Aromatase is the key enzyme responsible for catalyzing the conversion of C19 steroids to estrogens. Its inhibitors are widely used in breast cancer therapy. The CH2Cl2 partition of a crude ethanolic extract from the roots of Flemingia philippinensis showed potent inhibitory activity of aromatase. The constituents of the extract were analyzed and identified by liquid chromatography tandem mass spectrometry. Five purified prenylated isoflavones were evaluated for aromatase inhibition and their IC50 values ranged between 2.98 and 58.08 μm . In kinetic studies, all tested compounds behaved as reversible competitive inhibitors and their Ki values were calculated by Dixon plots. The most potent inhibitor (6,8‐diprenylorobol) had a Ki value of 1.42 μm . Furthermore, using UPLC and LC/MS, 6,8‐diprenylorobol was proven to be present in the native roots in high quantities.  相似文献

蚂蚁对热带森林中小种子的二次散布会影响种子最终到达地点和种子命运.本文以一种由鸟散布的榕树垂叶榕(Ficus benjamina)为研究对象,观察蚂蚁对不同处理种子二次散布的影响.共记录到菱结大头蚁(Pheidole rhombinoda)、法老小家蚁(Monomorium pharaonis)、横纹齿猛蚁(Odontoponera transversa)和布立毛蚁(Paratrechina bourbonica)等4种蚂蚁参与了种子的搬运,其出现频率分别为71.7%、23.3%、2.5%、2.5%.蚂蚁对种子的搬运距离为179 cm±13 cm(n=159).其中,最主要的散布者菱结大头蚁把种子搬进蚁巢后,取食了内果皮,并把58.6%的净种子抛弃在巢外垃圾堆中.蚂蚁取食内果皮显著提高了种子萌发特性(萌发率由49.3%提高到93.3%).去除内果皮后种子被蚂蚁搬走的比率显著下降(由75.0%降为29.5%).垂叶榕种子外着生的内果皮,明显提高了对蚂蚁的吸引力,而蚂蚁取食内果皮显著提高了种子萌发能力,垂叶榕和二次散布的蚂蚁之间存在着明显的互惠关系.  相似文献

How stand density and species richness affect carbon (C) storage and net primary productivity (NPP) changes with forest succession is poorly understood. We quantified the C storage of trees and the aboveground NPP in an early successional secondary birch forest (birch forest) and a late successional mixed broadleaf-Korean pine (Pinus koraiensis) forest (mixed forest) in northeastern China. We found that: 1) tree C storage in the mixed forest (120.3 Mg C ha?1) was significantly higher than that in the birch forest (78.5 Mg C ha?1), whereas the aboveground NPP was not different between the two forest types; and 2) only stand density had a positive linear relationship with tree C storage and aboveground NPP in the birch forest. In the mixed forest, both tree C storage and aboveground NPP were significantly affected by the combination of the stand density and species richness. The tree C storage to stand density and species richness relationships were hump-shaped. The aboveground NPP increased with increasing stand density, but its relationship to species richness was hump-shaped. We conclude that the effect of stand density and species richness on tree C storage and aboveground NPP was influenced by forest stand succession, and such effects should be considered in studying stand density- and species richness- ecosystem function (e.g., C storage and NPP) relationships in temperate forest ecosystems.  相似文献

Achene size and shape, surface sculpturing, and pericarp and testa wall structure of 23 Korean Saussurea spp. were investigated using scanning electron microscopy (SEM) and light microscopy to evaluate the infrageneric relationships and assess their systematic significance. Achene size categories and thickness of the testa epidermis were distinguished using biometric measurements. Four basic types of surface pattern were observed: (1) lineate; (2) striate; (3) reticulate; and (4) colliculate. Saussurea rorinsanensis was found to have some unique achene characteristics, such as a fusiform achene, uniform pappus, presence of epidermal hairs and tangentially elongated, narrow testa epidermal cells. The characteristic achene features for species were found to be achene size and shape, hilum position, surface sculpture, pappus composition, morphology of the pericarp wall and thickness of the testa epidermis. Based on 16 morphological and achene characters, a cladistic analysis resolved three well‐supported clades, with S. eriophylla as the first‐branching taxon. Saussurea pulchella and S. japonica, both belonging to Saussurea subgenus Theodorea, were distant from each other in the 50% majority rule consensus tree and the character distribution cladogram. This cladistic analysis of achene morphology and anatomy should be regarded as giving us a tentative picture of the phylogenetics of Saussurea, and this study may serve as a reference for future hypotheses and studies on the characterization and classification of Saussurea spp. in Korea.  相似文献

