The OsGS3 gene plays a principal role in controlling grain weight and grain length in rice. However, the function of an orthologous gene TaGS in wheat has not been analyzed to date. In the present study, we cloned the gDNA of TaGS gene, designated TaGS-D1, with four exons and three introns on chromosome 7DS by a comparative genomics approach. The cDNA of TaGS-D1 is 255 bp, and it encodes 85 amino acids. We also found a plant-specific organ size regulation domain in the deduced polypeptide, indicating that TaGS-D1, like OsGS3, does not belong to the PEBP family. DNA sequencing of the TaGS-D1 locus revealed no diversity in the coding sequence of exons, but there was a single nucleotide polymorphism (SNP) in the first intron, and 30 SNPs, a 40-bp InDel and a 3-bp InDel were found in the second intron between genotypes with higher and lower thousand grain weights (TGW). Based on the 40-bp InDel, a co-dominant STS marker, designated GS7D, was developed to discriminate the two alleles. GS7D was 8.0 cM from Xbarc184 located on chromosome 7DS by linkage mapping. A QTL for TGW and grain length at GS7D locus explained up to 16.3 and 7.7 %, respectively, of the phenotypic variances in a RIL population derived from Doumai/Shi 4185 grown in Shijiazhuang and Beijing. One hundred and seventy-five Chinese wheat cultivars were genotyped with GS7D, indicating that TaGS-D1 was significantly associated with grain weight. The allelic distribution at the TaGS-D1 locus showed that the frequencies of TaGS-D1a were high in cultivars from Serbia, Japan, Australia, Canada, and the Northeastern Spring Wheat and Northern Winter Wheat Regions of China.  相似文献

Purpose

To investigate the vitreous and plasma levels of vascular endothelial growth factor (VEGF) in patients with proliferative diabetic retinopathy (PDR) and to determine whether they predict a disease prognosis after primary vitrectomy.

Methods

Fifty patients (50 eyes) with PDR who underwent pars plana vitrectomy (PPV) and 56 healthy controls (56 eyes) were enrolled in this retrospective study. Clinical data were collected and analyzed. Vitreous and plasma VEGF concentrations were measured using enzyme-linked immunosorbent assays. VEGF levels and clinical data were compared and analyzed to see if they provide a prognosis of PDR progression after primary vitrectomy at more than 6 months follow-up. Correlation of VEGF concentrations between vitreous fluid and plasma was analyzed.

Results

The average BCVA was significantly improved after surgery (P<0.001). Vitreous and plasma VEGF levels were significantly elevated in PDR patients than those in healthy controls (P vitreous<0.001; P plasma<0.001). Both vitreous and plasma VEGF levels were significantly higher in PDR progression group than in stable group (P vitreous<0.001; P plasma = 0.004). Multivariate logistic regression analyses showed that the increased vitreous VEGF level was associated with the progression of PDR after primary PPV (OR = 1.539; P = 0.036). Vitreous VEGF level was positively associated with plasma VEGF level in PDR patients (P<0.001).

Conclusion

The increased VEGF level in vitreous fluid may be identified as a significant predictive factor for the outcome of vitrectomy in patients with PDR.  相似文献

保护区内放牧活动对野生动物保护存在负面影响,明确不同物种对放牧干扰的行为响应对制定更有针对性的保护管理政策具有重要意义。使用红外相机研究卧龙自然保护区放牧活动对多种珍稀野生动物的影响,分析放牧激励政策实施前后大熊猫(Ailuropoda melanoleuca)及其同域分布的小熊猫(Ailurus fulgens)、川金丝猴(Rhinopithecus roxellana)、水鹿(Rusa unicolor) 4种珍稀野生动物的照片数、空间分布以及活动模式的变化,探讨这4种动物对放牧的行为响应策略。结果表明:(1)一期(2012—2013),2012年10月实施了禁马政策,同年12月实施放牧(牛羊)激励政策)家畜照片数量很少,4种野生动物照片数相对较多。二期(2014—2015)家畜的照片数显著增加(P0.01),小熊猫(P0.05)与川金丝猴(P0.01)的照片数均显著减少,大熊猫、水鹿的照片数也呈减少趋势;到三期(2016—2017),大熊猫、小熊猫及水鹿3种关注野生动物的照片数基本回升到激励政策实施前的水平,无川金丝猴照片记录。(2)一期,4种野生动物在研究区域有较广的分布;二期,大熊猫、小熊猫的空间分布范围均缩小,无川金丝猴空间分布信息,而家畜、水鹿的空间分布范围有所增加;到三期,大熊猫、小熊猫的空间分布基本恢复到放牧激励政策实施前的区域,无川金丝猴的空间分布信息。(3)放牧激励政策实施前后,大熊猫、小熊猫及川金丝猴活动模式无明显变化,但水鹿的活动更加集中于傍晚,以避开人类与家畜的活动高峰。同域分布的不同的野生动物对人类活动(如放牧)的行为响应策略不同,各保护区在制定相关保护政策时应综合考虑人类干扰对多个物种的影响,增加决策的科学性与合理性。  相似文献

