In response to reported outbreaks of Legionnaires’ disease near a biological waste treatment plant, Legionella-containing aerosols were characterized at this location. Culturable bacteria-containing particles were assessed with slit-to-agar samplers and liquid impingement. Results showed that the air at this location contained a higher level of airborne Legionella particles compared to that of a control clean location 90 km away. On average, about 1 of 50 culturable particles contained Legionella (2%). The median particle size was estimated to 3.5 μm permitting an estimation of a maximum about 147 Legionella cells per aggregate. This implies that in <11 h, a human under such conditions (worse-case), may inhale an estimated lethal dose of individual Legionella cells (1,000 cells, 100% alveolar retained deposition level). Additionally, 44 taxonomically different airborne bacterial genera were measured at the treatment plant using denaturing gradient-gel electrophoresis (DGGE) and 16S rDNA analysis of air samples. Of these, 64% of the genera were not detected at the clean location. Our findings suggest that the treatment facility may emit respirable particles, as complex aggregates, containing a mixture of several Legionella cells and other bacterial cells. These findings can potentially have an impact on assessing environmental risk exposure and health hazard prediction modeling.  相似文献

Sequencing-based approaches have led to new insights about DNA methylation. While many different techniques for genome-scale mapping of DNA methylation have been employed, throughput has been a key limitation for most. To further facilitate the mapping of DNA methylation, we describe a protocol for gel-free multiplexed reduced representation bisulfite sequencing (mRRBS) that reduces the workload dramatically and enables processing of 96 or more samples per week. mRRBS achieves similar CpG coverage to the original RRBS protocol, while the higher throughput and lower cost make it better suited for large-scale DNA methylation mapping studies, including cohorts of cancer samples.  相似文献

Bench-scale experiments were performed to investigate the persistence of activated persulfate using citric acid (CA) chelated ferrous (Fe(II)), peroxide (H2O2), or hydroxide (OH?) activation in the presence of well-characterized aquifer solids. Chelation by citric acid was ineffective in controlling the interaction between persulfate and Fe(II), and oxidation of Fe(II) was observed, causing a rapid initial decrease in persulfate concentration. Subsequent to this loss, first-order persulfate degradation rate coefficients (kobs) were estimated, which were up to four times higher than the unactivated case due to the interaction with Fe(III), Fe(II), or CA. Total oxidation strength (TOS) was measured for peroxide activation experiments and was observed to decrease rapidly early due to peroxide degradation. This was followed by slow degradation kinetics similar to that of unactivated persulfate, implying that the initial TOS degradation was peroxide-dominated and the long-term kinetics were dominated by persulfate degradation. The kobs later used to capture TOS degradation were ~1 to 100 times higher than kobs for unactivated persulfate. For alkaline activation, kobs were only one to four times higher than unactivated persulfate, and therefore alkaline conditions demonstrated the least overall impact on persulfate stability among the various activation strategies explored.  相似文献

Live virtual machine migration can have a major impact on how a cloud system performs, as it consumes significant amounts of network resources such as bandwidth. Migration contributes to an increase in consumption of network resources which leads to longer migration times and ultimately has a detrimental effect on the performance of a cloud computing system. Most industrial approaches use ad-hoc manual policies to migrate virtual machines. In this paper, we propose an autonomous network aware live migration strategy that observes the current demand level of a network and performs appropriate actions based on what it is experiencing. The Artificial Intelligence technique known as Reinforcement Learning acts as a decision support system, enabling an agent to learn optimal scheduling times for live migration while analysing current network traffic demand. We demonstrate that an autonomous agent can learn to utilise available resources when peak loads saturate the cloud network.  相似文献

In the classical human leukocyte antigen (HLA) class I antigen processing and presentation pathway, the antigenic peptides are generated from viral proteins by multiple proteolytic cleavages of the proteasome (and in some cases other cytosolic proteases) and transported to the endoplasmic reticulum (ER) lumen where they are exposed to aminopeptidase activity. In human cells, two different ER-resident enzymes, ERAP1 and ERAP2, can trim the N-terminally extended residues of peptide precursors. In this study, the possible cooperative effect of generating five naturally processed HLA-B27 ligands by both proteases was analyzed. We identified differences in the products obtained with increased detection of natural HLA-B27 ligands by comparing double versus single enzyme digestions by mass spectrometry analysis. These in vitro data suggest that each enzyme can use the degradation products of the other as a substrate for new N-terminal trimming, indicating concerted aminoproteolytic activity of ERAP 1 and ERAP2.  相似文献