【目的】为探究UASB颗粒污泥启动的单室微生物电解池(Single-chamber microbial electrolysis cell,SMEC)对Ni(II)的去除途径和SMEC中微生物群落的动态特征。【方法】以乙酸钠为底物,采用单因子控制方法分析SMEC对Ni(II)的去除途径和应用Illumina高通量测序技术解析SMEC启动过程中微生物群落的组成和结构动态学特征。【结果】结果表明,SMEC对重金属的去除主要通过吸附和微生物作用。经培养驯化功能菌群发生变化。成熟单室微生物燃料电池(Single-chamber microbial fuel cell,SMFC)阳极生物膜菌群主要是Proteobacteria(变形菌门,91.42%)中的Geobacter sp.(地杆菌属,76.25%);阴极生物膜菌群主要是Bacteroidetes(拟杆菌门,47.99%)中的Niabella sp.(布鲁氏菌属,33.01%)和Proteobacteria(45.74%)中的Ochrobactrum sp.(苍白杆菌属,10.80%)。成熟SMFC改装成的SMEC在12.5 mg-Ni(II)/L下,阳极生物膜菌群由单一优势菌Geobacter sp.转变为Geobacter sp.(41.56%)和Proteobacteria中的Azospirillum sp.(固氮螺菌属,5.97%);阴极生物膜菌群由Niabella sp.和Ochrobactrum sp.转变为Firmicutes(厚壁菌门,25.21%)中的Acetoanaerobium sp.(19.28%)、Proteobacteria(51.42%)中的Dokdonella sp.(16.48%)和Azospirillum sp.(9.49%)。【结论】本研究表明,污泥微生物经SMFC和SMEC驯化过程及Ni(II)的淘汰和选择,在电极上形成了稳定、高效产电与除镍菌群,优势菌群为Proteobacteria。  相似文献

Fragile X syndrome is caused by the functional loss of the fragile X mental retardation 1 (FMR1) gene. Deletion of the FMR1 ortholog in Drosophila melanogaster (Fmr1) recapitulates many phenotypes associated with fragile X syndrome. We have discovered that Fmr1 mutant Drosophila die during development when reared on food containing increased levels of glutamate, which is consistent with the theory that FMR1 loss results in excess glutamate signaling. Using this lethal phenotype, we screened a chemical library of 2,000 compounds and identified nine molecules that rescued the lethality, including three that implicate the GABAergic inhibitory pathway. Indeed, GABA treatment rescued several known Fmr1 mutant phenotypes in flies, including mushroom bodies defects, excess Futsch translation and abnormal male courtship behavior. These data are consistent with GABAergic inhibition of the enhanced excitatory pathway in fragile X syndrome. In addition, our screen reveals that the muscarinic cholinergic receptors may have a role in fragile X syndrome in parallel to the GABAergic pathway. These results point to potential therapeutic approaches for treating fragile X syndrome.  相似文献

Controlling the uptake, transport, translocation, and accumulation of excessive amounts of cadmium from polluted environments is critical for plants and, consequently, humans with regard to food safety. Plants adopt various cellular and molecular mechanisms to minimize Cd toxicity. Upon exposure to Cd, plants initially implement avoidance strategies, such as production of organic acids, chelation, and sequestration, to prevent metal access to root cells. Nevertheless, Cd can be transported through the roots, stems, and leaves via apoplastic and symplastic pathways. These processes have been controlled by specific sites at the root surface and root cortex, in cells responsible for loading the root xylem, at the transition between the vascular systems of the root and the shoot, and in connecting tissues and cells at the stem. Although resistance to heavy metal cadmium can be achieved by either avoidance or tolerance, genetic basis to tolerance is therefore implied, in that these mechanisms are heritable attributes of tolerant mutants or genotypes.  相似文献

NaVPO4F has received a great deal of attention as cathode material for Na‐ion batteries due to its high theoretical capacity (143 mA h g?1), high voltage platform, and structural stability. Novel NaVPO4F/C nanofibers are successfully prepared via a feasible electrospinning method and subsequent heat treatment as self‐standing cathode for Na‐ion batteries. Based on the morphological and microstructural characterization, it can be seen that the NaVPO4F/C nanofibers are smooth and continuous with NaVPO4F nanoparticles (≈6 nm) embedded in porous carbon matrix. For Na‐storage, this electrode exhibits extraordinary electrochemical performance: a high capacity (126.3 mA h g?1 at 1 C), a superior rate capability (61.2 mA h g?1 at 50 C), and ultralong cyclability (96.5% capacity retention after 1000 cycles at 2 C). 1D NaVPO4F/C nanofibers that interlink into 3D conductive network improve the conductivity of NaVPO4F, and effectively restrain the aggregation of NaVPO4F particles during charge/discharge process, leading to the high performance.  相似文献

Gammadelta T cells suppress airway hyperresponsiveness (AHR) induced in allergen-challenged mice but it is not clear whether the suppression is allergen specific. The AHR-suppressive cells express TCR-Vgamma4. To test whether the suppressive function must be induced, we adoptively transferred purified Vgamma4(+) cells into gammadelta T cell-deficient and OVA-sensitized and -challenged recipients (B6.TCR-Vgamma4(-/-)/6(-/-)) and measured the effect on AHR. Vgamma4(+) gammadelta T cells isolated from naive donors were not AHR-suppressive, but Vgamma4(+) cells from OVA-stimulated donors suppressed AHR. Suppressive Vgamma4(+) cells could be isolated from lung and spleen. Their induction in the spleen required sensitization and challenge. In the lung, their function was induced by airway challenge alone. Induction of the suppressors was associated with their activation but it did not alter their ability to accumulate in the lung. Vgamma4(+) gammadelta T cells preferentially express Vdelta4 and -5 but their AHR-suppressive function was not dependent on these Vdeltas. Donor sensitization and challenge not only with OVA but also with two unrelated allergens (ragweed and BSA) induced Vgamma4(+) cells capable of suppressing AHR in the OVA-hyperresponsive recipients, but the process of sensitization and challenge alone (adjuvant and saline only) was not sufficient to induce suppressor function, and LPS as a component of the allergen was not essential. We conclude that AHR-suppressive Vgamma4(+) gammadelta T cells require induction. They are induced by allergen stimulation, but AHR suppression by these cells does not require their restimulation with the same allergen.  相似文献