Abstract

Identification of quantitative trait loci (QTL) for yield-related traits and closely linked molecular markers is important in order to identify gene/QTL for marker-assisted selection (MAS) in wheat breeding. The objectives of the present study were to identify QTL for yield-related traits and dissect the relationships among different traits in three wheat recombinant inbred line (RIL) populations derived from crosses Doumai?×?Shi 4185 (D?×?S), Gaocheng 8901?×?Zhoumai 16 (G?×?Z) and Linmai 2?×?Zhong 892 (L?×?Z). Using the available high-density linkage maps previously constructed with the wheat 90 K iSelect single nucleotide polymorphism (SNP) array, 65, 46 and 53 QTL for 12 traits were identified in the three RIL populations, respectively. Among them, 34, 23 and 27 were likely to be new QTL. Eighteen common QTL were detected across two or three populations. Eleven QTL clusters harboring multiple QTL were detected in different populations, and the interval 15.5–32.3 cM around the Rht-B1 locus on chromosome 4BS harboring 20 QTL is an important region determining grain yield (GY). Thousand-kernel weight (TKW) is significantly affected by kernel width and plant height (PH), whereas flag leaf width can be used to select lines with large kernel number per spike. Eleven candidate genes were identified, including eight cloned genes for kernel, heading date (HD) and PH-related traits as well as predicted genes for TKW, spike length and HD. The closest SNP markers of stable QTL or QTL clusters can be used for MAS in wheat breeding using kompetitive allele-specific PCR or semi-thermal asymmetric reverse PCR assays for improvement of GY.

Early flowering 3 (ELF3) is a regulator to modulate photoperiod flowering in Arabidopsis. The homologs of ELF3 in rice and barley also have been identified essential for regulation of flowering time. In the current study, TaELF3 genes, homologs of ELF3 in bread wheat (Triticum aestivum L.), were cloned by a comparative genomics approach and located on homologous group 1 chromosomes, designated as TaELF3-1AL, TaELF3-1BL, and TaELF3-1DL, respectively. A sequence-tagged site (STS) marker was developed based on sequence polymorphism at the TaELF3-1DL locus. A quantitative trait locus (QTL) for heading date (HD) co-segregating with TaELF3-1DL explained 7.7–20.6% of the phenotypic variance in a RIL mapping population derived from the Gaocheng 8901/Zhoumai 16 cross genotyped using the wheat 90K iSelect assay. The late HD allele of TaELF3-1DL was prevalently selected in China’s specific wheat-growing regions and other countries. This study produces novel information in better understanding HD and provides a reliable functional marker for molecular marker-assisted selection in wheat breeding.  相似文献

Gastric aspiration lung injury is one of the most common clinical events. This study investigated the effects of bone‐marrow‐derived mesenchymal stem cells (BMSCs) on combined acid plus small non‐acidified particle (CASP)‐induced aspiration lung injury. Enhanced green fluorescent protein (EGFP+) or EGFP? BMSCs or 15d‐PGJ2 were injected via the tail vein into rats immediately after CASP‐induced aspiration lung injury. Pathological changes in lung tissues, blood gas analysis, the wet/dry weight ratio (W/D) of the lung, levels of total proteins and number of total cells and neutrophils in bronchoalveolar lavage fluid (BALF) were determined. The cytokine levels were measured using ELISA. Protein expression was determined by Western blot. Bone‐marrow‐derived mesenchymal stem cells treatment significantly reduced alveolar oedema, exudation and lung inflammation; increased the arterial partial pressure of oxygen; and decreased the W/D of the lung, the levels of total proteins and the number of total cells and neutrophils in BALF in the rats with CASP‐induced lung injury. Bone‐marrow‐derived mesenchymal stem cells treatment decreased the levels of tumour necrosis factor‐α and Cytokine‐induced neutrophil chemoattractant (CINC)‐1 and the expression of p‐p65 and increased the levels of interleukin‐10 and 15d‐PGJ2 and the expression of peroxisome proliferator‐activated receptor (PPAR)‐γ in the lung tissue in CASP‐induced rats. Tumour necrosis factor‐α stimulated BMSCs to secrete 15d‐PGJ2. A tracking experiment showed that EGFP+ BMSCs were able to migrate to local lung tissues. Treatment with 15d‐PGJ2 also significantly inhibited CASP‐induced lung inflammation and the production of pro‐inflammatory cytokines. Our results show that BMSCs can protect lung tissues from gastric aspiration injury and inhibit lung inflammation in rats. A beneficial effect might be achieved through BMSC‐derived 15d‐PGJ2 activation of the PPAR‐γ receptor, reducing the production of proinflammatory cytokines.  相似文献

植物通过各类受体来感知外界环境的改变从而调节自身的生长和发育情况。在拟南芥中,植物主要通过隐花色素(Cryptochromes)和向光素(Phototropins)感知蓝光。同时ZEITLUPE (ZTL),FLAVIN-BINDING KELCH REPEAT F-box1 (FKF1)和LOV KELCH PROTEIN2 (LKP2)蛋白家族也作为蓝光受体参与调控植物生长发育过程。因其特殊的蛋白结构组成,在植物的光周期开花、节律性和光形态建成等方面发挥了重要的调控作用。近来,ZTL/FKF1/LKP2蛋白家族被发现参与植物逆境胁迫响应。本文归纳了ZTL/FKF1/LKP2的生物学功能研究进展,并对其作用机制进行了总结与讨论。  相似文献
