅱ活性的细胞核转录系统进而研究了cGMP、cAMP、cAMP丁酯及cAMP硫代环磷酰二乙胺对大鼠肝细胞核中RNA聚合酶Ⅰ与Ⅱ活性的影响。结果显示cGMP可以提高RNA聚合酶Ⅰ的活性;cAMP主要提高RNA聚合酶Ⅱ的活极,而cAMP分子结构变化产生的丁酯及硫代环磷酰二乙胺衍生物可增强cAMP的这种作用,为深入研究cAMP的构效关系提供了实验依据。     

自交作物群体的遗传组成分析

用数学方法推导出了自交作物群体遗传组成的分析公式。根据所推出的公式,可以计算出任何自交世代群体的基因型频率和同一表现型中各基因型的频率。对自交群体遗传组成的分析可用于质量性状的选择,确定选择时需要的有效群体大小和选出有效群体的大小,亦有助于确定选择的最佳世代。     

Isolation and reconstitution of the RNA replicase of the cytoplasmic polyhedrosis virus of silkworm,Bombyx mori

Summary After irradiation of the virus particles of CPV, the RNA replicase associated with the virion was isolated in the form of a genome-replicase complex with DEAE-Sephadex A-25 chromatography. This complex was then treated with Triton X-100 and purified by phosphocellulose column chromatography. The RNA replicase reconstituted with the doublestranded RNA of CPV showed both the enzyme activity of RNA polymerase and methyltransferase. The single-stranded RNA could not serve as the template for the RNA replicase. The role of the RNA replicase of CPV is discussed.     

Somatic embryogenesis and plant regeneration from immature inflorescence segments of Coix lacryma-jobi

Callus was obtained from segments of immature inflorescence of Coix lacryma-jobi cultured on N6 medium containing 1–2 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) and 3–5% sucrose. Plantlets were regenerated when embryogenic calluses were transferred onto MS medium with 0.5 mg/l kinetin and 0.01 mg/l naphthaleneacetic acid (NAA). Regenerated plants had the diploid chromosome number (2n=20).