How do the feedbacks between tectonics, sediment transport and climate work to shape the topographic evolution of the Earth? This question has been widely addressed via numerical models constrained with thermochronological and geomorphological data at scales ranging from local to orogenic. Here we present a novel numerical model that aims at reproducing the interaction between these processes at the continental scale. For this purpose, we combine in a single computer program: 1) a thin-sheet viscous model of continental deformation; 2) a stream-power surface-transport approach; 3) flexural isostasy allowing for the formation of large sedimentary foreland basins; and 4) an orographic precipitation model that reproduces basic climatic effects such as continentality and rain shadow. We quantify the feedbacks between these processes in a synthetic scenario inspired by the India-Asia collision and the growth of the Tibetan Plateau. We identify a feedback between erosion and crustal thickening leading locally to a <50% increase in deformation rates in places where orographic precipitation is concentrated. This climatically-enhanced deformation takes place preferentially at the upwind flank of the growing plateau, specially at the corners of the indenter (syntaxes). We hypothesize that this may provide clues for better understanding the mechanisms underlying the intriguing tectonic aneurisms documented in the Himalayas. At the continental scale, however, the overall distribution of topographic basins and ranges seems insensitive to climatic factors, despite these do have important, sometimes counterintuitive effects on the amount of sediments trapped within the continent. The dry climatic conditions that naturally develop in the interior of the continent, for example, trigger large intra-continental sediment trapping at basins similar to the Tarim Basin because they determine its endorheic/exorheic drainage. These complex climatic-drainage-tectonic interactions make the development of steady-state topography at the continental scale unlikely.  相似文献

Epigenetic regulations play important roles in plant development and adaptation to environmental stress. Recent studies from mammalian systems have demonstrated the involvement of ten-eleven translocation (Tet) family of dioxygenases in the generation of a series of oxidized derivatives of 5-methylcytosine (5-mC) in mammalian DNA. In addition, these oxidized 5-mC nucleobases have important roles in epigenetic remodeling and aberrant levels of 5-hydroxymethyl-2′-deoxycytidine (5-HmdC) were found to be associated with different types of human cancers. However, there is a lack of evidence supporting the presence of these modified bases in plant DNA. Here we reported the use of a reversed-phase HPLC coupled with tandem mass spectrometry method and stable isotope-labeled standards for assessing the levels of the oxidized 5-mC nucleosides along with two other oxidatively induced DNA modifications in genomic DNA of Arabidopsis. These included 5-HmdC, 5-formyl-2′-deoxycytidine (5-FodC), 5-carboxyl-2′-deoxycytidine (5-CadC), 5-hydroxymethyl-2′-deoxyuridine (5-HmdU), and the (5′S) diastereomer of 8,5′-cyclo-2′-deoxyguanosine (S-cdG). We found that, in Arabidopsis DNA, the levels of 5-HmdC, 5-FodC, and 5-CadC are approximately 0.8 modifications per 106 nucleosides, with the frequency of 5-HmdC (per 5-mdC) being comparable to that of 5-HmdU (per thymidine). The relatively low levels of the 5-mdC oxidation products suggest that they arise likely from reactive oxygen species present in cells, which is in line with the lack of homologous Tet-family dioxygenase enzymes in Arabidopsis.  相似文献

To date, most studies of white matter changes in Bipolar Disorder (BD) have been conducted in older subjects and with well-established disorders. Studies of young people who are closer to their illness onset may help to identify core neurobiological characteristics and separate these from consequences of repeated illness episodes or prolonged treatment. Diffusion tensor imaging (DTI) was used to examine white matter microstructural changes in 58 young patients with BD (mean age 23 years; range 16–30 years) and 40 controls. Whole brain voxelwise measures of fractional anisotropy (FA), parallel diffusivity (λ//) and radial diffusivity (λ⊥) were calculated for all subjects. White matter microstructure differences (decreased FA corrected p<.05) were found between the patients with BD and controls in the genu, body and splenium of the corpus callosum as well as the superior and anterior corona radiata. In addition, significantly increased radial diffusivity (p<.01) was found in the BD group. Neuroimaging studies of young patients with BD may help to clarify neurodevelopmental aspects of the illness and for identifying biomarkers of disease onset and progression. Our findings provide evidence of microstructural white matter changes early in the course of illness within the corpus callosum and the nature of these changes suggest they are associated with abnormalities in the myelination of axons.  相似文献
